The levels of the raffinose family of α-galactosides (raffinose, stachyose, verbascose and ciceritol) and of the nutritional carbohydrates (fructose, sucrose and starch) were determined in the seed of a range of lentil lines from germplasm collections and commercial Spanish cultivars. Significant levels of variation were found for all the measured characters, and particularly for the α-galactosides. The greatest variation was found in verbascose content, which ranged from about 1 % of the seed dry weight to undetectable amounts. The relationship between these chemical components and morphological seed characteristics was assessed. The only significant relationship between the different groups of characteristics was a positive correlation between the level of verbascose and the average seed weight for each genotype.  相似文献

【目的】研究甜瓜果实发育期坐果节位叶片光合能力、蔗糖合成能力和水苏糖装载能力对遮光的响应,分析不同耐弱光性甜瓜品种同化物输出能力的差异,为进一步研究甜瓜耐弱光品种果实糖分卸载与积累机理奠定基础。【方法】以耐弱光甜瓜品种‘玉金香’和不耐弱光品种‘钰雪三号’为试材,在日光温室条件下于授粉后进行遮光处理,每5 d取样一次,取坐果节位叶片测定叶绿素含量、气体交换参数、葡萄糖、果糖、蔗糖、肌醇半乳糖苷、棉子糖、水苏糖和淀粉含量,同时测定叶片蔗糖磷酸合成酶(SPS)、蔗糖合成酶(SS)、中性转化酶(NI)、酸性转化酶(AI)、肌醇半乳糖苷合成酶(GS)和水苏糖合成酶(STS)活性。【结果】遮光处理后,两个甜瓜品种叶片叶绿素a/b、净光合速率(Pn)、葡萄糖、果糖和蔗糖含量均显著降低,淀粉含量升高,‘玉金香’叶绿素a/b降幅(10.0%)大于‘钰雪三号’(5.8%),而Pn、蔗糖含量降幅(分别为30.3%和30.9%)和淀粉含量增幅(3.6%)均显著小于‘钰雪三号’(分别为45.2%、60.6%和20.4%)。遮阴条件下,两个品种叶片蔗糖代谢相关酶SPS、SS、AI和NI活性均显著降低,‘玉金香’SPS和SS酶活降幅(分别为16.5%和30.0%)显著小于‘钰雪三号’(31.6%和40.5%),而AI和NI降幅(分别为23.8%和12.7%)高于‘钰雪三号’。遮光后甜瓜叶片肌醇半乳糖苷含量和GS活性均显著降低,但品种间差异不显著。棉子糖、水苏糖和STS活性均显著降低,‘玉金香’棉子糖含量降幅(65.3%)显著高于‘钰雪三号’(35.0%),而水苏糖和STS活性降幅(分别为79.5%和23.8%)小于‘钰雪三号’。【结论】遮光条件下,耐弱光品种‘玉金香’叶片蔗糖合成能力和水苏糖装载能力下降较少,具有较强的同化物输出能力。  相似文献

Abstract

Selected organic and mineral analyses of common beans, peanut kernels, and cowpeas were determined. Simple correlations were computed between organic and mineral components to investigate possible linear relationships that might be useful. Most correlations between mineral and organic components were negative, and relatively low coefficients of determination (r2) indicated little merit in attempting to predict organic components from inorganic compositions and sample linear models. A positive correlation between stachyose and the N content of common beans was observed.  相似文献

This study was conducted to examine the effects of soybean oligosaccharide, stachyose, and raffinose on growth performance and cecal microbiota in broiler chickens. Three-hundred 1-day-old Arbor Acres broiler chickens were randomly assigned to five dietary treatments: a basal diet (control diet); diets supplemented with soybean oligosaccharide, stachyose, or raffinose at levels of 0.6% total sugar content; and diet with soybean meals (positive control). Results showed that feed-to-gain ratio (F/G) was improved (p < 0.05) in broiler chickens that received stachyose, but not raffinose, while addition of soybean oligosaccharide to the diet significantly (p < 0.05) resulted in decreases in the average daily feed intake (ADFI) and average daily gain (ADG). Supplementation with soybean oligosaccharide, stachyose, or raffinose had a positive effect (p < 0.05) on the nutrient digestibility, but not on the blood immune parameters (p > 0.05). The results of the gene sequencing indicated that, at the family level, Ruminococcaceae, Lachnospiraceae, and Lactobacillaceae were comparatively present in all the treatments but the cecal microbial composition was changed after dietary addition of different oligosaccharides. Our study had led to a greater understanding of prebiotic effects of these oligosaccharides on growth and intestinal health.  相似文献

Levels of carbohydrates and activities of metabolic enzymes were examined in leaves (source), phloem sap (flow) and mesocarp tissues (sink) in the course of cucumber (Cucumis sativus L.) fruit development, from 2 days before anthesis to 20 days after anthesis. While total sugar levels increased in all the three sampling organs, starch levels declined in leaves and mesocarp tissues as fruit development progressed. Glucose and fructose were the primary contributors to the soluble sugar pools in mature leaves. Stachyose was found as the most important component of the phloem sap extracts, followed by sucrose and raffinose. However, the primary sugars accumulated in mesocarp tissues were glucose and fructose, not stachyose or sucrose. Activities of sucrose synthesizing enzymes (sucrose phosphate synthase plus sucrose synthase in the synthesizing direction) exceeded that of sucrose degrading enzymes (acid invertase, neutral invertase plus sucrose synthase in the degrading direction) in leaves, which might cause a sucrose pool utilized in raffinose and stachyose biosynthesis. While alkaline a-galactosidase form I activity declined, stachyose synthase activity showed a rapid increase until 12 days after anthesis and only subsequently decreased in leaves. Activities of sucrose degrading enzymes were always much higher than that of sucrose synthesizing enzymes in mesocarp tissues. Thus, sucrose accumulation could not occur in mesocarp tissues. While stachyose synthase activity steadily decreased, alkaline a-galactosidase form I activity showed a moderate increase before decrease in mesocarp tissues. The relationship between levels of soluble sugars and activities of relative enzymes was also discussed.  相似文献

The soluble sugar content in soybean seeds, mainly sucrose, stachyose, raffinose and trace amounts of glucose and fructose, is important for the increasing global market demand for various soyfoods including tofu, soymilk, natto, bean sprouts and edamame due to their nutritional value and health benefits. The objective of this study was to conduct quantitative trait loci (QTL) analysis and identify molecular markers for soluble sugar content in soybean seeds for marker‐assisted selection (MAS) in soybean breeding. The content of the five previously mentioned sugars were measured and associated QTLs were mapped based on a F2 population that was derived from a cross between V97‐3000 and V99‐5089. Eleven QTLs were detected for the five sugar contents: one for glucose, three each for fructose and sucrose, and two each for raffinose and stachyose. However, only one QTL for sucrose, one QTL for raffinose, and two QTLs for stachyose were identified with LOD > 3.0 and R2 > 10% from this research. The QTL on chromosome 11 [linkage group (LG) B1] was identified as associated with sucrose, raffinose and stachyose in the same region as previously reported for sucrose and stachyose. The SSR marker, Satt359, on the QTL B1 region had an significant association with sucrose (LOD = 5.192; R2 = 0.134), raffinose (LOD = 3.95; R2 = 0.104), and stachyose (LOD = 13.572; R2 = 0.314); therefore it can be used to assist breeding selection for sucrose, raffinose and stachyose contents simultaneously.  相似文献

Abstract

Objectives in this study were to measure total sugar and individual sugar content in whole, mature, soybeans of certain strains and to determine the stability of these components in strains grown over a wide geographical area. Soybean seed of three selected strains from each Maturity Group in the 1970 Uniform Tests‐Northern States were analyzed for total sugar, sucrose, raffinose and stachyose content. For Maturity Groups 00, 0, I, II, III and IV there were 10, 12, 23, 33, 27 and 24 locations, respectively. The locations were in 17 states and 2 Canadian provinces.

The average content of total sugar, sucrose, raffinose and stachyose was 9.36, 5.96, 0.75 and 2.65 g per 100 g seed, respectively. Analysis of variance of total sugar content and individual sugars among the three strains within each Maturity Group revealed significant differences (P<.01) in total sugar among strains in Maturity Groups II and III but not in the other four Maturity Groups. Except for strains within Maturity Group IV, there were significant differences (P<.01 or P<.05) among the strains within each Maturity Group for sucrose, raffinose and stachyose. Few significant environmental index (linear) x strain interactions were observed for total sugar content or for any of the individual sugars.  相似文献

The oligosaccharide profile of raw mature seeds of seven different legumes indigenous to the Philippines was measured in 70% ethanol extracts of the seeds by thin layer chromatography using HPTLC plates and quantified by a densitometer. Based on the results, the legumes could be ranked according to decreasing oligosaccharide content or flatulence potential as follows: Sam-samping (Clitoria ternatea)>hyacinth bean (Dolichos lablab)> sabawel (Mucuna pruriens)>lima bean (Phaseolus lunatus)>swordbean (Canavalia gladiata)>rice bean (Vigna umbellata)>jack bean (Canavalia ensiformis). Sam-samping had 4.79% total oligosaccharides and hyacinth bean or batao, 3.66%. A jack bean accession had 1.79% oligosaccharides.Simple processing methods were tested to detoxify the oligosaccharides. Soaking the batao seeds had no effect while boiling even resulted in a net 23–31% increase in the levels of raffinose, stachyose and verbascose. On the other hand, two min of dry roasting resulted in complete removal of oligosaccharides whereas germination resulted in about 30–40% decrease after 1 and 2 days, respectively.Part of MS thesis of the senior author.  相似文献

Stachyose is an unfavorable sugar in soybean meal that causes flatulence for non‐ruminant animals. Understanding the genetic control of stachyose in soybean will facilitate the modification of stachyose content at the molecular level. The objective of this study was to identify quantitative trait loci (QTL) associated with seed stachyose content using simple sequence repeat (SSR) and single nucleotide polymorphism (SNP) markers. A normal stachyose cultivar, ‘Osage’, was crossed with a low stachyose line, V99‐5089, to develop a QTL mapping population. Two parents were screened with 33 SSR and 37 SNP markers randomly distributed on chromosome 10, and 20 SSR and 19 SNP markers surrounding a previously reported stachyose QTL region on chromosome 11. Of these, 5 SSR and 16 SNP markers were used to screen the F3:4 lines derived from ‘Osage’ x V99‐5089. Seed samples from F3:5 and F3:6 lines were analyzed for stachyose content using high‐performance liquid chromatography (HPLC). Composite interval mapping analysis indicated that two stachyose QTL were mapped to chromosome 10 and 11, explaining 11% and 79% of phenotypic variation for stachyose content, respectively. The SSR/SNP markers linked to stachyose QTL could be used in breeding soybean lines with desired stachyose contents. Chi‐square tests further indicated that these two QTL probably represent two independent genes for stachyose content. Therefore, a major QTL was confirmed on chromosome 11 and a novel QTL was found on chromosome 10 for stachyose content.  相似文献

试验研究了添加不同梯度水苏糖对肉鸡鼠伤寒沙门氏菌攻毒的控制效果。试验选用1日龄体重相近(45.52±1.48)g的健康AA肉鸡公雏72只,随机分为3个处理,设6个重复,每个重复4只。在玉米-大豆浓缩蛋白日粮基础上分别添加0、0.4%和0.8%水平的水苏糖。结果表明:日粮添加水苏糖降低了肉鸡体重;肉鸡肠道沙门氏菌控制效果不显著,第19天时,0.4%水苏糖添加组的大肠杆菌数显著高于其他2组(P<0.05),乳酸菌各处理之间差异不显著;pH值与盲肠指数无差异(P>0.05)。以上研究表明,该梯度水苏糖不能控制肉鸡沙门氏菌感染,对肉鸡无保护作用。  相似文献
