Enhancing capacity for freshwater conservation at the genetic level: a demonstration using three stream macroinvertebrates 下载免费PDF全文
- Species diversity is declining more rapidly in freshwater ecosystems than in any other, but the consequences for genetic diversity, and hence evolutionary potential, are poorly understood. In part this reflects limited use and development of modern molecular tools and genetic approaches to address conservation questions in rivers, lakes and wetlands. As widespread, diverse and functionally important organisms, freshwater macroinvertebrates are ideal candidates for genetic approaches to reveal, for example, the conservation consequences of demographic histories and past disturbances. However, the availability of microsatellite markers for this group is very limited.
- Using next generation sequencing, microsatellite markers were developed for Isoperla grammatica (Poda, 1761), Amphinemura sulcicollis (Stephens, 1836) and Baetis rhodani (Pictet, 1843) to enable conservation genetic investigations of these widespread invertebrate species. Fifty‐two robust microsatellite loci were developed (18, 21 and 13 per species), all with high levels of allelic diversity (7–27, 3–16, 5–13 alleles per loci, respectively).
- These tools will allow assessment of genetic structure, dispersal and demographic resilience in these model species as a function of environmental change and variation, thereby aiding freshwater monitoring and conservation. The authors urge further capacity building to support genetic applications to the conservation biology of other aquatic organisms.

Formal public sector soybean breeding in Africa spans over four decades, and it was initiated by the International Institute of Tropical Agricultural (IITA). As the demand of soybean continues to outstrip production, strategic projects such the Tropical Legume (TL) were initiated, in which the main goal was to enhance the productivity of soybean in the farmers’ fields in Sub‐Saharan Africa. One of the strategies to enhance the productivity of soybean in the farmers’ fields is through developing and deploying improved soybean varieties in the target countries. Through the TL I and TL II projects, a number of varieties were released in the target countries, Kenya, Nigeria, Malawi and Mozambique by employing participatory variety selection (PVS). This review provides highlights of the achievements made by IITA breeding programme and insights of what needs to be done to enhance yield improvement for soybean in Africa using demand‐driven breeding approaches. 相似文献

Irregular plagues of house mice, Mus musculus, incur major economic impacts on agricultural production in Australia. The efficacy of zinc phosphide (ZnP), the only registered broadacre control agent for mice, is reported as increasingly variable. Have mice become less sensitive over time or are they taking a sub-lethal dose and developing aversion? In this laboratory study, the sensitivity of mice (wild caught; outbred laboratory strain) was assessed using oral gavage of a range of ZnP concentrations. The estimated LD50 values (72–79 mg ZnP/kg body weight) were similar for each mouse group but are significantly higher than previously reported. The willingness of mice to consume ZnP-coated grains was determined. ZnP-coated grains (50 g ZnP/kg grain) presented in the absence of alternative food were consumed and 94% of wild mice died. Mice provided with alternative food and ZnP-coated wheat grains (either 25 or 50 g ZnP/kg grain) consumed toxic and non-toxic grains, and mortality was lower (33–55%). If a sublethal amount of ZnP-coated grain was consumed, aversion occurred, mostly when alternative food was present. The sensitivity of wild house mice to ZnP in Australia is significantly lower than previously assumed. Under laboratory conditions, ZnP-coated grains coated with a new higher dose (50 g ZnP/kg grain) were readily consumed. Consumption of toxic grain occurred when alternative food was available but was decreased. Our unambiguous findings for house mice indicate a re-assessment of the ZnP loading for baits used for control of many rodents around the world may be warranted. 相似文献
