The majority of the known prognostic factors in dogs with lymphoma have been evaluated before treatment commences or at the time of diagnosis. Prognostic factors evaluated during the initial phase of treatment are less described but may provide important clinical information. In this retrospective study, 82 canine lymphoma patients were categorized according to the weight change between diagnosis and after 5 weeks of chemotherapy. Dogs that gained greater than 5% or lost greater than 5% of initial body weight were categorized as increased‐ or decreased‐weight groups, respectively. Those in which weight changed less than 5% were categorized as the maintained‐weight group. The median progression‐free survival (PFS) in the increased‐weight group, maintained‐weight group and decreased‐weight group was 226, 256 and 129 days, respectively. The decreased‐weight group had significantly shorter PFS than the increased and maintained groups (P = .023, P = .003, respectively). The median survival time (ST) in the increased‐weight group, maintained‐weight group and decreased‐weight group was 320, 339 and 222 days, respectively. There was no significant difference in ST among the three groups (P = .128). In Cox‐regression results, weight change group and initial body weight were significant risk factors associated to PFS (P = .007, P = .001, respectively) while only patient's initial body weight was a significant risk factor to ST (P = .013). In conclusion, evaluation of initial body weight and weight changes over time can provide valuable information regarding PFS and ST in dogs with multicentric lymphoma. 相似文献

The aim of this retrospective study was to describe clinical features, treatment and outcome of 21 dogs with metastatic cancer of unknown primary (MCUP), a biopsy‐proven malignancy being diagnosed at a metastatic stage, in which the anatomical origin of the primary tumour cannot be detected. All dogs underwent total‐body computed tomography. Signalment, type and duration of clinical signs, metastasis site, pathology results, treatment and outcome were recorded. Carcinoma was the most common diagnosis (57.1%), followed by sarcoma, melanoma and mast cell tumour. The median number of disease sites per dog was 2, with bones, lymph nodes, lungs and spleen being the most frequent metastatic locations. The median survival for all dogs was 30 days. Overall, a primary site was not identified in 20 (95.2%) dogs. MCUP encompasses a variety of different pathologic entities and harbours a poor prognosis. 相似文献

Background Under anoxic conditions typically prevailing in disposal sites for dredged sediment, methanogenesis is the terminal step
during microbial degradation of sediment organic matter. Sediment gas production may pose several problems to site management
and post-closure utilisation. Depending on the magnitude of gas/methane formation and the intended utilisation of the site,
countermeasures will have to be effected during and after deposition of the dredged material. For this purpose it may be of
interest to pre-estimate the extent of gas formation from simple sediment variables. Therefore, the aim of the investigations
presented here was to analyse the interrelations between gas formation and sediment physical, chemical and biological qualities.
Methods Freshly sampled riverbed sediment from nine German federal waterways was analysed for standard solid physical and chemical
parameters, pore water composition, cell counts of methanogenic and sulfate-reducing bacteria, and gas formation over a period
of 500 days. Particle size and density fractionation were carried out in order to characterise organic matter quality. Correlations
between methane formation and sediment chemical, physical and microbiological characteristics were tested using linear and
multiple correlation analysis.
Results and Discussion The selected sediments, including two with marine influence, differed strongly regarding texture, chemical parameters, pore
water composition, and methanogen cell counts. The course of methane formation was found to follow distinct phases. The commencement
of methane formation was preceded by a lag phase of variable duration. The lag phase was followed by a strong increase of
the methane formation rate up to a sediment-characteristic maximum of 5–30 nmol CH4 h–1 g dw–1. Eventually, the rate of methane
formation decreased and reached a more stable, long-term level. The extent and amplitude of each phase varied strongly between
sediments and could be correlated well with only a few standard analytical parameters, despite the strong heterogeneity of
sediment with respect to chemical, physical, and biological characteristics. Lag phase duration depended strongly on the content
of inorganic electron acceptors and also on the number of methanogens present at the beginning of the experiment. Maximum
and steady state methanogenesis were mainly determined by sediment total nitrogen. As analysis of sediment density fractions
revealed that total nitrogen reflected the share of readily degradable macroorganic matter.
Conclusion and Outlook The results imply that the observed methane formation is a function of the temporally changing balance of the availability
of electron acceptors and H2, and the share of easily degradable organic matter. For fresh riverbed sediments, the latter
may be deduced from total nitrogen content. Overall, the results showed that methane formation by freshly dredged material
may well be pre-estimated from standard analytical data within the first few years of deposition. However, the differently
degradable organic matter pools will change over time with respect to size, chemical nature and association with the sediment
mineral phase. It can thus be expected that the correlations found in this study will not be readily transferable to older
materials. Further studies on the gas formation and organic matter quality by older sediments, e.g. from older dredged material
disposal sites of known age, should be conducted and results should be combined with existing organic matter degradation models
in order to improve the prediction of sediment gas formation over time. 相似文献

Objective – To test whether an initial plasma lactate≥6.0 mmol/L is associated with the presence of macroscopic gastric wall necrosis and overall survival in dogs presenting with gastric dilatation‐volvulus (GDV). Additionally, if no association was identified we sought to identify a different predictive initial plasma lactate concentration and to examine whether serial plasma lactate concentrations provide better prediction of survival. Design – Retrospective study over a 5‐year period (2003–2007). Setting – Urban private referral small animal teaching hospital. Animals – Eighty‐four client‐owned dogs with a diagnosis of GDV and plasma lactate measurements. Interventions – None. Measurements and Main Results – There was no statistically significant relationship found between survival and the presence of macroscopic gastric wall necrosis with the initial plasma lactate≥6 mmol/L. There was a significant relationship between the initial plasma lactate >2.9 mmol/L for predicting necrosis and <4.1 mmol/L for predicting survival to discharge. Forty dogs that had an increased initial plasma lactate (>2.5 mmol/L) also had a subsequent plasma lactate measured within 12 hours of presentation, with 37/40 dogs surviving and 70% of these surviving dogs having the subsequent lactate decrease by≥50% within 12 hours. The 3/40 that died failed to decrease their plasma lactate by≥50% from the initial blood lactate. Conclusion – The results of this study indicate that an initial presenting plasma lactate concentration≥6.0 mmol/L is not predictive of macroscopic gastric wall necrosis or survival in dogs presenting with GDV. A decrease in plasma lactate concentrations≥50% within 12 hours may be a good indicator for survival. Limitations to the study include its retrospective nature, the small number of patients, and the number of dogs that were euthanized rather than allowed to progress to a natural outcome. 相似文献

KIT, a transmembrane receptor tyrosine kinase, is one of the specific targets for anti-cancer therapy. In humans, its expression and mutations have been identified in malignant melanomas and therapies using molecular-targeted agents have been promising in these tumours. As human malignant melanoma, canine malignant melanoma is a fatal disease with metastases and the poor response has been observed with all standard protocols. In our study, KIT expression and exon 11 mutations in dogs with histologically confirmed malignant oral melanomas were evaluated. Although 20 of 39 cases were positive for KIT protein, there was no significant difference between KIT expression and overall survival. Moreover, polymerase chain reaction amplification and sequencing of KIT exon 11 in 17 samples did not detect any mutations and proved disappointing. For several reasons, however, KIT expression and mutations of various exons including exon 11 should be investigated in more cases. 相似文献
