Samples from multiple animals may be pooled and tested to reduce costs of surveillance for infectious agents in aquatic animal populations. The primary advantage of pooling is increased population‐level coverage when prevalence is low (<10%) and the number of tests is fixed, because of increased likelihood of including target analyte from at least one infected animal in a tested pool. Important questions and a priori design considerations need to be addressed. Unfortunately, pooling recommendations in disease‐specific chapters of the 2018 OIE Aquatic Manual are incomplete and, except for amphibian chytrid fungus, are not supported by peer‐reviewed research. A systematic review identified only 12 peer‐reviewed aquatic diagnostic accuracy and surveillance studies using pooled samples. No clear patterns for pooling methods and characteristics were evident across reviewed studies, although most authors agreed there is a negative effect on detection. Therefore, our purpose was to review pooling procedures used in published aquatic infectious disease research, present evidence‐based guidelines, and provide simulated data examples for white spot syndrome virus in shrimp. A decision tree of pooling guidelines was developed for use by peer‐reviewed journals and research institutions for the design, statistical analysis and reporting of comparative accuracy studies of individual and pooled tests for surveillance purposes.  相似文献

Calibration of the Granier thermal dissipation technique for measuring stem sap flow in trees requires determination of the temperature difference (DeltaT) between a heated and an unheated probe when sap flow is zero (DeltaT(max)). Classically, DeltaT(max) has been estimated from the maximum predawn DeltaT, assuming that sap flow is negligible at nighttime. However, because sap flow may continue during the night, the maximum predawn DeltaT value may underestimate the true DeltaT(max). No alternative method has yet been proposed to estimate DeltaT(max) when sap flow is non-zero at night. A sensitivity analysis is presented showing that errors in DeltaT(max) may amplify through sap flux density computations in Granier's approach, such that small amounts of undetected nighttime sap flow may lead to large diurnal sap flux density errors, hence the need for a correct estimate of DeltaT(max). By rearranging Granier's original formula, an optimization method to compute DeltaT(max) from simultaneous measurements of diurnal DeltaT and micrometeorological variables, without assuming that sap flow is negligible at night, is presented. Some illustrative examples are shown for sap flow measurements carried out on individuals of Erica arborea L., which has needle-like leaves, and Myrica faya Ait., a broadleaf species. We show that, although DeltaT(max) values obtained by the proposed method may be similar in some instances to the DeltaT(max) predicted at night, in general the values differ. The procedure presented has the potential of being applied not only to Granier's method, but to other heat-based sap flow systems that require a zero flow calibration, such as the Cermák et al. (1973) heat balance method and the T-max heat pulse system of Green et al. (2003).  相似文献

Under‐ as well as overfertilization with nitrogen (N) will result in economic loss for the farmer due to reduced yields and quality of the products. Also from an ecological perspective, it is important that the grower makes the correct decision on how much and when to apply N for a certain crop to minimize impacts on the environment. To aggravate the situation, N is a substance that is present in many compartments in different forms (nitrate, ammonium, organic N, etc.) in the soil‐plant environment and takes part in various processes (e.g., mineralization, immobilization, leaching, denitrification, etc.). Today, many N‐recommendation systems are mainly based on yield expectation. However, yields are not stable from year to year for a given field. Also the processes that determine the N supply from other sources than fertilizer are not predictable at the start of the growing season. Different methodological approaches are reviewed that have been introduced to improve N‐fertilizer recommendations for arable crops. Many soil‐based methods have been developed to measure soil mineral N (SMN) that is available for plants at a given sampling date. Soil sampling at the start of the growing period and analyzing for the amount of NO ‐N (and NH ‐N) is a widespread approach in Europe and North America. Based on data from field calibrations, the SMN pool is filled up with fertilizer N to a recommended amount. Depending on pre‐crop, use of organic manure, or soil characteristics, the recommendation might be modified (±10–50 kg N ha–1). Another set of soil methods has been established to estimate the amount of N that is mineralized from soil organic matter, plant residues, and/or organic manure. From the huge range of methods proposed so far, simple mild extraction procedures have gained most interest, but introduction into practical recommendation schemes has been rather limited. Plant‐analytical procedures cover the whole range from quantitative laboratory analysis to semiquantitative “quick” tests carried out in the field. The main idea is that the plant itself is the best indicator for the N supply from any source within the growth period. In‐field methods like the nitrate plant sap/petiole test and chlorophyll measurements with hand‐held devices or via remote sensing are regarded as most promising, because with these methods an adequate adjustment of the N‐fertilizer application strategy within the season is feasible. Prerequisite is a fertilization strategy that is based on several N applications and not on a one‐go approach.  相似文献

Arbuscular mycorrhizal (AM) development in different soil types, and the influence of AM fungal hyphae on their original soil were investigated. Plantago lanceolata, which can grow in soils of a very wide pH range, was grown in two closely related limestone soils and an acid soil from rock habitats. Plants were colonised by the indigenous AM fungal community. The use of compartmented systems allowed us to compare soil with and without mycorrhizal hyphae. Root colonisation of P. lanceolata was markedly higher in the limestone soils (30-60%) than in the acid soil (5-20%), both in the original habitat and in the experimental study. Growth of extraradical AM fungal hyphae was detected in the limestone soils, but not in the acid soil, using the signature fatty acid 16:1ω5 as biomass indicator. Analysis of signature fatty acids demonstrated an increased microbial biomass in the presence of AM fungal hyphae as judged for example from an increased amount of NLFA 16:0 with 30 nmol g−1 in one of the limestone soils. Bacterial activity, but not soil phosphatase activity, was increased by around 25% in the presence of mycorrhizal hyphae in the first harvest of limestone soils. AM fungal hyphae can thus stimulate microorganisms. However, no effect of AM hyphae was observed on the soil pH or organic matter content in the limestone soils and the available P was not depleted.  相似文献

Ectomycorrhizae (ECM) or the root‐fungal association in forest ecosystems provide a unique soil microenvironment where soil properties and processes differ from the bulk soil. In this study, we would like to better understand the role of ECM systems in mineral weathering and its implications to soil formation and nutrient cycling in forest ecosystems. Specifically, we would like to document the spatial variations in the composition of soil solution and mineralogy of the rhizosphere as influenced by the ECM of Norway spruce + Piloderma croceum. Two‐month‐old seedlings of Norway spruce (control and colonized by P. croceum) were cultivated in special rhizotrons designed to allow spatial collection of soil solution. We used A and C horizons of a Dystric Cambisol collected from Höglwald forest near Munich. Micro suction cups (5 mm x 1mm) were installed in colonized and control rhizotrons, and soil solution was collected from September to November 2000. Our results show that the concentrations of NH , Ca2+, and Mg2+ in the soil solution were lower in <1.0 cm than in >3.0 cm distance from the roots of Norway spruce, due to the possible range of influence of Piloderma mycelium reaching about 2–3 cm from the surface of the mycorrhizal root. In the rhizotron with soil from the A horizon, a higher phosphorus content in Piloderma‐colonized seedlings was observed. X‐ray diffraction data indicate that chlorite and possibly mica are being transformed to 2:1‐expanding clay minerals (probably smectite) within <1.0 cm distance from roots. The spatial variations in soil solution composition and mineral transformation are likely to be due to Piloderma colonization and concentrated mycelial growth within <1.0 cm distance from the roots. This is also evident in more intricate growth of mycelia on surfaces of micaceous minerals as compared to quartz. We assume that Piloderma modifies soil solution and mineralogy through acquisition of essential elements for its own survival and/or for the uptake by plant roots. However, the presence of spontaneous infection with wildtype ECM in the control plots may have altered the influence of Piloderma and must be taken into consideration when interpreting our results.  相似文献

Microscalic variability of soil solution chemistry - results of a laboratory experiment comparing standard - with micro suction cups In a laboratory experiment with an undisturbed soil column, the chemistry of soil solution collected by a standard suction cup (Ø 2 cm) was compared with that of 20 micro suction cups (Ø I mm) installed in the same soil depth. The standard cup showed comparable concentrations of inorganic anions with the soil column leachate, because preferably the main water paths of the soil column were sampled. In contrast, about 30 % of the micro suction cups sample soil compartments that have a different solution chemistry. In these cases the differences between standard and micro suction cups decrease in the order nitrate, chloride, sulfate. Standard suction cups seem to be the right sampling device for the investigation of element fluxes through soil. To get information about plant availability of ions they are inadequate, due to their dimension. Here micro suction cups are more appropriate, because their dimension is comparable to plant roots.  相似文献

Recent research shows that most soils are more or less water repellent. Already subcritical water repellency may cause incomplete soil wetting and preferential flow. Both processes potentially reduce the residence time of water and solutes in the vadose zone, resulting in an enhanced risk of groundwater contamination. The objective of the present paper is, therefore, to evaluate the impact of reduced soil wettability on the soil water infiltration rate and to investigate the tendency towards preferential flow with the analysis of the immobile water content in the infiltration zone. In november 2002, a field experiment was done in a coniferous forest, 30 km N of Hannover, Germany. Soil hydrophobicity was quantified by measuring the contact angles. The hydraulic conductivity of the podsolic sandy soil was measured depth‐dependent with a double‐ring tension infiltrometer in three soil horizons. To quantify possible preferential‐flow effects, a LiBr‐Tracer was added to the infiltrating water to evaluate the mobile water‐content fraction after infiltration. Additionally, infiltration rates of water were compared with infiltration rates of ethanol which were determined after water infiltration at the same locations. Results show that the actual water repellency of field‐moist soil was mainly subcritical (contact angle <90°). Water infiltration rates were reduced due to subcritical repellency by a factor of 3–170 compared with ethanol infiltration rates (exclusion of wetting effects). This spatially variable infiltration behavior was not clearly reflected neither by the small‐scale contact‐angle measurements nor by the analysis of the average immobile soil water content in the infiltration zone. We conclude that this specific infiltration behavior of water caused by small‐scale wettability effects may temporarily reduce the local connectivity of water‐flow pathways.  相似文献
