We developed 178 recombinant inbred lines from a southern‐by‐spring oat population designated as “TxH.” These lines were genotyped to generate a high‐quality linkage map that resolved 6,902 markers into 21 linkage groups that matched closely with the latest hexaploid oat consensus map. Three major quantitative trait loci (QTLs) affecting heading date were found in locations that are consistent with known QTLs and candidate genes, and two other QTLs affecting heading date were found in novel locations. Five QTLs affecting plant height were found. Both sets of QTLs are responsible for transgressive segregation observed for these two traits. Four QTLs affecting resistance to crown rust, caused by the pathogen Puccinia coronata f. sp. avenae, were identified. Two of these QTLs are consistent with known clusters of rust resistance genes, while two may represent new locations of novel rust resistance genes. A complete set of SNP sequences suitable for generating markers for molecular selection is provided.  相似文献

A comparison was made between the genes in 29 new selections of wild emmer wheat resistant to yellow rust over wide geographic areas and the previously extensively studied selectionTriticum dicoccoides G-25. In 23 selections the resistance may be conferred by 1 dominant gene; these include 11 selections in which the gene is different from the dominant gene in sel. G-25 and two others in which the genes were closely linked or allelic to the gene in G-25, differing from sel. G-25 by race-specificity. Two dominant genes different from the gene in sel. G-25, seem to be present in one selection. In five selections the resistance may be conferred by one or two recessive genes, including three instances in which the recessive gene was associated with a dominat gene. Our findings show that at least 19 out of the 29 selections studied possess genes which are different from the gene inT. dicoccoides sel. G-25.Samenvatting In dit onderzoek werden 29 nieuwe resistente wilde-emmer selecties (Triticum dicoccoides) gekruist met de reeds uitvoerig bestudeerde resistente selectie G-25, om na te gaan of de resistentie van de nieuwe selecties wordt veroorzaakt door genen op dezelfde locus als het dominante gen in sel. G-25 of dat er andere loci bij zijn betrokken. De ouders, de F1-en F2-populaties van een bepaalade selectie werden in het kiemplantstadium getoetst met één Israëlisch gele-roest isolaat van fysio 2E0 of van fysio 2E18. In de uitsplitsende F2-populaties werden de niet-sporulerende planten als resistent beschouwd en de sporulerende als vatbaar.In de F2-populaties van 12 herkomsten werden geen vatbare planten gevonden, hetgeen er op duidt dat de resistentie wordt veroorzaakt door een gen op dezelfde locus als het gen in G-25 of door een gen dat neuw gekoppeld is aan het gen in G-25. Voor twee van deze herkomsten kan op basis van een fysio-specifieke interactie worden vastgesteld dat de resistentie berust op allelen die verschillen van het allel in sel. G-25. In 11 herkomsten werd een uitsplitsing voor twee dominante gene gevonden (RS=151), waarbij het tweede dominante gen uit de getoetste nieuwe selectie afkomstig is. De aanwezigheid van twee dominante genen verschillend van het gen in sel. G-25 werd gevonden in één herkomst (631). In de overige vijf selecties bleek de resistentie te worden veroorzaakt door één of twee recessieve genen waarnaast in drie gevallen ook nog een dominant gen werd gevonden.De resultaten tonen aan dat tenminste 19 van de 29 bestudeerde selecties resistentiegenen bezitten die verschillen van het gen inT. dicoccoides sel.  小麦(Triticum aestivum)品种洛夫林10和叶锈菌小种366组成不亲和组合,小麦叶片发生过敏性坏死反应(HR)是小麦抵抗叶锈菌侵染的重要因素。在接种前给小麦叶片分别预注射微管解聚药物磺草硝(oryzalin)和微丝解聚药物细胞松弛素D (cytochalasin D,CD),结果表明2种药物注射使得寄主因叶锈菌侵染诱导的细胞过敏性坏死数目明显减少,并且注射药物的浓度越大,寄主细胞发生HR的数量越少。说明肌动蛋白和微管蛋白的聚合状态是诱发小麦叶片发生HR防卫反应所必需的,细胞骨架在小麦抵抗叶锈菌侵染过程中可能起着重要作用。  相似文献

Four foliar and two stem-base pathogens were inoculated onto wheat plants grown in different substrates in pot experiments. Soils from four different UK locations were each treated in three ways: (i) straw incorporated in the field at 10 t ha−1 several months previously; (ii) silicon fertilization at 100 mg L−1 during the experiment; and (iii) no amendments. A sand and vermiculite mix was used with and without silicon amendment. The silicon treatment increased plant silica concentrations in all experiments, but incorporating straw was not associated with raised plant silica concentrations. Blumeria graminis and Puccinia recondita were inoculated by shaking infected plants over the test plants, followed by suitable humid periods. The silicon treatment reduced powdery mildew ( B. graminis ) substantially in sand and vermiculite and in two of the soils, but there were no effects on the slight infection by brown rust ( P. recondita ). Phaeosphaeria nodorum and Mycosphaerella graminicola were inoculated as conidial suspensions. Leaf spot caused by P. nodorum was reduced in silicon-amended sand and vermiculite; soil was not tested. Symptoms of septoria leaf blotch caused by M. graminicola were reduced by silicon amendment in a severely infected sand and vermiculite experiment but not in soil or a slightly infected sand and vermiculite experiment. Oculimacula yallundae (eyespot) and Fusarium culmorum (brown foot rot) were inoculated as agar plugs on the stem base. Severity of O. yallundae was reduced by silicon amendment of two of the soils but not sand and vermiculite; brown foot rot symptoms caused by F. culmorum were unaffected by silicon amendment. The straw treatment reduced severity of powdery mildew but did not detectably affect the other pathogens. Both straw and silicon treatments appeared to increase plant resistance to all diseases only under high disease pressure.  相似文献

As a first step in the development of a mycoherbicide for biological control of Cirsium arvense indigenous fungal pathogens that had been isolated from diseased hosts were tested both singly and in combination on potted plants under environmental conditions. Disease symptoms (necroses, chloroses and macerations) and parameters of growth and development were evaluated to determine the potential for weed control. During 1998, Phoma destructiva, Phoma hedericola, a Mycelia sterilia and Puccinia punctiformis were inoculated singly. With the exception of the obligate biotroph P. punctiformis (local infections), the single inoculations reduced the measured parameters. A combined inoculum of P. punctiformis and Ph. hedericola was less effective than Ph. hedericola alone. In 1999, Ph. hedericola, Ph. destructiva, Ph. nebulosa and the Mycelia sterila were applied both singly and as a combined inoculum. Of all the tested inocula, the mixture of four pathogens had the greatest potential for biocontrol, significantly reducing reproductive capacity of the plants. These results demonstrate the importance of taking advantage of synergisms in developing a mycoherbicide.  相似文献

为持续控制小麦叶锈病及促进小麦的抗叶锈病育种工作,2019—2020年自江苏、浙江和安徽3个省采集自然感叶锈病的小麦病叶,经分离获得小麦叶锈菌单孢分离物,利用43个小麦叶锈病鉴别寄主材料对其致病类型进行鉴定,并对其毒性结构进行分析。结果显示,从170份小麦叶锈菌单孢分离物中共鉴定出67个致病类型,主要致病类型为THS、SHJ、PHS和SHS,出现频率分别为8.8%、7.6%、5.9%和5.9%。江苏、浙江和安徽3个省的单孢分离物对携带抗叶锈基因Lr10、Lr12、Lr22a、Lr22b、Lr29、Lr33、Lr35和Lr36的鉴别寄主材料的苗期毒性频率均超过90.0%,而对携带抗叶锈基因Lr9、Lr24、Lr25、Lr28、Lr38、Lr40、Lr41、Lr42、Lr43和Lr13+3ka的鉴别寄主材料的苗期毒性频率均小于10.0%。卡方检验及Fisher精确检验显示,3个省小麦叶锈菌群体对抗叶锈基因Lr1、Lr2a、Lr3、Lr14b、Lr18、Lr21、Lr26、Lr27+31、Lr32和Lr37的毒力存在显著分化。浙江省小麦叶锈菌群体具有较少的毒性因子(4.73)和毒性值(600...  相似文献

Phyllosphere yeasts may play an important role in suppressing necrotrophic pathogens. In a randomized block design, yeast densities on flag leaves and second leaves of field-grown wheat were left unchanged (D), or were lowered by sprayings with carbendazim (A), or were raised by sprayings with a mixture ofSporobolomyces roseus, Cryptococcus laurentii var.flavescens and nutrients (B), or with carbendazim, carbendazim-resistantS. roseus andC. laurentii var.flavescens and nutrients (C). The application of carbendazim to plots A and C would avoid a biased interference of carbendazim due to other effects than reducing the natural yeast population. Prolonged differences in yeast density would be reflected in differences in severity of diseases caused by carbendazim-insensitive necrotrophic pathogens.Both treatments B and C enhanced the yeast density about 10-fold at the beginning of leaf colonization, 2 to 3 weeks after leaf emergence. Untreated leaves (D), however, were rapidly leaves. Initially, the yeasts were suppressed by carbendazim (A). However, progressively the yeast population consisted of carbendazim-resistant strains which made the carbendazim sprayings ineffective. Therefore, the substantial differences in yeast density were of limited duration. The effect of the treatments on naturally occurringMycosphaerella graminicola (anam.Septoria tritici), Puccinia recondita f. sp.tritici and the total necrotic leaf area (D-value) is discussed.Samenvatting Fyllosfeergisten kunnen misschien een belangrijke rol spelen in de onderdrukking van necrotrofe pathogenen. De grootste verschillen in dichtheden van de gistpopulaties werden verwacht tussen de behandelingen A en B of C. Door alle veldjes A en C met carbendazim te behandelen wordt een eenzijdig effect van carbendazim, berustend op andere eigenschappen dan de onderdrukking van de natuurlijke gistpopulatie, voorkomen. Langdurige verschillen in de populatiedichtheden van de gisten zouden weerspiegeld kunnen worden in verschillen in aantasting door carbendazimongevoelige necrotrofe pathogenen.Aan het begin van de bladkolonisatie door gisten, 2 tot 3 weken na het verschijnen van het blad, leidde zowel behandeling B als C tot, een 10-voudige verhoging van de gistdichtheid. De onbehandelde bladeren (D) werden echter spoedig door de van nature voorkomende gisten gekoloniseerd en het verschil tussen de onbehandelde en met gist bespoten bladeren werd snel kleiner. Aanvankelijk werden de gisten door carbendazim (A) onderdrukt. Geleidelijk bestond de gistpopulatie echter uit carbendazim-resistente stammen, zodat de carbendazim bespuitingen nauwelijks meer effect hadden. Hierdoor werden grote verschillen in gistdichtheden niet langdurig gerealiseerd. A series of experiments was conducted with wheat stripe rust to analyse competition between simple and complex pathotypes in host mixtures. Two different pathotype combinations were tested, with different host components. Each combination included a complex (able to infect two host components) and two simple pathotypes. For one of the combinations, induced resistance was tested in a separate experiment as a possible interaction among pathotypes. Disease severity and pathotype frequencies were measured three times during the epidemic, on each host component grown in pure stands and in mixtures. In one of the experiments, pathotype frequencies were also measured within secondary foci. One of the complex pathotypes appeared to have a low fitness on one of the host components and did not significantly increase in frequency in host mixtures relative to pure stands. The average frequency of the other complex pathotype increased during the first epidemic cycles, but remained stable afterwards, below expected values. The results suggest that the development of complex pathotypes in host mixtures may be influenced by differential aggressiveness on the host components, by induced resistance and by random effects resulting from the formation of disease foci, and depends on pathogen autoinfection rate and dispersal mechanisms.  相似文献

采用人工接种喷雾法对国家甘蔗种质资源圃保存的31份甘蔗野生资源材料进行了抗蔗茅柄锈菌(Puccinia erianthi Padw et Khan)的抗性鉴定。结果表明,31份材料中,高抗材料19份(61.3%),抗性材料3份(9.68%),中抗材料6份(19.36%),中感材料3份(9.68%)。其中斑茅种对蔗茅柄锈菌的抗病性较强,细茎野生种抗病性较差,较易感锈病。  相似文献
