耐盐碱解磷菌的筛选鉴定及对绿豆的促生作用

摘    要:从大庆耐盐碱植物的根际土壤中分离得到100株解磷菌,对筛选得到的菌株解磷量进行测定,并结合其他促生特性检验,选出3株耐盐碱高效解磷菌。通过生理生化试验和16S rRNA鉴定出JC8、JC11和JP8这3株菌,分别为Pseudomonas sp.,Acinetobacter sp.和Microbacterium sp.;对3株菌株的耐盐碱能力进行测试,结果显示3株菌株均具有较高的盐碱耐受能力。在轻度盐碱缺磷土壤中分别施用JC8、JC11和JP8后种植绿豆,3株解磷菌均能显著提高绿豆幼苗的根长、根鲜质量以及植株鲜质量;同时降低绿豆叶片抗氧化酶活性,提高渗透性物质含量,增强土壤酶活性,说明解磷菌能够解磷并从多方面增强绿豆幼苗的盐碱抗性,促进其生长。

Abstract:In this paper,100 phosphate solubilizing bacteria strains were isolated from the rhizosphere soil of saline-alkali tolerant plants in Daqing,then 3 saline-alkali tolerant bacteria strains (JC8,JC11 and JP8) which have high-efficient phosphate solubilizing ability were screened out through phosphate solubilization capacity and growth-promoting characteristics tests. Through physiological and biochemical tests and 16S rRNA alignment,the above strains were identified as Pseudomonas sp.(JC8),Acinetobacter sp.(JC11) and Microbacterium sp.(JP8). The salt-tolerant ability test showed that the all the three strains had high salt-alkali tolerance. After application of JC8,JC11 and JP8 in mild saline-alkali and phosphorus-deficient soils,the mung beans were planted,each strain could significantly increase the root length,root fresh quality and plant fresh quality of mung bean seedlings,and reduce the antioxidant enzymes of mung bean leaves. The further investigation revealed that the osmotic substances in leaves and soil enzyme activity were enhanced. In conclusion,the phosphorus solubilizing bacteria strains screened and identified in this study can effectively solubilize phosphorus,enhance the salt-alkali resistance of mung bean seedlings and promote their growth.
