Routine gill swabbing is a non-destructive sampling method used for the downstream qPCR detection and quantitation of the pathogen Neoparamoeba perurans, a causative agent of amoebic gill disease (AGD). Three commercially available swabs were compared aiming their application for timelier AGD diagnosis (Calgiswab® (calcium alginate fibre-tipped), Isohelix® DNA buccal and cotton wool-tipped). Calcium alginate is soluble in most sodium salts, which potentially allows the total recovery of biological material, hence a better extraction of target organisms’ DNA. Thus, this study consisted of (a) an in vitro assessment involving spiking of the swabs with known amounts of amoebae and additional assessment of retrieval efficiency of amoebae from agar plates; (b) in vivo testing by swabbing of gill arches (second, third and fourth) of AGD-infected fish. Both in vitro and in vivo experiments identified an enhanced amoeba retrieval with Calgiswab® and Isohelix® swabs in comparison with cotton swabs. Additionally, the third and fourth gill arches presented significantly higher amoebic loads compared to the second gill arch. Results suggest that limiting routine gill swabbing to one or two arches, instead of all, could likely lead to reduced stress-related effects incurred by handling and sampling and a timelier diagnosis of AGD.  相似文献

Climate change will increase the frequency of extreme rain events, causing more flooding episodes. Willows are usually planted in marginal lands like flood prone areas. For willow plantations developed from rootless cuttings, the establishment phase is crucial, because the cuttings are still developing a shoot and root system and have a higher vulnerability to stress. A flooding episode during this early period may have a negative effect upon plants. We analyzed the responses to flooding of eight willow genotypes, representing important species from the economic and ecological point of view (Salix alba, S. matsudana, S. amygdaloides and S. matsudana × S. nigra hybrids). The treatments started when the plants were 2 months old and lasted for 3 weeks. They were identified as: Control (watered to field capacity); F10 (plants submerged 10 cm above soil surface) and F50 (plants submerged 50 cm above soil surface). The F50 treatment showed a greater growth reduction than the F10 treatment in most clones, either measured in height, diameter or total biomass. Both flooding treatments reduced significantly the root-to-shoot ratio compared to control plants. The F50 treatment increased the leaf nitrogen content and specific leaf area in all genotypes. Both treatments changed plant and leaf traits in different ways according to the depth of the floodwater. These changes may have lasting effects on growth recovery in the post-flooding period. Willow clones with a fast initial growth will be the best option to minimize growth reduction in areas prone to experience flooding episodes.  相似文献

The products that employ nanoparticles (NPs) in their composition have increased since the beginning of NP production; hence, their availability in the environment, especially in aquatic ecosystems, tends to increase. In these ecosystems, the phytoplankton is immersed in a complex matrix of nutrients, excreted materials, and other chemical compounds, which can influence the metabolic strategy of microalgae. One of the metabolic ways is mixotrophy, a situation whereby microalgae perform photosynthesis and use dissolved organic carbon at the same time. Most toxicity evaluations do not consider such a metabolic route, but this can represent a preferential metabolism in natural environments. The present study aimed at evaluating the effects of NP-TiO2 at a log concentration range of ??3.10 to 0.89, on photosynthesis, growth, viability, and biochemical composition of the microalgae Chlorella sorokiniana during photoautotrophic and mixotrophic growth (glucose as the organic carbon source). The results showed lower chlorophyll a and photosynthetic activity in mixotrophy than in photoautotrophy, which can be due to a decreased need for photosynthesis in mixotrophy. Photoautotrophy cultures were sensitive to NPs, reaching 39% of viability at log 0.89, while in mixotrophy, cell viability was not affected by NPs. The biochemical composition and cell density changed as a function of NP concentrations, with increase in the protein/carbohydrate ratio in both treatments. The results showed that C. sorokiniana is more resistant to NPs during mixotrophic growth, but with changes in biochemical composition, whereas the photoautotrophic cultures were more sensitive to the increase in NP concentrations.  相似文献

The Portuguese oyster Crassostrea angulata shows great potential in oyster farming. The conservation of pure populations of this species is important for production diversification and biodiversity preservation. In this way, the zootechnological development for seed hatchery production is extremely important. Broodstock conditioning is a key step in the process of rearing bivalves in a hatchery. Many factors regulate the reproductive cycle, being food one of the most important ones. To evaluate the effect of different diets on C. angulata reproductive performance, broodstock were conditioned with different food regimes formulated fundamentally by flagellates (Diet 1 – Pavlova lutheri and Isochrysis galbana clone T‐ISO; Diet 2 – P. lutheri, T‐ISO and Skeletonema costatum) and constituted fundamentally by diatoms (Diet 3 – S. costatum and Chaetoceros calcitrans; Diet 4 – P. lutheri, S. costatum and C. calcitrans). During conditioning, samples of oysters were collected to evaluate condition index, gonadal development and biochemical composition. At the end of the conditioning period, oysters were induced to spawn to evaluate reproductive output (fecundity, fertilization rate and D‐larvae development). The diets had an impact on the gametogenesis process, energy storage and reproductive output performance, being the best results those obtained in broodstock fed with the diatoms‐predominant diets. However, those fed with diets majority flagellates had an unsuccessful performance. Holistic approaches incorporating all results in this study reveal and reinforce the idea that the diatom species used presented the nutritional requirements to C. angulata broodstock, being essential in the conditioning phase.  相似文献

Conservation of threatened animals is frequently limited by lack of knowledge about their ecological preferences, and often artificial feeding is one of the few chances to save endangered species. We investigated the possibility to artificially feed two endangered flat bark beetles dependent on dead wood for their diet—namely, Cucujus cinnaberinus (Scopoli, 1763) and Cucujus haematodes Erichson, 1845—by examining their dietary preferences, life cycle duration, and survival in laboratory conditions. Individuals of the two species were caught in the wild and larvae and adults were fed in laboratory conditions by live or dead prey. Three species of saproxylic beetles: two cerambycids (Acanthocinus griseus Fabricius, 1793 and Rhagium inquisitor Linnaeus, 1758), one scolytid (Ips sexdentatus Börner, 1776) one tenebrionid (Tenebrio molitor (Linnaeus, 1758)) one dipteran (Lucilia sericata (Meigen, 1826)) and one ant (Lasius sp. Fabricius, 1804) were used as prey, with minced meat as a control. Our results indicated high survival and no difference in prey choice between the two flat beetle species. Larvae and adults preferred dead prey, but no significant preference was detected among dead prey taxa, supporting the hypothesis that the two species are opportunistic scavengers. Comparing data with previous results, both species and their developmental stages should be classified as obligate saproxylic organisms with preference to the dead and decaying organic material. Successful artificial feeding and rearing of these endangered species, followed by the release in the wild through rescue or reintroduction programs, therefore appear relevant for their protection and future conservation.  相似文献

The consequences of over-exploitation may seriously impair the integrity and functioning of ecosystems. When loss of species is accompanied by the loss of ecological processes, the effects are no longer just taxonomic and may affect the stability of the environments. Ornamental fishing is one of the main economic activities of the middle Negro River. Such activity is directed to a small number of species and has the Cardinal Tetra (Paracheirodon axelrodi) as the main target species. Given the potential effects this pressure and the lack of information on its consequences, the present study analysed, through simulations, the possible effects of depletion of ornamental fish populations on the functional structure of fish assemblages, represented by functional diversity (FD) and functional redundancy (FR) indexes. We sampled 13 streams exploited by the ornamental fishery, where we collected 4,286 specimens of 110 species, of which 22 were targeted as ornamental fishing. We found that the ornamental species corresponded, on average, to 27.8% of the species and 33.3% of the FD of the assemblages. In a scenario of complete exclusion of ornamental species, local communities would lose 24.6% of the FD and reduce up to 12% of the FR. With these results, it is possible to infer that the local extinction of exploited species would cause negative impacts on the multifunctionality and the resilience of the streams ecosystems. In this way, we emphasise the need for proper management of the ornamental fisheries aiming to the sustainability of the activity and to the conservation of ecosystem functionality.  相似文献

In this study, the effects of three diets were investigated to enhance Paracentrotus lividus production for commercial purposes. P. lividus were fed ad libitum for 80 days with: diet A—fresh Codium tomentosum Stackhouse, 1797; diet B—formulated using a jellified mix of macroalgae and vegetables, including C. tomentosum (20%), Coralina sp. Linnaeus, 1758 (17%), cabbage Brassica oleracea var. capitata Linnaeus, 1753 (30%), carrot Daucus carota Linnaeus, 1753 (30%) and agar (3%) as a gelling agent. Diet C consisted of maize Zea mays Linnaeus, 1753 (56%) and New Zealand spinach Tetragonia tetragonoides (Pallas, 1781) Kuntze, 1891 (44%). Their effects on the gonadal and somatic growths, gonadosomatic index (GI) and gametogenesis were evaluated, as well as on the total lipid content and fatty acid composition of sea urchin's gonads. Diet A provided high values of eicosapentaenoic acid (EPA). Gonads of sea urchins fed with diet A were found mostly in growth and maturation stages of gametogenesis and showed the lowest lipid content. Sea urchins fed with diet B presented their gonads in the reabsorption stage and had the highest values of omega‐3 polyunsaturated fatty acids (PUFAs). Sea urchins fed with diet C were in the early stages of gametogenesis and had the highest values of lipid content, plus omega‐6 PUFAs. Once as an ingredient in a balanced mix with vegetables, C. tomentosum can be a key factor to the development of new promising high‐quality and low‐cost feed for P. lividus roe enhancement.  相似文献

The aim of this study was to evaluate the effect of hydrogen peroxide, iodine solution (PVP) and methylene blue on eggs disinfection of three ornamental fish species, Danio rerio, Pterophyllum scalare and Gymnocorymbus ternetzi. The main idea was to create conditions to enhance the hatching rates. Eggs of each species were exposed to different concentrations of hydrogen peroxide (5, 10, 15 and 25 mg/L), PVP (0.25, 0.5, 0.75 and 1 mg/L) and methylene blue (0.5 1, 2 and 3 mg/L). The optimal doses ranged between species and chemicals: for G. ternetzi, the concentrations that high enhanced the hatching rate were 1 mg/L for the PVP treatment, 25 mg/L for the hydrogen peroxide treatment and 3 mg/L for methylene blue treatment; for P. scalare, the best results were achieved with 25 mg/L for hydrogen peroxide treatment and 3 mg/L for methylene blue treatment. By contrast, for all the different chemical did not increased the D. rerio hatching rate. Results showed that hydrogen peroxide and methylene blue are the most versatile, effective and safe to use in these species. On the other hand, PVP can be used but with many precautions due to very low safety margin. Results clearly show that the optimal concentration of chemicals for eggs disinfection is fish species dependent and it is completely wrong to extrapolate concentrations between different chemicals and fish species. Our study suggests that P. scalare can be used as a model in study of effectiveness of new chemicals with potential to disinfect water and increase hatching rates.  相似文献
