陆两优996种子纯度的SRAP指纹图谱鉴定

摘    要:利用270对SRAP引物对陆两优996和2个亲本的DNA指纹图谱,获得能在父、母本间表现出多态性的特异SRAP标记引物有54对,其中差异性条带136个。利用Me3Em8引物,条带在杂交种完全互补,对200株样本进行了纯度鉴定,结果鉴定纯度为97.50%,与田间种植鉴结果97.50%(海南鉴定)一致,表明SRAP标记技术适用于种子纯度鉴定。

Studies on SRAP Fingerprinting in Purity Identification of Hybrid Seeds of Lu Liangyou 996

Abstract:In this research, DNA fingerprints of 1 male sterile lines and 1 restorer lines of two-line hybrid rice were constructed by using 270 SRAP (Sequence related amplified polymorphism) primers. The 54 SARP primers could amplify 136 polymorphic bands totally with a mean of 2. 5 bands per primer. The 200 single plants of Luliangyou 996 were tested in laboratory by a specific SRAP marker(Me3Em8)whose bands were complement in hybrid combination seeds. The seed purity was 97.50%, which was same to the field purity (97.50% in Hainan). The results of this research suggested that SRAP technique could be used in seed purity identification of two-line hybrid rice.
