This study aimed to evaluate the intake, digestion, blood parameters, and feeding behavior of crossbred dairy cows (Holstein?×?Gir) managed on Panicum maximum Jacq. cv. Tanzania-1 and provided supplementation with groundnut cake, sunflower cake, or palm kernel cake (to replace soybean meal). Sixteen cows were randomly assigned in a Latin square design with four treatments and four experimental periods. The consumption of nutrients from the pasture did not vary between experimental treatments. Cows receiving the palm kernel cake supplement had a reduced crude protein, non-fibrous carbohydrate, and total digestible nutrient intake and an increase in the average consumption of ether extract. There was also a reduction in the digestibility of dry matter. The inclusion of supplements in the diet did not influence the average time spent eating, ruminating, or resting. The mean values of respiratory and heart rates showed thermal comfort during the trial period. There was a reduction in blood urea nitrogen with palm kernel cake supplementation, and the values of total protein, albumin, and glucose were also significantly different with this supplement. It is recommended that cakes of groundnut cake and sunflower cake seed be used for a total replacement of soybean meal supplements for lactating cows, but the use of palm kernel cake is not recommended.
Myostatin (MSTN) is an interesting negative growth‐regulating gene that has been well characterized in vertebrates but scantly described in invertebrates. The current study focuses on the downregulation of the MrMSTN gene and subsequently records any histological changes for giant freshwater prawn, Macrobrachium rosenbergii (Mr). In addition, the study also deals with the MrMSTN gene's influence on other growth‐related genes, which include myosin heavy chain, dystrophin‐dystroglycoprotein complex, tropomyosin, farnesoic acid o‐methyl transferase, arginine kinase, cyclophilin, and acyl CoA desaturase. The preliminary histological analysis following MrMSTN silencing favors muscle regeneration, which supports its functional role as a negative growth regulator and its significant effect on the expression of other growth‐related genes. Overall, our results show that the MrMSTN gene could therefore be a potential target for gene manipulation aimed at enhancing the growth and muscle development of M. rosenbergii, which could be beneficial in increasing the total mass production in the postlarva phase at the hatchery level. 相似文献
Important marine mammal areas (IMMAs) are discrete portions of habitat, important to marine mammal species, that have the potential to be delineated and managed for conservation. Although IMMAs are not a blueprint for marine protected areas or other conservation designations, they are useful for providing a foundation for marine spatial planning and systematic conservation planning that can then lead to protected areas or special spatial regulations. To be most useful for supporting management and conservation, however, the information coming out of IMMAs needs to reflect current conditions.
An ‘early warning system’ is proposed with a generic set of indicators to flag when marine mammal species in IMMAs require management interventions due to changing distributions or decreasing populations. Rather than signifying that quantitative thresholds have been reached, these indicators comprise alerting information derived from visual or acoustic census, satellite imagery analysis, whale‐watching logs, or increases in mortality reported by stranding networks that can trigger additional targeted research.
Although it is possible that in some regions data will be sufficient to provide quantifiable indicators, the system is meant to rely on existing data sources, and be adaptable to the circumstances of each region.
Regional expert groups can utilize early warning system information and feed it into IMMA‐related spatial planning in two ways: by nominating additional areas of interest, and by providing a scientific rationale for revising IMMA boundaries, to be considered at the next decadal IMMA regional expert workshop.
IMMA‐driven consolidation of information that is as current as possible will prove valuable for enhancing regional cooperation to conserve marine mammals, and will be useful as countries implement new protected areas to conserve marine mammals and other marine biodiversity.
Dried gill plates from manta and devil rays, some of the world's most biologically vulnerable fishes, have become a valued commodity in Asian dried‐seafood and traditional Chinese medicine markets. This trade is a primary driver of fisheries, which have led to declines in many mobulid populations.
With no reliable trade statistics and scarce data on mobulid fisheries, this study estimates the number and species of mobulids required to supply this trade, and investigates the consumers and suppliers involved and drivers of demand. Following preliminary market research, 525 trader surveys were conducted in Hong Kong, Singapore, Macau, Taiwan, and southern China.
Guangzhou, China was identified as the centre of the trade accounting for 99% of total estimated market volume of 60.5 tons of dried gill plates in 2011, increasing to 120.5 tons by 2013. The estimated number of mobulids converted from tons of gill plates more than doubled over the period to 130 000, comprising 96% devil rays, Mobula japanica, Mobula thurstoni, and Mobula tarapacana, and 4% Manta spp. By 2015 the Guangzhou market had declined sharply, reportedly due to conservation campaigns and government policies. However Hong Kong's gill plate sales increased dramatically between 2011 and 2015.
China, Indonesia, Vietnam, Sri Lanka, and India were reported most frequently as gill plate sources.
Vendors recommend gill plates (trade name pengyusai) for ailments ranging from acne to cancer and as a general health tonic. While pengyusai is a new addition to traditional Chinese medicine literature and is rarely prescribed by traditional medicine practitioners, it is readily available over the counter and aggressively marketed by vendors.
Working in concert with consumer demand reduction efforts, increased measures to restrict mobulid fisheries and trade are recommended to prevent further population declines of these highly vulnerable species.
The rice Pi2/9 locus harbors multiple resistance (R) genes each controlling broad-spectrum resistance against diverse isolates of Magnaporthe oryzae, a fungal pathogen causing devastating blast disease to rice. Identification of more resistance germplasm containing novel R genes at or tightly linked to the Pi2/9 locus would promote breeding of resistance rice cultivars.
Results
In this study, we aim to identify resistant germplasm containing novel R genes at or tightly linked to the Pi2/9 locus using a molecular marker, designated as Pi2/9-RH (Pi2/9 resistant haplotype), developed from the 5′ portion of the Pi2 sequence which was conserved only in the rice lines containing functional Pi2/9 alleles. DNA analysis using Pi2/9-RH identified 24 positive lines in 55 shortlisted landraces which showed resistance to 4 rice blast isolates. Analysis of partial sequences of the full-length cDNAs of Pi2/9 homologues resulted in the clustering of these 24 lines into 5 haplotypes each containing different Pi2/9 homologues which were designated as Pi2/9-A5, ?A15, ?A42, ?A53, and -A54. Interestingly, Pi2/9-A5 and Pi2/9-A54 are identical to Piz-t and Pi2, respectively. To validate the association of other three novel Pi2/9 homologues with the blast resistance, monogenic lines at BC3F3 generation were generated by marker assisted backcrossing (MABC). Resistance assessment of the derived monogenic lines in both the greenhouse and the field hotspot indicated that they all controlled broad-spectrum resistance against rice blast. Moreover, genetic analysis revealed that the blast resistance of these three monogenic lines was co-segregated with Pi2/9-RH, suggesting that the Pi2/9 locus or tightly linked loci could be responsible for the resistance.
Conclusion
The newly developed marker Pi2/9-RH could be used as a potentially diagnostic marker for the quick identification of resistant donors containing functional Pi2/9 alleles or unknown linked R genes. The three new monogenic lines containing the Pi2/9 introgression segment could be used as valuable materials for disease assessment and resistance donors in breeding program.
Protoplast fusion can be used to produce somatic hybrids of species that cannot be obtained by sexual hybridization. The possibility to introgress genes from Solanum species into the cultivated tomato species Lycopersicon esculentum, and to obtain novel cytoplasm-nucleus combinations (cybrids) was considered as an important strategy to extend the genetic variation available for tomato breeding. Somatic hybrids between L. esculentum and other Lycopersicon species, as well as between L. esculentum and Solanum or Nicotiana species, have been produced. Specific mutants, genotypes with antibiotic resistances, and metabolic inhibition by iodoacetate or iodoacetamide and irradiation were used for the selection of hybrids. In addition, the improvement of protoplast culture techniques and the use of the favourable tissue culture traits derived from species such as L. peruvianum, which have been introduced into tomato by classical breeding, allowed the efficient recovery of somatic hybrids. However, the occurrence of somatic incongruity in fusion combinations of L. esculentum and Solanum and even more in L. esculentum and Nicotiana, did not allow the production of true cybrids and/or fertile hybrids, indicating the importance of both cytoplasm-nucleus and nucleus-nucleus interactions in somatic incongruity. Another problem with fusions between distantly related species is the strongly reduced fertility of the hybrids and the very limited homoeologous recombination between chromosomes of the parental species. Partial genome transfer from donor to recipient through microprotoplast (+) protoplast fusion, and the production of monosomic or disomic chromosome addition lines, light overcome some of these problems. In symmetric somatic hybrids between L. esculentum and S. tuberosum the occurrence of limited somatic and meiotic recombination was demonstrated. Fertile progeny plants could be obtained, though at a low frequency, when embryo rescue was performed on a large scale after backcrossing hexaploid somatic tomato (+) potato hybrids with a tetraploid potato genotype. The potential value of genomic in situ hybridization (GISH) and RFLPs for the analysis of the genome/chromosome composition of the hybrids has been demonstrated for intergeneric somatic hybrids between Lycopersicon and Solanum.Abbreviations cpDNA
