A vineyard of Vitis vinifera cv. Sangiovese was surveyed for incidence of esca and xylem sap collection. Sap samples were collected from healthy vines and from those with dual infection by Phaeomoniella chlamydospora (Pch) and Togninia minima (Tmi) or triple infection by Pch, Tmi and Fomitiporia mediterranea (Fme), during each early spring in a 3-year period (2001–2003). In order to analyse the possible trends in the climatic data, temperature and rainfall were assessed. At sap harvesting, aliquots of sap were assayed for phytotoxicity and extracted with ethyl acetate for phytotoxin recovery. Moreover, the exopolysaccharide (EPS) content was evaluated on several sap samples during the bleeding period. Conidia of Pch and Tmi, mycelium of Fme and their secondary metabolites were found in the sap of the esca-affected vines, indicating that the pathogens and their by-products together with some defence substances were accumulated and then translocated. Bioactivity tests showed toxicity of the sap from esca-affected vines to healthy detached leaves of cv. Sangiovese. The daily amount of sap, the pH, and the volume (Jv) and solute (Js) fluxes were analysed as a function of the infecting fungi. Pullulan, glucogalactomannan(s) and arabinogalactan(s) are the main EPS in the esca-infected vines, whereas in the sap of healthy vines no traces of pullulan were found. Scytalone and isosclerone usually produced in vitro by Pch and Tmi were also detected in the sap of vines infected by Pch and Tmi or by Pch, Tmi and Fme. The endogenous phytohormone content of healthy vines evaluated by the cutting bioassay was different from that of infected vines. Four phenolics belonging to three classes e.g., benzoic acid derivatives, stilbenes and flavonol-glycosides were separated and identified by HPLC.  相似文献

Nodal root anatomy was compared among twelve upland and lowland rice (Oryza sative L.) varieties with tropical origin which were grown in hydroponic culture and under field conditions. The traditional upland japonica varieties showed the largest diameter of root, stele, and xylem vessel followed by modern upland varieties. There was a clear varietal difference in the ratio of stele to root diameter, which was associated with the genetic group rather than with the ecosystems. The japonica varieties had a significantly larger stele diameter relative to the root diameter than indica and aus varieties. The indica and aus varieties displayed more xylem vessels per unit area of stele than the japonica varieties, but the diameter of xylem vessel was smaller. Equivalent xylem vessel diameter (De) was more dependent on the number of xylem in the indica varieties than in the japonica varieties. Distinctly different types of sclerenchyma anatomy were identified among the varieties. The development of sclerenchyma was classified into four different types based on thickening of cell wall in the outer cortical parenchyma and the number of sclerenchymatous cell layers. Like the xylem anatomy, the varietal differences in sclerenchyma development were more associated with genetic group rather than the ecotype. The japonica varieties had higher frequency of the types which have a doubled cell layer in sclerenchyma with thick cell wall than indica and aus. The difference among the genetic groups was nearly consistent across growing conditions, aerobic and submerged soils. These results indicated that sclerenchyma development is controlled by a genetic factor.  相似文献

Abstract

The objective of this study was to clarify the effects of soil temperature in the stage from late tillering to panicle initiation (SI) and during the grain-filling stage (SII) on grain setting, dry matter production, photosynthesis, non-structural carbohydrate (NSC), xylem exudation and abscisic acid (ABA) levels in rice (Oryza sativa L. cv. Koshihikari). Rice plants were exposed to four different soil temperatures during SI or SII: 17.5, 25, 31.5 and 36.5°C (ST18, ST25, ST32 and ST37, respectively). The yield, yield components, grain filling and quality in SI were negatively influenced by high soil temperature of 37°C. On the other hand, there was no significant difference in those characters among temperature treatments in SII. The root/shoot ratio was smallest in the ST37 plants in both SI and SII, mainly due to their lighter root weight. At 7 days after initiation of treatment (DAT) in both SI and SII, the photosynthetic and xylem exudation rate tended to increase slightly as soil temperature increased up to 32°C. At 21 DAT, however, the photosynthetic rate was lowest in ST37, with concurrent decrease of diffusion conductance and SPAD value. In addition, decrease of NSC concentration in stem and xylem exudation rate, and increase of ABA level in leaves and xylem exudate were observed in ST37 plants at 21 DAT. These results suggested that high soil temperature before heading especially influenced yield, grain quality and plant growth. Possible mechanisms of the effect of soil temperature are discussed.  相似文献

The rates of sap flow and xylem vessel features were studied in two-year-old nongrafted and grafted avocado (Persea americana Mill.) trees. Daily sap flow rates were measured with heat and balance stem gauges in clonal Duke 7 (D7) and Toro Canyon (TC) trees and ‘Hass’ clonal scions grafted onto clonal D7 (H/D7) and TC (H/TC) rootstocks. Vessel features as size, number and total vessel area were determined histologically in the stem of the scion and rootstock and the roots of the grafted trees. Significant differences in the sap flow rate were found among the rootstocks, where D7 had a 29% higher sap flow rate than did TC (grafted and nongrafted trees). There were no differences among xylem vessel features in the stems of any of the varieties. However in the roots, D7 had wider and fewer vessels then TC do. Also, D7 had a 19% higher total vessel area than TC. These results suggest that the differences in water consumption of ‘Hass’ on different rootstocks may be associated with differences in the efficiency of the roots to absorb water across conductive tissue which may be linked to differences in the area of xylem vessels in the root.  相似文献

A preliminary field study was conducted on root mass of young hybrid polar trees (Populus deltoides x P. nigra) using the electrical capacitance method. The objective of this study was to develop a simple calibration relationship that would enable root mass (fresh or dry) to be estimated for young hybrid poplar trees grown in mineral soils using simple, non-intrusive measurements of electrical capacitance. For first-year potted saplings (n = 33), strong linear and positive correlations were found between both root dry mass (adjusted r2 = 0.895, P<0.001) and root fresh mass (adjusted r2 = 0.866, P<0.001) and root electrical capacitance for the particular DN hybrid poplar clone studied. When augmented with data from third-year DN saplings grown in a more harsh landfill environment, a somewhat less strong curvilinear relationship was indicated with root dry mass (adjusted r2 = 0.780, P<0.001, n = 50). This rapid field method may be capable of providing adequate estimates of root mass for young trees in a variety of applications, such as afforestation, agroforestry or phytoremediation studies. Oak mortality is often associated with a complex of decline factors. We describe the morphological and physiological responses of coast live oak, Quercus agrifolia Née, in California to an invasive insect, the goldspotted oak borer (GSOB), Agrilus auroguttatus Schaeffer (Coleoptera: Buprestidae), and evaluate drought as a potential inciting factor. Morphological traits of 356 trees were assessed and physiological traits of 70 of these were monitored intensively over one growing season. Morphological characteristics of tree health included crown thinning and dieback; bole staining resulting from larval feeding; density of GSOB adult exit holes; and holes caused by woodpecker feeding. These characteristics were used to rank GSOB infestation/injury into four classes, and taken together, they explained 87% of the variation in a principal component analysis. Drought stress on various size/age and infestation classes of Q. agrifolia was measured by assessing branchlet pre-dawn and solar noon xylem water potential, leaf cell turgor potential, and water use efficiency over one growing season. Both morphological and physiological traits were highly variable in mature and old growth trees. Early summer plant water status (branchlet xylem water potential and water use efficiency) was similar between uninfested and newly colonized trees, suggesting that GSOB are not pre-selecting drought-stressed Q. agrifolia for oviposition. By late summer, leaf water and cell turgor potentials were lower in infested than in uninfested mature trees, suggesting that GSOB infestation causes drought stress in these trees. Among the tree size/age classes, infested old growth trees exhibited the greatest change in water use efficiency over the growing season, and showed greater morphological injury symptoms of decline than infested mature trees. Morphological attributes of decline in Q. agrifolia associated with GSOB were correlated weakly with increasing physiological drought stress among infestation classes of trees. We propose that the collection of morphological responses of Q. agrifolia to GSOB described here can be used to monitor the future expansion of the GSOB distribution as well as the GSOB-induced decline of Q. agrifolia in California.  相似文献

Recent findings suggest that stomatal conductance (gs) may be as closely linked to plant chemical variables as to hydraulic variables. To test this in an urban field setting, we examined seasonal gs in relation to a number of plant and environmental variables in five temperate, deciduous tree species. Stomatal conductance was generally more closely correlated with abscisic acid concentrations in xylem sap than with shoot water potential, shoot osmotic potential, pH of xylem sap or environmental variables. Seasonal gs was mostly poorly correlated with shoot water potential and osmotic potential. Among environmental variables, PPFD accounted for most variability in gs. We tested a model, developed previously in maize, that describes regulation of gs by abscisic acid concentration of xylem sap with leaf water status acting to modify stomatal sensitivity to the abscisic acid signal. This model explained somewhat more variation in gs than abscisic acid concentrations alone. Response surface models, especially those incorporating environmental variables, were most successful at explaining gs. Our findings with urban trees are consistent with the theory of regulation of gs by root-sourced abscisic acid.  相似文献

为探讨木质部栓塞修复过程监测到大量AE信号现象的原因,基于Rayleigh-Plesset方程建立描述栓塞修复过程中空化泡的运动方程,并用数值模拟方法得到空化泡运动方程的解,对AE信号进行动力学分析。结果表明:当木质部液水势升高时,空化泡会塌缩为半径较小的气泡。但经过一次塌缩并不会使栓塞修复。在压强升高的情况下,气泡会经多次塌缩直到形成稳定的微小气泡,栓塞才会完全修复。在气泡多次塌缩的过程中,气泡半径减小,但是并不是单调减小而是以减幅振动的方式减小,从而形成振源,成为AE信号的来源之一。  相似文献

以郑单958为材料, 采用田间试验方法, 在50 025株/hm2、67 500株/hm2、100 050株/hm2密度下分别设两种株行距配置, 探讨增密条件下调控株行距配置对玉米根系性状及产量影响。结果表明, 3个种植密度下总体表现为小行距种植方式在0~60 cm土层内的根重密度、根长密度及根表面积密度均高于大行距处理。50 025株/hm2下吐丝期时0~60 cm土层内的根重密度和根长密度在两个株行距配置间差异显著(P<0.05), 两个株行距配置的根表面积密度在10叶展、吐丝期和吐丝后25 d差异显著(P<0.05);67 500株/hm2下不同时期两个株行距配置根重密度差异显著(P<0.05), 根长密度和根表面积密度在10叶展和吐丝后25 d差异显著(P<0.05);100 050株/hm2下, 两个株行距配置间根重密度和根长密度在10叶展和吐丝后25 d差异显著(P<0.05), 不同时期根表面积密度差异显著(P< 0.05)。同一密度下缩行增株后单株木质部伤流液体积增大, 根系供应能力增强, 产量增加, 50 025株/hm2、67 500株/hm2下产量分别提高了6.76%和4.89%。不同时期0~60 cm土层内根重密度、根长密度和根表面积密度与产量呈正相关, 其中10叶展时各根系性状均与产量呈显著正相关。  相似文献
