This research aims to investigate the relationship between the solvent retention capacity (SRC) test and quality assessment of hard red spring (HRS) wheat flour samples obtained from 10 HRS cultivars grown at six locations in North Dakota. The SRC values were significantly (P < 0.05) correlated with flour chemical components (protein, gluten, starch, and damaged starch contents, except arabinoxylan); with farinograph parameters (stability [FST], water absorption, peak time [FPT], and quality number); and with breadmaking parameters (baking water absorption [BWA], bread loaf volume [BLV], and symmetry). Differences in locations and cultivars contributed significantly to variation in quality parameters and SRC values. Suitability of SRC parameters for discriminatory analysis of HRS wheat flour is greatly influenced by molecular weight distribution (MWD) of SDS‐unextractable proteins. SRC parameters, except for sucrose SRC, showed significant (P < 0.01) and positive correlations with high‐molecular‐weight (HMW) polymeric proteins in SDS‐unextractable fractions, whereas only lactic acid SRC exhibited significant (P < 0.01) correlations with low‐molecular‐weight polymeric proteins. HMW polymeric proteins also exhibited positive associations with FPT, FST, BWA, and BLV. The discrepant variation in association of SRC parameters with respect to MWD of SDS‐unextractable proteins could improve segregation of HRS wheat flour samples for quality.  相似文献

The use of molecular methods for rotavirus characterisation provides increased sensitivity for typing and allows the identification of putative reassortant strains. Reagents and methods for genotyping the virus need constant modification because of the reassortant nature of the virus. This study was aimed at designing and evaluating new oligonucleotide degenerate primer pairs that provide increased sensitivity and specificity for detecting avian rotavirus. Gene-specific primer pairs were designed by analysing different rotavirus strains isolated during the last decade by downloading them from the GenBank. The alignments were generated using clustal analysis from the BioEdit program. Degenerate nucleotides were included due to the reassortant nature of rotavirus. The consensus sequences were aligned using the BioEdit program and then treated with the Fast PCR software to derive the primers. The derived primer sequences were submitted for a BLAST search to ensure alignment was exclusive to the desired target genes. The designed primers had specific bands and were efficient in detecting rotavirus in faecal samples than previously published primers. Thus, a successful surveillance of rotaviruses requires that primer pairs be updated regularly in order to detect the emergence of novel or “unusual types”, which have occurred by genetic drift causing nucleotide changes at the primer binding sites that result in typing failures. We recommend the use of the proposed primers in molecular surveillance studies for efficient detection of avian rotavirus.  相似文献

A 45-day experiment was carried out to evaluate the growth performance,digestive enzymes activity and biochemical response of Cirrhinus mrigala fed graded levels of oil.Three isonitrogenous purified diets(6%,8%and 10%of oil)were formulated.A total of 108 juveniles of Cirrhinus mrigala(5.95±0.25)g were equally distributed in triplicate groups in nine plastic tanks of 100 L capacity and fed under natural light cycle.Growth performance of the fish significantly reflected the effect of inclusion levels of oil tested but the diet with 8%lipid had the best significant growth performance[%weight gain,Feed Conversion Ratio(FCR)and Specific Growth Rate(SGR)],across different experimental groups.Levels of secretion of digestive enzymes in the fish fed different experimental diets resulted in significant values of(p0.05)amylase and lipase,while Aspartate transaminase(AST),Alanine transaminase(ALT),Lactate dehydrogenase(LDH)and Malate dehydrogenase(MDH)values in the muscle and liver decreased significantly(p0.05)as the levels of oil in the diet increased.However,serum biochemical parameters did not record any significant difference(p0.05)with the exception of the total lipid across diets.The study confirmed the inclusion effect of graded levels of oil in juvenile Mrigal diet,but the inclusion should not be more than 8%to avert growth depression.  相似文献

Purpose

Bioremediation of co-contaminated environments is difficult because of the mixed nature of the contaminants and the fact that the two components often must be treated differently. This study investigated the use of inorganic treatment additives, namely calcium carbonate (CaCO3), gypsum (CaSO4·2H2O), and disodium phosphate (Na2HPO4) to improve remediation of soil co-contaminated with 1,2-dichloroethane (1,2-DCA) and arsenic or cadmium.

Materials and methods

The soil samples were collected from a specific site in the Westville area in Durban, KwaZulu-Natal, South Africa. Microcosms were set up by artificially co-contaminating the soil sample (100 g mixed with 75 ml of synthetic groundwater in sterile screw-capped 250-ml serum bottles) with 1,2-DCA + risk elements; As3+ (150 mg/kg); or Cd2+ (170 mg/kg). Thereafter, each microcosm was amended with either 5 g CaCO3, 2 g CaSO4·2H2O, or 1.12 g Na2HPO4 + 0.046 g NaCl, separately. The samples were analyzed for the degradation of 1,2-DCA using GC–MS, while total 1,2-DCA degrading bacterial populations were determined at different sampling times using a standard spread plate technique. Soil dehydrogenase and urease activities were also monitored during the experimental period using standard enzyme assays.

Results and discussion

Addition of CaCO3 resulted in an approximately 2-fold increase in 1,2-DCA degradation in both the As3+ and the Cd2+ co-contaminated soil as compared to the co-contaminated soil without CaCO3. All the treatment additives were more effective in the As3+ co-contaminated soil resulting in 11.19, 9.25, and 5.63% increase in 1,2-DCA degradation in the presence of CaCO3, Na2HPO4 + NaCl, and CaSO4·2H2O, respectively, compared to the Cd2+ co-contaminated soil. The total 1,2-DCA degrading bacterial population increased in treated soils over time. Overall, soil dehydrogenase and urease activities were lower in the heavy metal co-contaminated samples compared to the treated soil. The inhibitory effect of heavy metal was less in As3+ co-contaminated soil for both CaCO3- and Na2HPO4 + NaCl-treated soil, with up to 7.92% increase in dehydrogenase activity obtained compared to soil co-contaminated with Cd2+.

Conclusions

Results from this study indicate that treatment additives can be used to reduce bioavailable fractions of risk elements in the soil matrices, thereby limiting the toxicity of these risk elements to 1,2-DCA degrading microorganisms. Thus, this approach can be applied to enhance organic compound degradation in co-contaminated soil environments.

The objective of this study was to evaluate the potential role of gramineous weeds present near paddy fields as alternative hosts for the Fusarium graminearum species complex (FGSC) that causes fusarium head blight (FHB) in rice. A total of 142 weed samples were collected from 10 gramineous weed species near paddy fields from August to October 2018 in Jiangsu Province, China. Of the 145 isolates of seven Fusarium species isolated from the weed samples, F. asiaticum was the most abundant (86.9%), followed by F. fujikuroi (5.5%), F. proliferatum (2.8%), F. graminearum (2.1%), F. tricinctum (1.4%), F. acuminatum (0.7%), and F. sporotrichioides (0.7%). Genotype and mycotoxin analyses confirmed that 72.2% of F. asiaticum isolates were producers of deoxynivalenol (DON) with 3-acetyl deoxynivalenol (3ADON), and the remainder were nivalenol (NIV) producers. Pathogenicity assays showed that both 3ADON and NIV chemotypes of F. asiaticum could cause FHB in rice, but NIV chemotypes were significantly (p < .05) more aggressive than 3ADON chemotypes. Three Fusarium mycotoxins, DON, NIV, and zearalenone, occurred naturally at low concentrations in the weed samples. Taken together, this study provides insight into the mycotoxin production and aggressiveness of F. asiaticum isolates from gramineous weeds in China.  相似文献

Avian coccidiosis is one of the most important diseases of poultry and it is responsible for a large number of all broiler mortalities worldwide. The control of this disease relies mainly on the use of anticoccidial drugs. However, herbal preparations could be an alternative for the treatment against coccidiosis in chickens. The direct effects of Moringa oleifera acetone extracts on broiler chickens naturally infected with mixed Eimeria species was investigated to determine the relative efficacy of the extracts against coccidiosis in birds. The investigations were carried out in seven groups (ten chickens per group). The birds were given various doses (1.0, 2.0, 3.0, 4.0 and 5.0 g/kg body weight) of acetone extract of leaves of M. toltrazuril® (positive control) and untreated (negative control). The extract was evaluated for anticoccidial activity by means of inhibition of oocyst output in faeces, faecal score, weight gain and mortality. Haematological indices were evaluated by standard methods. The group treated with 1.0 g/ kg body weight Moringa oleifera extract produced the least inhibitory effect on oocyst shed in the faeces (96.4 %), while the groups treated with 2.0 g/kg, 3.0 g/kg, 4.0 g/kg and 5.0 g/kg body weight of the extract produced 97.4, 98.7, 99.1 and 99.8 %, respectively. Body weight gains of infected chickens treated with the extract significantly improved (p?<?0.05), and faecal scores were milder. Packed cell volume, haemoglobin concentration and red blood count of the treated birds were significantly (p?<?0.05) higher than those of the infected untreated group. Moringa oleifera leaves could find application in the treatment of avian coccidiosis in veterinary practice.  相似文献

The combination of inorganic fertilizers and compost is a technique aimed at improving crop growth and maintaining soil health. Understanding the rate of nutrient release from enriched compost is important for effective nutrient management. A laboratory incubation study was conducted for 112 days to study the nutrient mineralization pattern of poultry manure compost enriched with inorganic nitrogen (N) and phosphorus (P) fertilizer nutrients in an Ultisol. Compost applied at the rate of either 5 or 10 g kg?1 was blended with N (50 kg N ha?1) and P (30 kg P ha?1). Carbon dioxide evolution and N and P mineralization were measured fortnightly. The bacterial and fungal populations were determined at the mid and end of the experiment. The combination of compost and inorganic N and P increased carbon (C) and P mineralization by 4?8% and 56?289%, respectively, over the application of either compost or inorganic N and P. However, P addition influenced the amount of C mineralized. Inorganic N and P, on the other hand, were better at increasing N mineralization than compost blended with inorganic N and P over a short time. The addition of compost stimulated bacterial and actinomycete populations, while fungal populations were unaffected. Actinomycetes and bacteria had similar and higher relationship trend with C (R2 = 0.24) and P (R2 = 0.47) mineralization and were key determinants in nutrient mineralization from compost in this Ultisol. Integrating compost with inorganic fertilizers improves nutrient availability through the growth and activities of beneficial microorganisms.  相似文献

A case of a rapidly fatal disease in polo horses caused by African horsesickness (AHS) virus serotype 2 is described. The pattern of polo tournaments, environmental/management conditions (moist and warm), as well as probable importation strategies with no regard for import control and quarantine, favored introduction and spread of the virus. The outbreak, which involved a large number of horses, was characterized by severe respiratory distress, fever, supraorbital edema, and death. This is the first time a widespread epidemic of AHS has been reported in Nigeria and this is the first report of AHSV serotype 2 in the northern hemisphere. In addition, we amplified the complete genome of the virus using RNA extracted from clotted blood. This report indicates that AHS is expanding its geographical territory northwards and assuming a new microbial ecology.  相似文献

The current control of parasitic nematodes in small ruminants relies on the use of chemical anthelminthics, but the development of resistance and the problem of drug residues require research into alternatives. Acetone extract and solvent–solvent fractions of Vernonia amygdalina Del. (Compositae) were evaluated in vitro for potential anti-parasitic effects against the eggs and larvae of Haemonchus contortus. Significant effects were obtained with the extract and fractions but differences were observed depending on the parasitic stage. The acetone extract and fractions inhibited egg hatching and inhibited larval development and killed larvae of H. contortus in a concentration-dependent manner. Best-fit 50% lethal concentration (LC50) values were 957.0, 76.0, 524.0, 309.0 and 224.0 μg/ml for the acetone extract, and the butanol, hexane, chloroform and 35% water in methanol fractions, respectively, when tested against nematode eggs. Best-fit LC50 values for the larval viability test were 508.2, 485.5, 569.3, 348.9 and 196.6 μg/ml for the acetone extract, and the butanol, hexane, chloroform and 35% water in methanol fractions, respectively. The butanol fraction was most active against nematode eggs while the 35% water in methanol fraction was the most active on nematode larvae, although differences in activity between fractions were not significant (p > 0.05). Overall, these in vitro results suggest that V. amygdalina, traditionally used by small farmers in Western Africa, does possess anti-parasitic properties.  相似文献
