This study aimed to evaluate the effect of Lactococcus lactis K-C2 on the growth performance, microbial diversity, and release of free amino acids in the intestinal tract and the edible parts of young amberjack, Seriola dumerili. Fish were fed a diet with or without strain K-C2 (2?×?1010 cfu/g feed) for 25 days. The results indicated that the growth performance of fish in the treated group was significantly higher than those in the control group (p?<?0.05). The amount of five amino acids (aspartate, sarcosine, taurine, alanine, and arginine) in the gut content and 13 of 21 amino acids in the edible parts of fish in the treated group were significantly higher (p?<?0.05) than those in the control group. Sphingomonas, Propionibacterium, and Mycobacterium were observed in gut microflora of fish in both the control and treated groups. Staphylococcus and Kocuria were detected in one sample from the control and treated groups; Acinetobacter and Acidobacteria were found in one sample from the control group. L. lactis was only found in one sample in the treated group. In conclusion, the dietary administration of probiotic L. lactis stimulated growth, reduced feed consumption, and improved the nutritional value of cultured amberjack.  相似文献

Recently, we reported that intracerebroventricular (i.c.v.) injection of reduced glutathione (GSH) induces hypnotic and sedative effects under an acute stressful condition in chicks. However, no information is available on the effects of GSH on energy expenditure (EE) under stressful conditions. The purpose of the present study was to clarify whether i.c.v. injection of GSH affects EE of neonatal chicks, and whether EE is correlated with behavioral changes after isolation-induced stress. The EE was rapidly decreased by i.c.v. injection of GSH, but was increased 27 min after injection. This change in EE was correlated with behavioral changes in which GSH induced hypnotic and sedative effects shortly after injection, followed by a period in which activity increased. The present study demonstrates that central GSH initially causes lowered EE through hypnotic and sedative effects under an acute stressful condition in chicks.  相似文献

Hibiscus syriacus, as a national flower of Korea, is most popularly used for ornamental purposes and includes numerous cultivars, and it is widely planted in temperate zones that feature hot summers. We investigated Choanephora flower rot on H. syriacus from 2012 to 2014 in Korea and Japan and confirmed Choanephora infection in several localities in both countries. Here, our objectives were to identify the main causal agent of Choanephora flower rot on H. syriacus and describe its morphological and molecular characteristics. We identified 44 out of 50 isolates as Choanephora cucurbitarum and the remainder as C. infundibulifera based on morphological characterization and phylogenetic analysis. The sequences of the internal transcribed spacer region (ITS) of ribosomal DNA and the D1/D2 region of the large subunit (LSU) rDNA of examined isolates were compared with sequences obtained from GenBank, and the analysis of the results revealed 100 % identity with the corresponding sequences of C. cucurbitarum and C. infundibulifera strains. Classification of the Choanephora species performed here according to the key described by Kirk (1984) corresponded with the results of the phylogenetic analysis of this study. Through intraspecific and interspecific mating tests, the characteristics of zygospore were described in details. Pathogenicity tests using both species showed the same symptoms, causing blossom blight and soft rot on the flowers, which were identical to those observed in the field. All identified causal agents of Choanephora rot were indeed Choanephora species, where C. cucurbitarum was identified in the majority, while the others were in the minority of examined samples.  相似文献

In the present study, the loop-mediated isothermal amplification (LAMP) assay was developed to amplify the fragments of the O9 Salmonella-specific insertion element and evaluated in the laboratory for its potential use in a field situation, such as poultry farms. Among the bacteria tested, a positive reaction was observed only for 128 strains of 6 serovars of the O9 group Salmonella, such as Enteritidis (SE) and Pullorum. The detection limit of the LAMP assay was 10(3)CFU/ml, which was more sensitive than that of the polymerase chain reaction (PCR) assay with the same target gene (10(6)CFU/ml). The final results were obtained within 30 min for the LAMP assay, while the PCR assay needed a total of 120 min. When the LAMP assay was applied to the enrichment broth mixed with cecal dropping samples either spiked with SE in vitro or excreted by SE-inoculated hens, the results were comparable to those of the conventional plating method including 2 separate enrichments. In conclusion, the LAMP assay developed in the present study is an effective method for the specific detection of the O9 group Salmonella serovars, including SE.  相似文献

This study compared the immunosuppressive effects of dexamethasone (DEX), flunixin meglumine (FLU) and meloxicam (MEL) on the peripheral blood mononuclear cells (PBMCs) of seven healthy Holstein calves in vitro. DEX significantly inhibited lymphocyte proliferation and expression of interferon (IFN)-γ, interleukin (IL)-2 and IL-4 messenger RNA (mRNA) in comparison with FLU and MEL. FLU and MEL dose-dependently inhibited lymphocyte proliferation, but did not significantly reduce mRNA expression. Our in vitro study indicates that steroidal anti-inflammatory drugs (SAIDs) as well as nonsteroidal anti-inflammatory drugs (NSAIDs) have immunosuppressive effects on calf PBMCs. These findings are important for assessing the indications and complications of NSAIDs in calves.  相似文献

The objective of this research was to develop a low-cost attitude sensor for agricultural vehicles. The attitude sensor was composed of three vibratory gyroscopes and two inclinometers. A sensor fusion algorithm was developed to estimate tilt angles (roll and pitch) by least-squares method. In the algorithm, the drift error of the gyroscopes was estimated using the inclinometers. In addition to tilt angles, the attitude sensor also estimated the absolute heading angle and position with inclination error correction by integrating a GPS. Tests were conducted on a flat field, a sloping ground and a bumpy road. Results showed that the attitude sensor was able to estimate the roll angle with the maximum root mean square error of 0.43°, the pitch angle with 0.61° and the heading angle with 0.64°. Moreover, the attitude sensor dramatically improved the positioning accuracy from 25.9 cm to 3.0 cm in the sloping ground test and from 8.4 cm to 3.7 cm in the bumpy road test. The proposed technology used in the attitude sensor will help to develop advanced agricultural applications.  相似文献

Japanese Black (JB) calves are more susceptible to infectious diseases compared to Holstein (Hol) calves. To clarify the immunological differences between JB and Hol calves, expression of cytokine messenger RNA (mRNA) was examined using peripheral CD4+, CD8+ and γδ T cells. Healthy calves, 24 from each species, were examined. Blood samples were obtained from calves at 1 week, 1 month and 3 months old, eight calves for each age of each species. Peripheral blood mononuclear cells were stimulated with phytohemagglutin (PHA), and T cell subsets were isolated by positive selection using magnetic cell sorting (MACS). Levels of interlekin (IL)‐2, IL‐4, IL‐10 and interferon (IFN)‐γ mRNA in three T cell subsets were analyzed. WC1‐N1+ γδ T cell percentages were significantly lower in JB calves at 1 week and 1 month of age compared to Hol calves. In addition JB calves had significantly lower IL‐2, IL‐10 and IFN‐γ mRNA in WC1‐N1+ γδ T cells at 1 and 3 months of age, whereas there were no significant differences in cytokine mRNA of CD4+ and CD8+ cells between the two groups. Decreased cytokine mRNA and cell number of peripheral γδ T cells may affect negatively on the immune system of JB calves.  相似文献

Use of live and dead probiotic cells in tilapia Oreochromis niloticus   总被引:3,自引:0,他引:3  
ABSTRACT:   To investigate the effect of live and dead probiotic cells on the non-specific immune system of tilapia Oreochromis niloticus , probiotics were introduced by feeding either in the form of live or dead cells, or supplying live cells to the rearing water in a closed recirculating system. The probiotics treatment enhanced non-specific immune parameters such as lysozyme activity, migration of neutrophils and plasma bactericidal activity, resulting in improvement of resistance to Edwardsiella tarda infection. Especially, oral administration of live cells seemed to be more effective compared with other probiotic treatments such as oral administration of dead probiotic cells and supply of live probiotic cells to the rearing water. These results indicate that probiotics treatment is promising as an alternative method to antibiotics for disease prevention in aquaculture, and the viability of probiotic bacteria is a key factor to induce more potential effect of probiotics used for fish production.  相似文献
