| 摘 要： | 根据猪流感病毒M基因序列设计了一对扩增M基因684 bp片段的特异性引物,建立了RT-PCR快速诊断猪流感的方法,采用该方法检测出H1、H3、H5和H9 4个亚型猪流感病毒标准参考株为阳性,猪副黏病毒、圆环病毒2型和猪繁殖与呼吸综合征病毒检测结果呈阴性.RT-PCR最少可检测到1 000EID50的病毒量核酸,可直接从猪流感病毒感染小鼠的组织样品中检测到病毒.

| Abstract: | A RT-PCR diagnostic technique for the detection of swine influenza viruses was developed,which was used to amplify 684 bp segment of the Mtrix (M) gene of H1,H3,H5 and H9 subtype swine influen- za viruses.Sensitivity to detection of highly purified influenza virus by RT-PCR reached approximately 1 000 EID_(50).All the influenza viruses tested by RT-PCR were positive,whereas the Paramyxovirus,Por- cine reproductive and respiratory syndrome virus,Porcine circovirus type 2,Classical swine fever virus used as controls were negative.The efficiency of the system for detection of influenza viruses directly in clinical samples of infected mice was confirmed by virus embryonated eggs inoculation. |
