The survival of Leptosphaeria maculans , which causes phoma stem canker (blackleg), on oilseed rape residues ( Brassica napus ) in South Australia was investigated. Using a quantitative polymerase chain reaction (PCR) assay for L. maculans DNA, the pathogen was mainly detected in the upper 5 cm of the soil profile, including residues on the soil surface. As the size of organic matter particles in the soil decreased, so did the quantity of L. maculans detected in them. To obtain representative data for a field, at least 30 subsamples needed to be collected over the 0·81 ha area studied. In a survey of 49 commercial fields in South Australia, most L. maculans was detected in fields 1 year after oilseed rape had been grown, with less detected after 2 years and negligible amounts 3 years or more after cropping. The diagnostic DNA-based assay for L. maculans reduced the time and cost of studying L. maculans survival in soil and increased the sensitivity and accuracy of results compared with estimates of propagule number of colony-forming units on a semiselective medium.  相似文献

In controlled environment experiments, sporulation of Pyrenopeziza brassicae was observed on leaves of oilseed rape inoculated with ascospores or conidia at temperatures from 8 to 20°C at all leaf wetness durations from 6 to 72 h, except after 6 h leaf wetness duration at 8°C. The shortest times from inoculation to first observed sporulation ( l 0), for both ascospore and conidial inoculum, were 11–12 days at 16°C after 48 h wetness duration. For both ascospore and conidial inoculum (48 h wetness duration), the number of conidia produced per cm2 leaf area with sporulation was seven to eight times less at 20°C than at 8, 12 or 16°C. Values of Gompertz parameters c (maximum percentage leaf area with sporulation), r (maximum rate of increase in percentage leaf area with sporulation) and l 37 (days from inoculation to 37% of maximum sporulation), estimated by fitting the equation to the observed data, were linearly related to values predicted by inserting temperature and wetness duration treatment values into existing equations. The observed data were fitted better by logistic equations than by Gompertz equations (which overestimated at low temperatures). For both ascospore and conidial inoculum, the latent period derived from the logistic equation (days from inoculation to 50% of maximum sporulation, l 50) of P. brassicae was generally shortest at 16°C, and increased as temperature increased to 20°C or decreased to 8°C. Minimum numbers of spores needed to produce sporulation on leaves were ≈25 ascospores per leaf and ≈700 conidia per leaf, at 16°C after 48 h leaf wetness duration.  相似文献

Orobanche ramosa is a parasitic Angiosperm responsible for severe yield losses in several economical crops. It is a serious threat in oilseed rape in France and Morocco and is appearing in carrot crops in Morocco. In this study, several varieties of oilseed rape and carrot were screened in order to identify resistant cultivars and to characterize the resistance mechanisms involved. All the 15 oilseed rape varieties tested were susceptible. In carrot, the varieties 'Colmar à coeur rouge' and 'Nantaise demi-longue' were susceptible, whereas 'Palaiseau' and 'Buror' were resistant. In the susceptible 'Colmar à coeur rouge' carrot no defence reactions were found and the development of the parasite inhibited carrot tap root formation. In the resistant carrot varieties, the parasite germinated, became attached to the host root but became necrotic before emergence. In 'Buror' carrot, formation of a mechanical barrier was associated with the restriction to the cortex of the parasite. In maize cv. 'Vigni', a non-host of O. ramosa, thickening of xylem vessels, cell divisions in the central cylinder and formation of an encapsulation layer were observed in association with restricted development of Orobanche haustoria.  相似文献

为探明安徽省直播冬油菜种植中适宜的氮肥用量、氮肥种类和施氮方式,通过田间小区试验,研究尿素不同用量及等氮量尿素和控释尿素不同施用方式对直播冬油菜产量、氮肥累积量及氮肥利用率的影响。结果表明:施氮水平和施氮方式对直播冬油菜产量具有显著影响,成株率与产量之间呈显著正相关,施氮量180 kg N·hm-2条件下,尿素分次施用处理(N180)和控释尿素一次性施用处理(CRU180)在生育中后期能有效提高直播冬油菜密度。施氮量240 kg N·hm-2、尿素分次施用处理(N240)角壳和茎秆氮素累积量分配比例增加,籽粒分配比例减少,氮肥表观利用率、氮肥农学效率、氮肥生理效率和氮肥偏生产力显著降低,籽粒产量不增反降,较CRU180和N180分别降低2.87%和9.67%。各施氮处理中,N180处理产量最高,较其他施氮处理显著增产7.53%~82.15%。施氮量180 kg N·hm-2条件下,相比尿素一次性施用处理(U180),CRU180和N180显著增产8.53%和16.69%,籽粒氮素累积量显著增加8.80%和16.02%,氮肥表观利用率显著增加11.68%和14.30%,氮素农学效率显著增加12.53%和24.46%,氮素生理效率增加0.74%和9.13%,氮肥偏生产力显著增加8.59%和16.76%。综合本试验直播冬油菜产量、氮素累积量和氮肥利用率结果来看,合理的氮肥用量和氮肥品种可以有效增加直播冬油菜的成株率和氮素累积量,进而增加产量、提高氮肥利用率。安徽省直播冬油菜施氮量180 kg N·hm-2,尿素分次施用和控释尿素一次性施用,均能达到产量和氮肥利用率的双向提升。考虑到直播冬油菜轻简化发展大趋势,建议采用控释尿素一次性基施。  相似文献

The field experiments conducted on the grey‐brown podzolic soil in the four growing seasons (1998–2001) at Krzeslice Farm, central‐western Poland comprised seven fertilization variants: 80NF + 80CAN; 80CAN + 80CAN; 80AN + 80AN; 80NF + 50CAN + 30CN; 80CAN + 50CAN +30CN; 80AN + 50AN + 30CN (where NF – nitrofos NPK; CAN – calcium‐ammonium nitrate; AN – ammonium nitrate; CN – calcium nitrate) and control (without N) applied in split rates at the beginning of spring regrowth (80 kg N ha?1), stem elongation (80 or 50) and flower buds visible stages (30). The yielding effect of tested fertilization variants was significant in comparison with the control (2.24 t ha?1). The highest mean seed yield (3.64 t ha?1) was collected from 80AN + 80AN and 80CAN + 80CAN variants. Mean values of 4 years indicate that the second N rate division (80 + 50 + 30) decreased yield, although not significantly in comparison with these two N treatments. Plants grown on these treatments have developed different patterns of growth to yield the seeds. These patterns were characterized by very high crop growth rate during flowering (above 21 g m?2 day?1) and negative at maturation (down to ?2.5 g m?2 day?1). Plants fertilized with ammonium nitrate (80AN + 80AN) reached maximum growth rate earlier (65 days), which lasted longer (20 days) than plants fertilized with calcium‐ammonium nitrate (71 days lasting 17.5 days). A sugar-beet-infecting isolate of beet mild yellowing luteovirus (BMYV), and aBrassica-infecting isolate of beet western yellows luteovirus (BWYV) were used to produce monoclonal antibodies for epidemiological studies with BMYV and related field strains. Thirty-four monoclonal antibodies were tested for their reaction with 9 luteoviruses in triple-antibody-sandwich enzyme-linked immunosorbent assay. One (MAFF 24) is now routinely used in the UK for detecting BMYV and BWYV in plants and aphids, although it does not discriminate between them. Heterologous reactions were detected between some of the monoclonal antibodies and potato leafroll virus (PLRV), bean leafroll virus (BLRV) and barley yellow dwarf virus (BYDV-RPV). 38% of antibodies raised to BWYV reacted with PLRV compared with 4% of those raised to BMYV. Monoclonal antibodies were produced which distinguished a sugar-beet-infecting isolate of BMYV with differing host range and serological properties from the commonly-occurring field strain.  相似文献

研究了油菜秸秆作为基质培养植物病原菌核盘菌的重寄生菌盾壳霉分生孢子,并从盾壳霉分生孢子萌发及其抑制核盘菌菌核子囊盘萌发等方面评价了所获得的盾壳霉分生孢子的质量。结果表明:盾壳霉野生菌株Chy-1和Zs-1,以及Chy-1的突变菌株SV-5-2(抗杀菌剂vin-clozolin)可以利用油菜秸秆为基质进行培养,有利于3个菌株的菌丝生长、分生孢子器及分生孢子的产生,分生孢子产量可达2·4×109~3·4×109个孢子/g干秸秆。水分含量和发酵时间影响盾壳霉分生孢子产量。在接种量为5×105个孢子/g干秸秆的条件下,以干秸秆中含水量为3~6ml/g,在20℃下发酵12d较为适宜。水琼脂平板试验表明:在20℃下培养48h,盾壳霉分生孢子的萌发率达到90%以上。将油菜秸秆基质培养的盾壳霉分生孢子接种于土壤中,无论是夏季试验,还是秋季试验,对核盘菌菌核萌发及存活具有显著的抑制作用。2003年夏季,4·0×106个孢子/m处理其核盘菌菌核萌发数比对照减少26·3%;秋季该处理比对照减少57·1%~88·0%。  相似文献

2003年秋至2005年春2个油菜生长季分别到湖北和安徽2省12县市油菜产区进行病毒病发病情况调查,除局部地区和地块发病率达到20%~40%,多数大面积油菜发病率在0.1%以下。应用间接酶联免疫吸附法检测258份病害样品,TuMV占样品总数的90.7%,CMV占样品总数的8.9%,ORMV占样品总数的0.8%。  相似文献

 从油菜植株体内分离出的内生细菌BY-2,经过生物学鉴定为枯草芽孢杆菌(Bacillus subtilis)。用BY-2回接油菜,重新分离到具有BY-2相同形态特征和抑制病原真菌能力的内生菌株。油菜接种后的第10 d,体内的BY-2菌数达(2.24~9.02)×103 cfu/g鲜植株,25 d仍然保持在(3.13~8.59)×103 cfu/g鲜植株。BY-2与油菜核盘菌[Sclerotinia sclerotiorum(Lib.) de Bary]对峙培养可以形成直径为3.1 cm的抑菌圈;可使油菜核盘菌菌丝细胞浓缩变短,细胞壁破裂,原生质外溢,从而抑制真菌生长发育;同时还能抑制菌核的萌发,抑制率达60%~70%;在油菜离体叶片试验中,BY-2对菌核病的防治效果达100%。  相似文献
