Discovery of epigenetic modifications associated with feed efficiency or other economically important traits would increase our understanding of the molecular mechanisms underlying these traits. In combination with known genetic markers, this would provide opportunity to improve genomic selection accuracy in cattle breeding programs. It would also allow cattle to be managed to improve favorable gene expression. The objective of this study was to identify variation in DNA methylation between beef cattle of differential pre-natal nutrition and divergent genetic potential for residual feed intake (RFI). Purebred Angus offspring with the genetic potential for either high (HRFI) or low (LRFI) RFI were prenatally exposed to either a restricted maternal diet of 0.5 kg/d average daily gain (ADG) or a moderate maternal diet of 0.7 kg/d ADG from 30 to 150 d of gestation. We performed DNA methylation analysis of differentially methylated regions (DMR) of imprinted genes (Insulin-like growth factor 2 (IGF2) DMR2, IGF2/H19 imprinting control region (ICR) and IGF2 receptor (IGF2R) DMR2) using post-natal samples of longissimus dorsi (LD) muscle taken from male and female calves at birth and weaning, and of LD muscle, semimembranosus (SM) muscle, and liver samples collected from steers at slaughter (17 months of age). Interestingly, for all three DMR investigated in liver, LRFI steers had higher levels of methylation than HRFI steers. In LD muscle, IGF2/H19 ICR methylation differences for heifers at birth were due to pre-natal diet, while for steers at birth they were mostly the result of genetic potential for RFI with LRFI steers again having higher levels of methylation than HRFI steers. While results from repeated measures analysis of DNA methylation in steers grouped by RFI revealed few differences, in steers grouped by diet, we found higher methylation levels of IGF2 DMR2 and IGF2R DMR2 in LD muscle of restricted diet steers at weaning and slaughter than at birth, as well as increased methylation in LD muscle of restricted diet steers compared with moderate diet steers at weaning and/or slaughter. Our results suggest that differential pre-natal nutrition, and divergent genetic potential for RFI, induces tissue- and sex-specific alterations in post-natal IGF2 and IGF2R methylation patterns and that these patterns can vary with age in Angus beef cattle.  相似文献

ObjectivesTwo-dimensional (2D) speckle tracking echocardiography (STE) is a new angle-independent ultrasound technique based on tracking of speckles within the myocardium on 2D grayscale images. The aims of this prospective study were as follows: (1) to assess the variability of left ventricular peak systolic radial strain (St) and strain rate (SR) in awake dogs using STE (Protocol 1); and (2) to quantify these variables in a healthy canine population and compare them with tissue Doppler imaging (TDI)-based St and SR values (Protocol 2).BackgroundSt and SR may be assessed using TDI, which is limited by angle dependency.Animals, materials and methodsThirty-six STE examinations were performed on 6 healthy dogs for Protocol 1 and 37 healthy dogs were recruited for Protocol 2. In both studies, STE measurements were obtained offline from the right parasternal short-axis view by the same trained observer using automatic frame-to-frame tracking of grayscale speckle patterns.ResultsAll within- and between-day coefficients of variation were <10% (Protocol 1). In Protocol 2, St (46.7 ± 12.2%) and SR (2.7 ± 0.6 s−1) measured by STE were correlated with heart rate (p < 0.01), but not with the ratio of early mitral inflow velocity to early mitral annular velocity. There was a good correlation between STE and TDI for both St and SR values (p < 0.001).ConclusionsSTE is a repeatable and reproducible non-Doppler method for assessing radial St and SR. The combination of these indices with conventional echo-Doppler variables could provide a new approach for accurately quantifying canine systolic function.  相似文献

根据番茄的形态结构特点及其演变规则,应用有限态自动机的数学理论,研究建立了番茄动态发展的随机结构模型。该模型建立在对番茄结构的宏观抽象和生理年龄(PHYAGE)与生长年龄(GA)这两个最重要的基本概念之上,利用“事件驱动”的状态转移机制模拟了番茄生长过程、分枝过程和死亡过程,其中利用了二项随机分布模拟番茄的动态生长,离散的Markov链用于模拟番茄的分枝模式。番茄的有限态自动机模型完成了对主茎上节间长度、果实个数以及叶子结构的随机模拟。并利用试验统计分析获得的参数结果,模拟出番茄在不同生长阶段的3D结构。该动态的番茄结构模型,为与生理生态模型进行接口建立互影响、互反馈的结构-功能模型打下了基础。  相似文献

建立了番茄动态生长的功能模型,从个体和器官的角度,研究生物量在植物体内的生产和分配规律、器官的动态生长规则。模型根据蒸腾作用计算物质的生产;根据节间、叶柄、叶片和果实等器官的汇强和扩展规律对物质进行分配;根据与物质量的动态关系计算各器官的几何尺寸;利用试验数据对模型参数进行估计。通过对参数进行比较和分析得出它们对各器官生长的影响程度和器官本身的扩展规律。并对模型输出与测量数据的拟合程度进行了残差分析,验证了模型的合理性和有效性。  相似文献

About 50% of cancer patients receive radiation therapy. Here we investigated the hypothesis that tumor response to radiation is determined not only by tumor cell phenotype but also by microvascular sensitivity. MCA/129 fibrosarcomas and B16F1 melanomas grown in apoptosis-resistant acid sphingomyelinase (asmase)-deficient or Bax-deficient mice displayed markedly reduced baseline microvascular endothelial apoptosis and grew 200 to 400% faster than tumors on wild-type microvasculature. Thus, endothelial apoptosis is a homeostatic factor regulating angiogenesis-dependent tumor growth. Moreover, these tumors exhibited reduced endothelial apoptosis upon irradiation and, unlike tumors in wild-type mice, they were resistant to single-dose radiation up to 20 grays (Gy). These studies indicate that microvascular damage regulates tumor cell response to radiation at the clinically relevant dose range.  相似文献

Rooibos and honeybush teas significantly (P < 0.05) enhanced the activity of cytosolic glutathione S-transferase alpha. A significant (P < 0.05) to marginal (P < 0.1) increase in the activity of the microsomal UDP-glucuronosyl transferase was obtained with unprocessed rooibos and honeybush teas, respectively. Oxidized glutathione (GSSG) levels were significantly (P < 0.05) reduced in the liver of all tea treated rats while reduced glutathione (GSH) was markedly increased in the liver of the herbal tea treated rats. These changes resulted in a significant (P < 0.05) increase in the GSH/GSSG ratio by the unprocessed, processed rooibos and unprocessed honeybush teas. Green and black teas markedly to significantly decreased the oxygen radical absorbance capacity in liver homogenates, respectively. Modulation of phase II drug metabolizing enzymes and oxidative status in the liver may be important events in the protection against adverse effects related to mutagenesis and oxidative damage.  相似文献

Runoff events were analysed in separated sewer systems in the town of Luxembourg. The relationships between Event Mean Concentrations of different pollutants and runoff patterns were evaluated. In addition, the inter-storm and intra-storm variability of the material transport were determined. Primarily, the variations in pollutant concentrations and loads are determined by the antecedent weather conditions. The presence of illicit sanitary inputs in one of the sewers produced a significant first flush effect as well as higher Event Mean Concentrations for pollutants. Furthermore, near the town of Trier 40 storms were analyzed in a small natural basin mainly influenced by runoff from a separated sewer system. Natural and artificial storm events were investigated in order to estimate the relationship between the pollutant sources in the channel and from the separated sewer system. Just like in the canalization of Luxembourg City the pollutant dynamics during natural storms are strongly influenced by pre-event hydrological conditions. The artificial storms behave differently. Despite little pre-rain, the maximum concentrations of toxic substances are comparatively low. A resuspension of sediment only occurs in the natural channel system, without the introduction of fines from the sewer system.  相似文献
