以美洲黑杨‘D189’和辽宁杨为杂交亲本,利用花粉管通道法,经柱头滴注,将携带山葡萄CBF3(VaCBF3)基因的质粒导入杨树中。结果显示:在杂交授粉后,经过目的基因导入,共获得了52株转化后代,经PCR检测,有5株扩增出目的条带,初步推断目的基因已导入到杨树中。本研究结果可为进一步利用花粉管通道培育抗寒杨树新品系提供参考。 相似文献

根据已克隆的戊型肝炎病毒株swCH189株结构蛋白基因(ORF2)的抗原性及水溶性分析,在其序列上设计套式引物,采用RT-PCR技术扩增猪戊型肝炎病毒(HEV)结构蛋白基因ORF2主要抗原基因片段,长度为728bp,将其克隆于PMD18-T载体,测序结果表明插入的片段属于猪戊型肝炎病毒结构蛋白基因ORF2部分;将该片段插入pet32a表达载体,经酶切、测序鉴定证实获得了带有目的基因的重组表达质粒。将重组质粒转化Rosetta菌,经0.3mmol/L IPTG诱导得到融合表达,得到相对分子质量为45 000的重组蛋白,以包涵体形式存在。Western blot分析表明,该蛋白可以与猪戊型肝炎阳性血清反应,表明该蛋白具有良好的反应原性。并用SDS电泳切胶纯化和透析浓缩方法进行纯化,用纯化的蛋白免疫兔子,经每2周免疫1次,连续3次免疫后,对免疫组和对照组,分别在45,60,90d采血,每只兔子采集1份。用夹心ELISA法测定免疫血清抗体,结果表明免疫组在45d后均产生具有HEV抗原结合活性的抗体。本试验对猪swCH189株CP239基因的克隆和表达研究,为进一步研究结构蛋白基因ORF2的生物学功能奠定了基础。 相似文献

A two year field experiment was conducted in field conditions using wheat (Triticum aestivum ssp. vulgare L. cv. Bezostaja) as the test plant for the evaluation of phosphate solubilizing (+PS) microorganisms. Bacterial strains significantly (P < 0.05) increased plant biomass (by 13 to 36%) without plant P enrichment. Only Bacillus sp. #189 significantly (P < 0.05) raised plant P-content, biomass and rhizosphere soil Olsen P at Zadoks Scale 62 simultaneously. Inoculations variably increased plant potassium (K-), magnesium (Mg-), zinc (Zn-), and managenese (Mn-) contents at harvest (Zadoks 92) but not earlier (Zadoks 31 and 61). The enrichment of the inoculated plants with nutrients other than P indicates the presence of alternative plant growth promoting mechanisms. This study showed that promising phosphate solubilizing microorganisms could increase not only the P content of the plants, but also the soil available P in rhizosphere transiently. 相似文献

In the present study, the effects of the anti-estrogen ZM 189,156 and the aromatase inhibitor fadrozole were evaluated in
a 40-day juvenile assay developed for screening of endocrine disrupting chemicals. Juvenile zebrafish were exposed from 20
to 60 days post hatch (dph) to ZM 189, 154 (100 μg l−1 and 200 μg l−1) and fadrozole (10, 32, and 100 μg l−1). VTG concentrations were measured at 38 dph by the use of a direct non-competitive sandwich ELISA and sex ratios were determined
at 60 dph by histological examination of the gonads. A small but significant increase in VTG concentrations was observed in
fish exposed to ZM 189, 156 (100 and 200 μg l−1) and fadrozole (10 and 100 μg l−1) compared to control groups. In fish exposed to ZM 189, 156 and fadrozole, the percentage of females declined and the number
of undifferentiated fish increased. These findings show that exposure of juvenile zebrafish to an aromatase inhibitor or an
anti-estrogen during early development inhibits differentiation and development of female gonads. The data presented, furthermore,
show that the 40-day juvenile assay may be suitable for screening endocrine disrupting chemicals acting as anti-estrogens
