Like other influenza A viruses, equine influenza virus undergoes antigenic drift. It is therefore essential that surveillance is carried out to ensure that recommended strains for inclusion in vaccines are kept up to date. Here we report antigenic and genetic characterisation carried out on equine influenza virus strains isolated in North America and Europe over a 2-year period from 2008 to 2009. Nasopharyngeal swabs were taken from equines showing acute clinical signs and submitted to diagnostic laboratories for testing and virus isolation in eggs. The sequence of the HA1 portion of the viral haemagglutinin was determined for each strain. Where possible, sequence was determined directly from swab material as well as from virus isolated in eggs. In Europe, 20 viruses were isolated from 15 sporadic outbreaks and 5 viruses were isolated from North America. All of the European and North American viruses were characterised as members of the Florida sublineage, with similarity to A/eq/Lincolnshire/1/07 (clade 1) or A/eq/Richmond/1/07 (clade 2). Antigenic characterisation by haemagglutination inhibition assay indicated that the two clades could be readily distinguished and there were also at least seven amino acid differences between them. The selection of vaccine strains for 2010 by the expert surveillance panel have taken these differences into account and it is now recommended that representatives of both Florida clade 1 and clade 2 are included in vaccines. 相似文献

The molecular analysis of the arbuscular mycorrhizal (AM) symbiosis started at the beginning of this decade. The paper summarises the work of the ‘Laboratorium für Molekulare Genetik arbuskulärer Mykorrhiza’ at the MPI für terrestrische Mikrobiologie in Marburg on three major topics. Firstly, the plant response to the mycorrhizal colonisation was analysed using both, targeted and non‐targeted approaches. As an example, the localisation of the gst1 mRNA in potato mycorrhiza is shown. Second, molecular techniques were established to analyse gene expression of the fungal partner of the symbiosis. We present a differential RNA display analysis of spore germination in two AM fungi and the cloning of a gene from Glomus mosseae that shows expression at all stages of the fungal life cycle. In the last part, we introduce the work we are carrying out with a new root endophytic fungus, Piriformospora indica. Infection experiments on maize showed that despite the fungus performs as a root necrotroph, it has a positive effect on plant growth. 相似文献

This study was conducted to improve our understanding of how earthworms and microorganisms interact in the decomposition of litter of low quality (high C : N ratio) grown under elevated atmospheric [CO2]. A microcosm approach was used to investigate the influence of endogeic earthworm (Aporrectodea caliginosa Savigny) activity on the decomposition of senescent Charlock mustard (Sinapis arvensis L.) litter produced under ambient and elevated [CO2]. Earthworms and microorganisms were exposed to litter which had changed in quality (C : N ratio) while growing under elevated [CO2]. After 50 d of incubation in microcosms, C mineralization (CO2 production) in the treatment with elevated‐[CO2] litter was significantly lower in comparison to the ambient‐[CO2] litter treatment. The input of Charlock mustard litter into the soil generally induced N immobilization and reduced N2O‐emission rates from soil. Earthworm activity enhanced CO2 production, but there was no relationship to litter quality. Although earthworm biomass was not affected by the lower quality of the elevated‐[CO2] litter, soil microbial biomass (Cmic, Nmic) was significantly decreased. Earthworms reduced Cmic and fungal biomass, the latter only in treatments without litter. Our study clearly showed that A. caliginosa used the litter grown under different [CO2] independent of its quality and that their effect on the litter‐decomposition process was also independent of litter quality. Soil microorganisms were shown to negatively react to small changes in Charlock mustard litter quality; therefore we expect that microbially mediated C and N cycling may change under future atmospheric [CO2]. 相似文献

Breeding for organic agriculture (BFOA) is a strategy for a commercial breeding company based on the exploitation of the frequently
observed high correlation for many traits between conventional, low input (LI) and organic agriculture (ORG). Indirect selection
under LI can be useful to roughly divide the germplasm into the genotypes better adapted to high input and those better adapted
to LI or ORG conditions. BFOA is an evolving process, in which two methods are currently applied: early generations are either
selected following the pedigree system under LI, or grown as bulk populations on ORG fields. In the latter case the system
switches to LI after individual ear selection under ORG conditions. In both methods, the first replicated yield trial is grown
parallel under ORG and LI. Subsequently, the genotypes are grouped into conventional or ORG advanced trial series. The BFOA
strategy allows that the larger genetic variability of both the organic and conventional gene pool can be exploited in the
selection for ORG. Hitherto, seven winter wheat varieties were released in Austria after exclusive organic VCU testing. 相似文献

Einsatz bakterieller Antagonisten zur Bekämpfung von Krankheiten verursacht durch Rhizoctonia solani
Rhizoctonia solani is an important soil-borne pathogen causing diseases in numerous economically important crops. The pathogen R. solani can be responsible for relevant yield losses as well as on lettuce and potato. To develop a biocontrol strategy, two bacterial strains, Pseudomonas fluorescens L13–6-12 and Serratia plymuthica 3Re4–18 were evaluated against R. solani causing black scurf in potato and bottom rot in lettuce. The disease suppression effect of the two antagonists was tested as well as in a climate chamber and in the field during two vegetation periods. The results of the climate chamber experiments showed, that R. solani can significantly reduced the lettuce growth. The dry mass losses on lettuce and the disease severity on potato sprouts were significantly limited through bacterization. The antagonist L13–6-12 showed best disease suppression effect in climate chamber experiments on both crops. A significant lower disease severity was to observed in treatments with bacterial antagonists as well as on lettuce plants and harvested potato tubers during both vegetation periods. Also the dry mass losses on lettuce were clear reduced in treatments with bacterial antagonists, whereas only a partly limitation of yield losses on potato was to achieved. In summary, the results supported that the use of bacterial antagonists can be part of a control strategy against R. solani. 相似文献

ABSTRACT: Understanding protein and gene function requires identifying interaction partners usingbiochemical, molecular or genetic tools. In plants, searching for novel protein-proteininteractions is limited to protein purification assays, heterologous in vivo systems such as the yeast-two-hybrid or mutant screens. Ideally one would be able to search for novel proteinpartners in living plant cells. We demonstrate that it is possible to screen for novel proteinproteininteractions from a random library in protoplasted Arabidopsis plant cells and recoversome of the interacting partners. Our screen is based on capturing the bi-molecularcomplementation of mYFP between an YN-bait fusion partner and a completely random preyYC-cDNA library with FACS. The candidate interactions were confirmed using in plantaBiFC assays and in planta FRET-FLIM assays. From this work, we show that the wellcharacterized protein Calcium Dependent Protein Kinase 3 (CPK3) interacts with APX3,HMGB5, ORP2A and a ricin B-related lectin domain containing protein At2g39050. Objective To describe the clinical data of dogs with neurogenic Keratoconjunctivitis sicca (KCS) and an ipsilateral dry nose without other neurologic deficits. Procedure The retrospective case study included 11 dogs diagnosed with neurogenic KCS and an ipsilateral dry nose between 2006 and 2010. Medical records were reviewed for breed, age, sex, history, suspected cause of neurogenic KCS, clinical signs, and treatment modalities. Follow‐up information was obtained by re‐examination of patients or completion of a telephone survey with the referring veterinarian or the owners. Results Mean age of the dogs was 6.6 ± 4.5 years. Neurogenic KCS was diagnosed in three females, five spayed females, one male, and two castrated males representing 10 different breeds. Ophthalmic signs of KCS (mean Schirmer tear test [STT] value of 1.9 ± 2.9 mm/min) combined with an ipsilateral dry nose were diagnosed in seven left and four right eyes. The suspected cause of neurogenic KCS was idiopathic in nine and trauma in two cases. Systemic therapy consisted of oral pilocarpine 1–2% eye drops combined with case‐specific topical treatment with cyclosporine 0.2% and tear substitutes. Duration of systemic treatment with pilocarpine until healing was 125 days (range 84–204, median 98 days) for five dogs. One dog was lost to follow‐up, and the remaining five dogs are still under systemic treatment with pilocarpine. Conclusions Neurogenic KCS with an ipsilateral dry nose seems to be a predominantly idiopathic disease of middle‐aged female dogs without breed predisposition, which may be self‐limiting in some cases. 相似文献
