君子兰八氢番茄红素合成酶基因的克隆与序列分析

摘    要:根据GenBank上登陆的黄水仙八氢番茄红素合成酶基因(登录号:X78814.1)设计特异引物,通过PCR扩增从君子兰cDNA中克隆出一个1.4 kb的片段。序列分析表明,这个序列全长1 395 bp,包含一个1 272 bp的开放阅读框,并且编码423个氨基酸。与水仙(DQ984674)、文心兰(FJ859988)和茶花(EF545005)相比,此cDNA序列显示出98%~76%的同源性,并且与水仙、猕猴桃、番茄和柿相比,其氨基酸序列显示出达到了99%~80%的同源性。由此可知,研究所克隆的序列为君子兰的PSY全长基因,该PSY基因已登陆GenBank(登录号:JF499694)。

Abstract:A pair of specific primers was designed according to the PSY mRNA sequene of Narcissus pseudo narcissus(GenBank accession number X78814.1) and a novel 1.4 kb cDNA was obtained from Clivia miniata Rogel by PCR method.The sequence analysis showed that the length of cDNA was 1 395 bp with an open reading frame of 1 272 bp.This gene encoded a polypeptide of 423 animo acids.Its cDNA sequence showed 98%-76% identity with PSY genes from Narcissus tazetta(DQ984674),Oncidium gower Ramsey(FJ859988) and Camellia sinensis(EF545005).Its amino acid sequence shared 99%-80% identity with PSY protein from Narcissus tazetta,Actinidia deliciosa,Solanum lycopersicum and Diospyros kaki,respectively.The results indicated that the sequence obtained in this study was the full length PSY cDNA from Clivia miniata Rogel and the GenBank accession number was JF499694.
