山药蛋白酶解液的分离纯化

摘    要:以山药(Dioscorea opposite Thumb.)为原料,选取碱性蛋白酶酶解山药蛋白粗提物制备山药蛋白酶解液。分别采用纤维素DE-52阴离子交换层析和Sephadex G-50凝胶层析,对酶解制备的山药蛋白酶解液进行分离纯化。结果表明:经过纤维素DE-52阴离子交换层析,得到4个吸收峰,收集每个峰组分,其单位质量的还原能力分别为峰2(P2)>峰1(P1)>峰3(P3)>峰4(P4)。采用Sephadex G-50凝胶分别用蒸馏水和Tris-HCl缓冲液对P1和P2组分继续分离纯化,均能得到2个吸收峰(组分)。

Abstract:The Chinese yam protein was hydrolyzed by Alkaline protease to prepare hydrolytic products. They were isolated and purified by cellulose DE-52 anion-exchange chromatography and Sephadex G-50 gel chromatography. Four peaks of absorption were obtained with cellulose DE-52 anion-exchange chromatography, which showed the levels of reducing power as Peak 2 (P2)〉Peak 1 (P1)〉Peak 3 (P3)〉Peak 4 (P4). Distilled water and Tris-HC1 buffer were employed for further isolation of the peaks P1 and P2 with Sephadex G-50 gel chromatography, 2 peaks of absorption were obtained, respectively.
