Carp (Cyprinus carpio L.) is a pest species in Australian waterways, and cyprinid herpesvirus 3 (CyHV‐3) is being considered as a potential biological control (biocontrol) agent. An important consideration for any such agent is its target specificity. In this study, the susceptibility to CyHV‐3 of a range of non‐target species (NTS) was tested. The NTS were as follows: 13 native Australian, and one introduced, fish species; a lamprey species; a crustacean; two native amphibian species (tadpole and mature stages); two native reptilian species; chickens; and laboratory mice. Animals were exposed to 100–1000 times the approximate minimum amount of CyHV‐3 required to cause disease in carp by intraperitoneal and/or bath challenge, and then examined clinically each day over the course of 28 days post‐challenge. There were no clinical signs, mortalities or histological evidence consistent with a viral infection in a wide taxonomic range of NTS. Furthermore, there was no molecular evidence of infection with CyHV‐3, and, in particular, all RT‐PCRs for viral mRNA were negative. As a consequence, the results encourage further investigation of CyHV‐3 as a potential biocontrol agent that is specific for carp.  相似文献

BACKGROUND

Ethanedinitrile (EDN) is a fumigant being commercialized worldwide as an alternative phytosanitary treatment to methyl bromide (MB) for forest products. The penetration characteristics of MB and EDN were measured through the bark of wooden blocks (100 × 100 × 50 mm) cut from the upper (average bark thickness 5 ± 2 mm) and lower (average bark thickness 25 ± 5 mm) trunk of recently felled pine (Pinus radiata D.Don) trees. Doses of 48 g m−3 MB and 50 g m−3 EDN were applied to chambers at 10 and 20°C for 10 h.

RESULTS

Penetration of MB was influenced by the interaction between fumigation time and temperature, with concentrations increasing at a higher rate at 20°C compared with at 10°C. After 10 h, an average concentration of 8.05 ± 0.89 g m−3 had penetrated the bark of log sections at 20°C, whereas 5.20 ± 0.89 g m−3 was measured at 10°C. By contrast, the factors examined in this study did not significantly impact the penetration of EDN. Concentration × time (CT) values for MB under the bark were 35.20 ± 2.30 g h m−3 at 10°C and 55.85 ± 9.58 g h m−3 at 20°C; whereas for EDN, CT values were 19.50 ± 6.80 g h m−3 at 10°C and 19.08 ± 4.10 g h m−3 at 20°C.

CONCLUSION

MB can achieve a higher concentration under the bark of log sections during simulated fumigations, but all of the factors examined affected the ability of MB to penetrate the bark of wooden blocks. By comparison, the penetration of EDN through the bark is more consistent than MB under laboratory conditions. © 2022 The Authors. Detailed studies have been undertaken to define pruning regimes for Eucalyptus nitens, but little is known of E. globulus responses to pruning although this is a more commonly planted species. This paper describes experiments that aimed to identify (a) levels of pruning that reduce E. globulus growth, (b) physiological processes contributing to those growth responses, (c) the incidence of decay and factors influencing decay incidence following pruning of E. globulus, and (d) comparative responses of E. nitens and E. globulus to live branch pruning. Results of a field experiment indicated that removal of between 30 and 50% of the crown length was appropriate for E. globulus plantations verging on canopy closure. The significant reduction in height growth associated with removal of 50 or 70% of crown length suggested pruning should remain below 50% of crown length if reduced stem growth of pruned trees was to be avoided. Stem volume was only significantly reduced over the period of the experiment by 70% pruning, but it was estimated that standing volume following removal of 50% of crown length would be reduced by 82 m3 ha−1 over a 20-year rotation if there were no other silvicultural interventions. The growth responses observed were probably related to large reductions in leaf area following 50 or 70% removal of crown length. Trees responded to pruning by changing patterns of biomass partitioning to favour stem growth at the expense of branch growth. A glasshouse study determined that light-saturated net CO2 uptake (Amax) increased following pruning. E. nitens seedlings had both a higher baseline Amax and higher Amax following pruning than did E. globulus, which could partially explain the greater effect of pruning on E. globulus growth than has been observed for E. nitens in other studies. This result, as well as apparently different patterns of foliage distribution through the crowns of E. globulus and E. nitens, suggested that models of pruning responses need to be parameterised for both species. In addition, a more conservative pruning regime may be appropriate for E. globulus than E. nitens. Pruning increased the frequency of branch traces with decay infection, and there was a trend towards increasing decay outbreaks with increasing pruning severity. Decay outbreaks were more likely to occur following pruning of high angle or larger diameter branches.  相似文献

  1. Measuring the energy stores, or body condition, of cetaceans is vital for monitoring population health. Cetaceans are exposed to a range of anthropogenic stressors, including, for example, noise, contaminants, habitat degradation, reduced prey availability, ship strike, and entanglement. Tools to assess body condition in these inaccessible and vulnerable animals are required to better understand the energetic consequences of anthropogenic stressors that can impact population health and, ultimately, conservation status.
  2. Remotely obtained, dart biopsy samples are becoming an increasingly standard method of tissue collection from live, large cetaceans. Here, the potential applicability of using total lipid content in such biopsy samples to estimate overall body condition was assessed using full‐depth dorsal blubber samples from stranded ziphiids (n = 8) and balaenopterids (n = 9). First, variation in total lipid content through the blubber depth was investigated to assess the representativeness of shallow‐depth dart biopsies taken from live animals. Second, how lipid content varies by species, cause of death, sex, age class, and morphometric indices of body condition was assessed to evaluate what information about individual energy stores can be gained from such analyses.
  3. Total lipid content in dorsal, shallow‐depth blubber biopsy samples from both cetacean families provides little information on overall body condition. Stratification of lipid content through the blubber layer in the balaenopterids means that superficial biopsy samples are not representative of the lipid stores available for mobilization through the rest of the tissue. A lack of variation in blubber lipid content, both within and between the ziphiid individuals, resulted in no ability to correlate these measures with morphometrics or other health, sex, or age class covariates.
  4. Other potential markers in the blubber from remote biopsy sampling should be explored in order to further develop robust tools for estimating the body condition of free‐ranging cetaceans.

Soybean yield components and agronomic traits are connected through physiological pathways that impose tradeoffs through genetic and environmental constraints. Our primary aim is to assess the interdependence of soybean traits by using unsupervised machine learning techniques to divide phenotypic associations into environmental and genetic associations. This study was performed on large scale, jointly analyzing 14 quantitative traits in a large multi-parental population designed for genetic studies. We collected phenotypes from 2012 to 2015 from a soybean nested association panel with 40 families of approximately 140 individuals each. Pearson and Spearman correlations measured phenotypic associations. A multivariate mixed linear model provided genotypic and environmental correlations. To evaluate relationships among traits, the study used principal component and undirected graphical models from phenotypic, genotypic, and environmental correlation matrices. Results indicate that high phenotypic correlation occurs when traits display both genetic and environmental correlations. In genetic terms, length of reproductive period, node number, and canopy coverage play important roles in determining yield potential. Optimal grain yield production occurs when the growing environment favors faster canopy closure and extended reproductive length. Environmental associations found among yield components give insight into the nature of yield component compensation. The use of unsupervised learning methods provides a good framework for investigating interactions among various quantitative traits and defining target traits for breeding.  相似文献

Twenty cowpea (Vigna unguiculata L. Walp.) genotypes, comprising Kenyan cultivars and Kenyan and introduced breedings lines, were grown in 15 replicated field trials carried out at three locations in semi-arid eastern Kenya over four seasons. Grain and straw yields and the carbon isotope discrimination (Δ) of this material were determined, and days to flowering and maturity observed.None of the seasons in which the trials were conducted was appreciably drier than the long term average for the sites, and some trials received excessive rain. Averaging the data for the 15 trials, one genotype yielded much less grain (77 g m−2) than the 19 others, whose yield ranged from 125 to 177 g m−2 (average 148 g m−2). Trial mean yield trials had a high straw Δ (r = +0.567) and had received more rain between flowering and maturity (r = +0.428) than lower yielding ones. Among genotypes, averaging over trials, the correlation coefficient (r) between grain yield and grain Δ was +0.394, and between grain yield and straw Δ, +0.460. Early genotypes had the highest grain Δ and straw Δ. The correlation among genotypes between date of flowering and grain Δ was −0.632 and with straw Δ, −0.502. When comparisons were made among trials, there was no clear relationship between the strength of the correlations among genotypes between grain yield and straw Δ or grain Δ and the degree of stress experienced by a trial (as indicated by the rainfall it had received). It appeared that this was because the variances of grain Δ and straw Δ were greater in the most droughted trials, whereas the variance of yield was greatest in the least droughted trials.It is concluded that the genetic correlation between grain yield and either grain Δ or straw Δ is unlikely to be strong enough to make either Δ a useful surrogate or adjunct in selecting for high yield in the segregating generations of a breeding programme for semi-arid eastern Kenya, but that Δ may be of value in the selection of parent genotypes.  相似文献

Summary Thornless Loganberry (TL) is a periclinal chimeral blackberry in which a layer of mutant (thornless) epidermis surrounds a core of wild-type (thorny) tiusse. Due to its chimeral arrangement, TL produces thorny adventitious root cuttings and thorny offspring. To separate the chimera into its components parts, meristems of TL were grow in vitro on modified Murashige and Skoog medium to yield callus and adventitious shoots. One of these shoots has survived, flowered, and produced thornless offspring from seed.
