在试管中筛选烟草抗黑胫病突变体

用烟草体细胞愈伤组织与烟草黑胫病菌共同培养,在209块G_(140)烟草体细胞愈伤组织中有1块长出抗菌突变系,离开选择压力后经3次140天继代培养仍能表达抗性。用烟草黑胫病菌接种在单倍体试管苗上,从546株八大香单倍体试管苗中筛选到1株(代号RB_1)、从2431株小黄金1025单倍体试管苗中筛选到2株(代号RS_(1.3))抗病突变系。初步鉴定RB_1、RS_(1.3)、的抗病性均超过其亲本和高抗品种G_(140);取RB_1、RS_(1.3)叶片培养,其体细胞无性系植株群体仍具抗病性。

Selection of Mutant Resistance to Phytophthora Parasitica var.Nicctianae Tucker from Tobacco in Vitro

A mutant RG140-1 was selected from 209 uninduced mutation calluses of tobacco varietyG140 inoculated with Phytophthora parasitica var. nicotianae Tucker. It expressed the resistance afterRG140-1 left selection pressure and had been cultured for 3 times 140 days. Another as RB_1 from546 haploid plants of the variety Ba Da Xing and yet 2 as RB_(1.3) from 2431 haploid plants of thevariety Xiao Huang Jing 1025 in vitro that were inoculated with the same disease-producing germwere also selected. The 3 mutants showed that resistance to the disease-producing germ exceededtheir parents and high resistant variety G140. The regenerated plants from their leaved culturedin vitro also expressed the resistance.
