This study was conducted to identify and analyse the expression of gametogenesis‐associated genes and proteins in foetal and adult buffalo gonads of both the sexes. Relative quantification of the genes was determined by qPCR and Western blotting. Immunohistochemistry was also performed for various gametogenesis‐associated proteins in foetal and adult gonads of both the sexes. We observed significantly (p < 0.05) increased expression of primordial germ cell‐specific, meiotic as well as genes associated with oocyte maturation and development in foetal ovaries as compared to the adult ones. However, significantly (p < 0.05) increased expression of proteins associated with oocyte maturation like GDF9 and ZP4 was found in adult ovaries, indicating temporal regulation of mRNA translation during oogenesis. Meiotic genes showed significantly (p < 0.05) increased expression in adult testes as compared to foetal testes and ovaries, indicating onset of meiosis at a later stage in spermatogenesis. In general, the expression of primordial germ cell‐associated as well as meiotic genes was higher in adult testes, indicating the increased biological activity in the organ. Immunohistochemistry revealed localized expression of gametogenesis‐associated proteins in ovarian follicles and seminiferous tubules of testes, while the surrounding somatic tissues were devoid of these proteins. The study gives an understanding of the sequential and temporal events of gene expression as well as mRNA translation during male and female gametogenesis. It could also be concluded that follicles and seminiferous tubules are the functional units of the female and male gonads, respectively, and their function could be enhanced by appropriate chemical and genetic intervention of the somatic tissue immediately surrounding them. This assumes importance in the context that buffalo attains sexual maturity at an older age of 2–3 years and have smaller ovaries with lesser number of primordial follicles in comparison with cattle, which is suggested to be the main reason of their poor breeding performance.  相似文献

The degradation of penoxsulam [2-(2,2-difluoroethoxy)-N-5,8-dimethoxy[1,2,4]triazolo[1,5-C]pyrimidin-2-yl-6-(trifluoromethyl)benzene-sulfonamide] was studied in flasks simulating flooded rice field conditions using four representative rice field soils from the Sacramento Valley. Degradation half-lives (t(1/2) values) ranged between 2 and 13 days. Increased degradation rates were observed in flask systems with steeper redox gradients between the flooded soil layer and the overlaying water. Two transient metabolites were identified that were temporarily formed in amounts exceeding 5% of the total initial mass of penoxsulam. The results of high-performance liquid chromatography/(14)C radiodetection studies indicate that the degradation of the triazolopyrimidine system and its substituents is the main pathway of microbial transformation processes. Microbial activity, as measured by dehydrogenase activity, was not affected by penoxsulam concentrations corresponding to the proposed maximum annual use rate of 40 g active ingredient/ha.  相似文献

The microbial degradation of etofenprox, an ether pyrethroid, was characterized under anaerobic (flooded) and aerobic (nonflooded) California rice field soil conditions by determination of its half-life (t1/2) and dissipation rate constant (k) and identification and quantification of degradation products at both 22 and 40 °C using LC-MS/MS. The overall anaerobic t1/2 at 22 °C ranged from 49.1 to 100 days (k=-0.0141 to -0.0069 days(-1)) compared to 27.0 days (k=-0.0257 days(-1)) at 40 °C, whereas under aerobic conditions the overall t1/2 was 27.5 days (k=-0.0252 days(-1)) at 22 °C compared to 10.1-26.5 days (k=-0.0686 to -0.0262 days(-1)) at 40 °C. The biphasic dissipation profiles were also fit to a first-order model to determine the t1/2 and k for both the fast and slow kinetic regions of the dissipation curves. Hydroxylation at the 4'-position of the phenoxy phenyl ring was the major metabolic process under anaerobic conditions for both 22 °C (maximum% yield of applied etofenprox mass=1.3±0.7%) and 40 °C (max % yield=1.2±0.8%). Oxidation of the ether moiety to the ester was the major metabolite under aerobic conditions at 22 °C (max% yield=0.5±0.1%), but at 40 °C increased amounts of the hydroxylated form were produced (max% yield=0.7±0.2%, compared to 0.3±0.1% for the ester). The hydrolytic product of the ester, 3-phenoxybenzoic acid (3-PBA), was not detected in any samples. Sterilized control soils showed little etofenprox degradation over the 56-day incubation period. Thus, the microbial population in a flooded soil was able to transform and contribute to the overall dissipation of etofenprox. The simulated summer temperature extreme (40 °C) increased the overall degradation.  相似文献

Penoxsulam (trade name Granite) is a new acetolactate synthase (ALS) inhibitor herbicide for postemergence control of annual grasses, sedges, and broadleaf weeds in rice culture. This study was done to understand the equilibrium phase partitioning of penoxsulam to soil and air under conditions simulating California rice field conditions. Partitioning of penoxsulam was determined between soil and water (Kd) by the batch equilibrium method and between air and water (K(H)) by the gas-purge method. In four representative soils from the Sacramento Valley, the Kd values ranged from 0.14 to 5.05 and displayed a modest increase with soil pH. In soil amended with manure compost, soil sorption increased 4-fold with increasing soil organic matter content, but was still low with a Kd of 0.4 in samples with high organic carbon contents of 15%. Penoxsulam was confirmed to be extremely nonvolatile and did not partition into air at any measurable rate at 20 or 40 degrees C. K(H) (pH 7) was estimated at 4.6 x 10(-15) Pa x L x mol(-1) on the basis of available water solubility and vapor pressure data. The results imply that soil and air partitioning of penoxsulam do not significantly affect its potential for degradation or offsite movement in water.  相似文献

Outbreaks of proliferative haemorrhagic enteropathy on two pig farms   总被引:2,自引:0,他引:2  
SUMMARY Clinical signs of proliferative haemorrhagic enteropathy (PHE) including anaemia, dysentery and sudden death were observed in finisher pigs and young breeding stock on 2 farms. On farm A, PHE occurred 12 months after repopulation of the farm. Other outbreaks of PHE occurred after the withdrawal of therapeutic concentrations of in-feed antibacterial agents (farm A), or after monensin sodium (100 g/t) replaced olaquindox (100 g/t) in feed (farm B). The outbreaks, the possible sources of contamination and the role of antibacterial feed additives in the treatment and control of PHE are described.  相似文献

A number of insecticides used for ectoparasite control in the livestock industry were screened for their efficacy against larvae of the screw-worm fly, Chrysomya bezziana, using in vivo and laboratory tests. Proprietary screw-worm fly treatments (after exposure to outdoor conditions for up to 10 days) were also tested against eggs and adults of C bezziana. Three of these were also evaluated on naturally acquired screw-worm infestations. Residual protection was generally of short duration. Among the organophosphorus compounds, the most effective formulations contained relatively high concentrations (3 to 4% al) of coumaphos, 2.5% fenchlorphos or low concentrations (0.05 to 0.5% al) of diazinon, chlorfenvinphos and fenthion methyl. Two chlorinated hydrocarbon insecticides containing 3% lindane and 5% dieldrin were very effective but are now prohibited for use in Australia. Preparations had serious deficiencies when used under field conditions, especially for treating large, deepseated myiases for which systemic insecticides are recommended. A comparison of methods demonstrated that a laboratory test could supersede live animal experimentation, at least for the initial screening of potential insecticides.  相似文献

Two Scottish Deerhound puppies had clinical and pathological features consistent with the diagnosis of congenital non-goitrous hypothyroidism. They were from separate litters, but were the progeny of the same sire and dam. The puppies were smaller, had shorter limbs and shorter, broader heads than their littermates. They also had histories of weakness, difficulty in walking and somnolence. A characteristic radiographic feature was the absence of epiphyseal growth centres. Both had depressed serum thyroxine (T4) levels and one did not respond to exogenous thyroid stimulating hormone. On necropsy, the thyroid glands were small, the follicles varied in size and contained little or no colloid. The adenohypophysis contained many cells with markedly vacuolated cytoplasm. It is suggested that the clinicopathological pattern is the result of a primary thyroid abnormality. Possible mechanisms include either primary thyroid hypoplasia or an unresponsiveness to thyroid stimulating hormone.  相似文献

A long-term field experiment was initiated in November, 1967 at Research Area of Department of Soil Science to study the response of nitrogen to pearl millet-wheat cropping system at various doses and modes of farmyard manure application. The soil organic carbon increased with farmyard manure application and ranged from 0.68% in control to 1.82% in the plot receiving the highest annual dose (90 Mg ha?1) of farmyard manure. To study the contribution of farmyard manure on the productivity of pearl millet and wheat crops, the constants (intercept and slope) were determined between the grain yield of pearl millet and wheat crops with increasing dose of fertilizer nitrogen. Another linear regression was fitted between the intercept of the linear model and the soil organic carbon content. It has been observed that with each unit increase in the soil organic carbon, the productivity of pearl millet increased by 273 kg ha?1 and that of wheat by 1591 kg ha?1. The regression between the slope and soil organic carbon was linear in case of pearl millet (R2 0.49) but in case of wheat there was no relationship.  相似文献
