DPI对尾叶桉愈伤组织诱导和抗氧化酶活性的影响

摘    要:将不同浓度DPI添加到尾叶桉(Eucalyptus urophylla)愈伤组织诱导培养基中,分析DPI对其愈伤组织诱导和SOD、POD、CAT、APX 4种抗氧化酶活性的影响。结果表明,高于1.5μmol/L的DPI显著抑制尾叶桉愈伤组织的诱导,0~0.25μmol/L的DPI能减轻褐化,0.1μmol/L DPI抗褐化效果最好,褐化率仅9.82%。0~0.25μmol/L的DPI处理后,尾叶桉APX活性随DPI浓度升高而升高,SOD、POD、CAT活性随DPI浓度升高呈先升高后降低的趋势。添加0.10μmol/L DPI可提高保护酶活性,减轻褐化,促进尾叶桉愈伤组织诱导。

Abstract:The callus of Eucalyptus urophylla was induced by adding different concentrations of diphenyleneiodonium (DPI) to the callus induction medium. And SOD,POD,CAT and APX activities of the callus were measured. The concentration of DPI higher than 1.5μmol/L significantly inhibited callus induction. 0~0.25μmol/L DPI can reduce browning,of which 0.1μmol/L concentration can get the best anti-browning effect,browning rate only 9.82%. In the DPI treatment of 0~0.25μmol/L,the ac-tivity of APX increased,while the activities of SOD,POD and CAT increased first and then decreased with the increase of DPI concentration. The results showed that addition of 0.10μmol/L DPI could promote the callus of Eucalyptus urophylla in-duction by increasing the protective enzyme activity and reducing the browning.
