摘    要:通过比较SDS和CTAB 2种方法 ,CTAB法更适于油茶DNA的提取 ,并在CTAB法提取过程中将一些有效的去多糖、多酚的步骤进行组合 ,建立适合油茶DNA的提取方法 ,同时构建了油茶DNA的PCR扩增程序 ,并从预变性温度、Mg2 +浓度、dNTPs、TaqDNA聚合酶、模板DNA等方面确定了PCR扩增条件

Abstract:Two methods(SDS method and CTAB method) for DNA extrcating were compared.The CTAB method was the better for Camellia oleifera DNA extracting.Some process such as eliminating amylose and eliminating polyphenols were united and were ameliorated,then the suited method for Camellia oleifera DNA extracting was defined.PCR amplified procedure was established and PCR amplified conditions were defined on the factors of pre-denaturating temperature,Mg 2+ density,dNTPs density,Taq DNA polymerase and DNA density.
