The denitrifying woodchip bioreactor is designed to remediate nitrate-rich water, including those produced from aquaculture effluents. Reuse of treated bioreactor outflows in recirculating aquaculture would offer considerable water savings and valuable alkalinity recuperation. However, such bioreactors may leach detrimental wood-bound contaminants, preventing outflow reuse. To determine water reuse potential, woodchip media from two hardwood species (white ash, Fraxinus americana; Norway maple, Acer platanoides) were evaluated for 206 d under a range of operating conditions (start-up, steady-state, reducing conditions, and drying-rewetting cycles) for a spectrum of potentially harmful dissolved contaminants. Aerated outflows also were evaluated for acute and chronic toxicity to the biologically sensitive invertebrate Ceriodaphnia dubia. Dissolved metal leaching subsided within the first few weeks of operation, though initial concentrations of copper and zinc were detected at concentrations of concern. Elevated concentrations of tannins-lignin and total ammonia nitrogen were detected throughout the study and were influenced by operational phase. Acute toxicity was not generally detected, though chronic toxicity was observed during drying-rewetting cycles in the maple outflows. The measured toxicity was not correlated with water chemistry, indicating an additive effect of several toxicants. Overall, significant differences in outflow water quality between ash and maple wood species were negligible. Results indicated that bioreactor outflows may be applicable for aquacultural reuse, though reusing outflows immediately following start-up or restarting after a dry period would not be recommended. 相似文献

Two isoforms of the full-length cDNA of the growth hormone receptor (GHR) of the Atlantic salmon (Salmo salar; ss) were cloned by a PCR approach using RACE. Respectively, the cDNA sequences of ssGHR isoforms 1 and 2 are 2654 and 2608 nucleotides long, with 1782 and 1773 nucleotide ORFs. The resulting coded proteins
are 594 and 590 aa long, with 19 and 20 aa signal peptides. The two isoforms share 86% protein and 87% cDNA sequence similarity.
Isoform 1 is most similar to other salmonid GHR isoforms 1 while isoform 2 is most similar to salmonid GHR isoforms 2 (93–95%).
Similarity with other teleost species was lower (37–44%). The bioactivity of the cloned ssGHR was tested by transfecting the ssGHR isoform 1 cDNA into CHO-K1 hamster cells, incubating with recombinant salmon GH (sGH) or native ovine prolactin (oPRL),
and measuring cell proliferation by the MTT assay. The ssGHR-transfected cells significantly increased proliferation when stimulated by sGH at all concentrations. oPRL stimulated
ssGHR-transfected cells at higher concentrations due to receptor cross reaction. ssGHR isoforms 1 and 2 contain a single transmembrane domain and the typical conserved motifs found in other teleost GHRs, including
four paired cysteine residues and five potential N-glycosylation sites in the extracellular domain, Box I and Box II, as well as seven potential tyrosine phosphorylation sites
in the intracellular domain. However, in salmonids, these motifs differ from those of other teleosts, and could be responsible

An experimental design and statistical analysis providing information on the reliability of pooled test procedures is described. It involves estimating the relationship between the probability of a positive pooled test result (dependent variable) and the expected number of infected individuals in a pool (explanatory variable). The intercept is an estimate of the proportion of false positives (1 − pooled specificity) and pooled sensitivities can be estimated for indicative prevalences of infected individuals. Simulations for a theoretical infection are used to investigate the advantages and limitations of the approach. The approach is used to evaluate the reliability of a virus isolation and qRT-PCR test procedure detecting Salmonid alphavirus the pathogenic agent necessary for the development of Pancreas Disease in Atlantic salmon (Salmo salar). 相似文献

We studied partial replacement of fish meal (FM) by krill meal (KM) and how fluoride from KM enriched-muscles and bones of rainbow trout reared in freshwater affected these fish. Diets that replaced FM with KM at proportions of 0, 7, 15 and 30% (control, KM7, KM15, and KM30) were fed to groups of rainbow trout for 92 days and growth was observed. In fish fed KM7 and KM15, weight gain (WG), feed intake (FI) and specific growth rate (SGR) were unchanged compared with fish fed the control diet, but in fish fed KM30, WG, FI and SGR significantly decreased. After the experiment, fluoride concentration in dorsal muscles of each experimental group was below the detected limit (1 mg/kg), but in vertebral bones, the fluoride concentration increased with each increase in KM inclusion in the diets: 490 mg/kg (control), 755 mg/kg (KM7), 1100 mg/kg (KM15), and 2400 mg/kg (KM30). Tissue specimens of liver of each experimental group had no histopathological changes. Therefore, accumulation of fluoride in vertebral bones apparently adversely affected growth of the vertebral bones thus affecting the growth performance as shown by the decrease in WG, FI and SGR. 相似文献

The composition and structure of riparian forests in the coast redwood region were analyzed in relation to the length of time since timber harvest, and the width of riparian buffer zone. Ten sites were sampled in the central range of the coast redwood forest type within a variety of post-harvest age groups and riparian buffer zone widths. Data was collected using randomly selected sample plots adjacent to perennial coastal steams. Correlation Fisher's r to z tests and two-tailed t-test were used to examine the relationship between the sample variables and the experimental parameters “years since harvest”, and “width of buffer.” Results indicate that canopy cover was negatively correlated to “years since harvest” with the highest level of canopy cover found on the youngest sites and the lowest level found on the old-growth sites. The hardwood to conifer dominance ratio and the basal area of Alnus rubra (red alder) were correlated negatively to both “years since harvest” and “buffer width” indicating that timber harvest favored hardwood species. Late seral associates such as Oxalis oregana (redwood sorrel), Anthyrium filix-femina (lady fern), and Vaccinium parviflorum (billberry) were found preferentially in older forests and sites with wider buffer zones, while non-native species such as Hedera helix (English ivy), Pampas cortedaria (pampas grass), and Myosotis latifolia (forget-me-not) were found preferentially in younger forests and areas with smaller buffer zones. 相似文献

Predator conditioning can be used to improve post‐release antipredator recognition of hatchery‐reared salmonids. However, possible negative stress‐related effects of prolonged predator conditioning on juvenile fish physiology are poorly understood. We studied the effects of prolonged (91 days) predator odour exposure on whole‐body cortisol level and spleen size in six full‐sib families of juvenile hatchery‐bred Arctic charr (Salvelinus alpinus). Chemical cues from water containing charr‐fed pikeperch (Sander lucioperca) were used as the predator exposure stimuli and lakewater was used as a chemical control. Our study revealed that juvenile body cortisol levels post‐predator conditioning were affected by treatment, fish size and their interaction. Importantly, among the smaller (i.e. slowest growing) charr, the predator‐exposed fish had higher cortisol levels than control fish, while the opposite pattern was true for the larger fish. These results suggest that chemical cues from charr‐fed predators induce a prolonged stress response in juvenile charr. As prolonged predation exposure seems to elevate stress levels in a size‐dependent manner, the larger, faster growing fish could possibly have intrinsically lower stress responses to predation threats than smaller, slower growing fish. Possible coupling between stress sensitivity and growth requires further attention due to the likely implications for the management of unintended domestication among captive‐reared salmonids. 相似文献

One of the key factors in determining the accumulation of sediment and soluble nutrients within a lease is water movement. Water masses entering and leaving the farm determine the finfish ecosystem interaction. Understanding the hydrodynamic interaction with the farm is therefore key to understanding the potential ecological effects of individual farms. In addition, finfish farms are now being proposed in exposed offshore environments and have caused concern regarding their potential down stream impacts on currents and wave climate. Seven current meters, oxygen probes and CTD were deployed to examine the hydrodynamic interactions inside and outside a 270 m long Salmon farm in Newfoundland, Canada. Current meter results indicate that the finfish farm cages have a clear shadowing effect on the currents. Currents upstream were found to be considerably faster than those recorded downstream during the sampling period. Current speeds inside the farm were also found to be considerably slower than those found outside of the farm especially during high flow events. In situ observations of currents were found to be similar to those predicted by previous CFD and hydrodynamic modelling studies. Modeling was also undertaken to calculate the energy lost as currents enter and leave a series of fish cages. In comparison to the observed flow the model compares relatively well. Flow recorded downstream of the farm was observed to be in the range predicted by the model as was the flow recorded inside and outside the cages. Current speed downstream of the farm is clearly affected by farm orientation which has important implications for the dispersal of farm debris. Average oxygen saturation within the cages over the 5 day sampling period was 80.2 %± SD 5.7 %, compared to nearly 100 %, 20 m and 50 m from the farm site. Orientation of the farm may play some part in determining the location and amount of oxygen depletion within the farm. The farm also acts to push water from deeper in the water column up into the cages which has implications for farms situated in heavily stratified environments. 相似文献
