In this study, we analysed the impact of heavy metals and plant rhizodeposition on the structure of indigenous microbial communities in rhizosphere and bulk soil that had been exposed to heavy metals for more than 150 years. Samples of the rhizosphere of Silene vulgaris and non-rhizosphere soils 250 and 450 m from the source of emission that had different metal concentrations were collected for analyses. The results showed that soils were collected 250 m from the smelter had a higher number of Cd-resistant CFU compared with the samples that were collected from 450 m, but no significant differences were observed in the number of total and oligotrophic CFU or the equivalent cell numbers between rhizosphere and non-rhizosphere soils that were taken 250 and 450 m from the emitter. Unweighted pair group method with arithmetic mean (UPGMA) cluster analysis of the denaturing gradient gel electrophoresis (DGGE) profiles, as well as a cluster analysis that was generated on the phospholipid fatty acid (PLFA) profiles, showed that the bacterial community structure of rhizosphere soils depended more on the plant than on the distance and metal concentrations. The sequencing of the 16S rDNA fragments that were excised from the DGGE gel revealed representatives of the phyla Bacteroidetes, Acidobacteria, Gemmatimonadetes, Actinobacteria and Betaproteobacteria in the analysed soil with a predominance of the first three groups. The obtained results demonstrated that the presence of S. vulgaris did not affect the number of CFUs, except for those of Cd-resistant bacteria. However, the presence of S. vulgaris altered the soil bacterial community structure, regardless of the sampling site, which supported the thesis that plants have a higher impact on soil microbial community than metal contamination.  相似文献

A radiation dose assessment model was applied to determine naturally occurring radionuclides 238U, 210Po and 232Th in trees like Elaeocarpus oblongus, Evodia roxburghiana (Juice of the leaves that are used to treat fever), Vaccinium neilgherrense, Viburnum hebanthum and Michelia nilagirica and shrubs like Lasianthus coffeoiaes and Hedyotis stylosa in the agroforests of the Western Ghats, India and these areas were used for different agroforestry models of land development systems. The concentration of radionuclides in the biota and corresponding soil was measured using a gamma ray spectrometer and an alpha counter. The concentration ratios of these radionuclides varied substantially between the species, and E. oblongus showed a preferential uptake of all the radionuclides and hence useful for bio-indication of radionuclides in such soils. ERICA assessment tool was employed to provide an assessment of the potential doses to biota’s growing in agro forest E. oblongus appears to be much prone to radiation absorption from the dose risk point of view. The annual effective dose (AED) due to ingestion of these radionuclides in E. roxburghiana was also estimated, and was found to be lower than the world average.  相似文献

BACKGROUND

Transgenic maize (Zea mays L.) event TC1507 (Herculex® I insect protection), expressing Cry1F δ‐endotoxin derived from Bacillus thuringiensis var. aizawai, was commercialized in 2003 in the Americas. Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) susceptibility to Cry1F was monitored annually across several regions in Argentina using diagnostic concentration bioassays. Reduced performance of TC1507 maize against S. frugiperda was reported in 2013. A resistant population was established in the laboratory and the dominance of Cry1F resistance was characterized.

RESULTS

During 2012–2015, high‐survivorship of several populations was observed in the resistance monitoring program. Reciprocal crosses of a Cry1F‐resistant population with a Cry1F‐susceptible population were evaluated to calculate effective dominance (DML) based on mortality levels observed at 100 µg/ml Cry1F. Two additional dominance levels (DLC and DEC) were calculated using lethal (LC50) or effective concentration (EC50) derived from concentration–response bioassays. Estimates indicated that Cry1F resistance in S. frugiperda in Argentina was either highly recessive (DML = 0.005) or incompletely recessive (DLC < 0.26 and DEC < 0.19).

CONCLUSION

This study is the first documented confirmation and characterization of S. frugiperda Cry1F field‐evolved resistance in Argentina. The resistance to Cry1F in S. frugiperda populations collected in Argentina, is autosomal and incompletely recessive similar to the resistance reported in Brazil. © 2017 The Authors. Shoe-string disease caused by Cucumber mosaic virus (CMV) is one of the major threats to tomato production worldwide. The alteration in some biochemical parameters in leaves of the susceptible tomato genotype (Nagina) associated with CMV infection and the effect of exogenous application of salicylic acid (SA) and benzothiadiazole (BTH) were studied in this paper. Results showed that exogenous treatment with SA and BTH not only led to plants which gave significantly more yield than diseased controls (DC), but also delayed symptom expression and reduced disease severity. Analysis of biochemical parameters indicated that exogenous application of elicitors and viral infection, significantly affected the activity of peroxidase (POD), superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX). Compared to the DC plants, minimum disease severity and maximum number of fruit were recorded after a single dose of SA + BTH. Maximum plant height was recorded after weekly application of SA and maximum fruit yield per plant was gained with single dose of SA. Moreover, the activity of POD was significantly elicited many-fold after weekly application of SA + BTH, while higher amount of SOD was recorded with single dose of SA. The activity of CAT was also significantly accelerated after weekly application of SA + BTH while increased level of APX was noticed with single dose of BTH. In conclusion, the combined application of SA and BTH played an important role in induction of defense mechanism against CMV infection and can be useful in tomato disease management programs.  相似文献

Xanthomonas fragariae is the causative agent of angular leaf spot of strawberry, a quarantine organism in plant propagation material in the European Union. Field experiments were conducted to assess the risks for infection of strawberry plants through dispersal of an aerosolized inoculum. In practice, pathogen aerosols can be formed during mowing of an infected crop or by water splashing on symptomatic plants during overhead irrigation or rain. In our experiments, aerosols were generated by spraying suspensions of X. fragariae with a density of 108 cfu ml?1 or water under pressure vertically up into the air. In strawberry plants (cv Elsanta) placed at 1.3, 5 and 10 m distance downwind from the spray boom, infections were found, as evidenced with a combination of dilution–plating and molecular techniques, but more frequently in plants wetted prior to inoculation than in plants kept dry. A logarithmic decrease in infection incidence was found with the distance to the inoculum source. Symptomatic plants were found up to 5 m distance from the inoculum source. No infected plants were found in plants placed 4 m upwind or treated with water. In glasshouse studies, it was shown that under conditions favorable for disease development, spray-inoculation of strawberry plants with estimated densities of X. fragariae as low as 2000 cfu per plant were able to cause symptoms both in cv Elsanta and cv Sonata. Results indicate that there is a considerable risk on infections of strawberry plants exposed to aerosolized inoculum.  相似文献
