Rice (Oryza sativa) plants acquired excess photosynthates in the form of nonstructural carbohydrates (NSCs) in their stems and grain. Despite keen interest in rice NSC, the dynamics of NSC accumulation, translocation and re-accumulation have not yet been well investigated. AG1 and AG2 QTLs associated with flooding tolerance through catalyzing starch into soluble sugar in germinating seeds. Here we conducted three experiments, greenhouse and field to lay the groundwork for large-scale diversity studies on grain NSC and some agronomic traits under direct-seeded rice (DSR) system, using elite lines incorporating AG1, AG2 and AG1-AG2 QTLs into the popular varieties PSB Rc82 and Ciherang-Sub1 along with the donors Kho Hlan On (AG1) and Ma-Zhan Red (AG2). In germinating seedlings, soluble sugars increased, while starch concentration decreased gradually especially in the tolerant checks and AG1-AG2 introgression lines under flooded soil. Soluble sugar accumulation in stem started to increase from the vegetative stage and peaked at the panicle initiation stage then gradually decreased towards the maturity stage. But Sub1-AG lines had higher sugar and starch concentrations at different growth stages than other genotypes in wet season 2016 and dry season 2017. Plant survival rate was positively correlated with the stem NSC at the early vegetative stage (21 days after sowing), and stem NSC was positively associated with plant height at different growth stages. Among the tested seeding rate, the most suitable seeding rate, 4 g/m2 with shallow burial depth (0.5 cm), resulted in better seedling establishment, relatively higher seedling vigor index and higher leaf area index under flooding in DSR system. Introgression of AG1-AG2 QTLs had no any negative impact on nonstructural carbohydrate, germination rate, and growth and biomass production.  相似文献

Stagnant flooding (SF) during vegetative growth triggers stem elongation usually at the cost of tiller production in rice, reducing grain yield. To explore physiological mechanisms associated with tillering suppression under SF, three contrasting genotypes (Swarna and Swarna‐Sub1, both sensitive and IRRI154, tolerant) were evaluated under standing water depths of 0, 5, 30 and 50 cm. SF significantly suppressed tiller formation but increased plant height, root biomass, shoot elongation (ratio of plant height before and after flooding), leaf emergency and non‐structural carbohydrate (NSC) concentration (in root–shoot junction) in all genotypes at the early stage of development. Chlorophyll concentration in the upper leaves (upper most fully expanded leaf at top) was higher than in lower leaves (lowest green leaf at base), but decreased under SF in both. SF increased hydrogen peroxide (H2O2) at the early stage of treatment, with concomitant increase in malondialdehyde (MDA) production by stems and leaves. MDA concentration in root–shoot junction increased but delayed. Tiller number correlated negatively with plant height, shoot elongation, leaf emergency, MDA concentration in leaves and root–shoot junction, root biomass, and NSC concentration in the root–shoot junction. The results suggested existence of compensatory mechanisms between tiller growth and shoot elongation in rice for resilience under SF, where energy is mainly diverted for shoot elongation to escape flooding. The SF‐tolerant genotype produced less H2O2 and maintained energy balance for higher survival and better growth under stagnant flooding.  相似文献

A series of active biodegradable coatings based on chitosan, gelatin, starch, and sorbitol with or without monoterpenes (geraniol and thymol) were prepared and applied on fresh strawberry fruit as postharvest treatments. The coated fruits were inoculated with fungal spores of Botrytis cinerea and stored for 7 days at 4 °C. Decay incidence, weight loss, anthocyanins, total soluble solids (TSS), total soluble phenolics (TSP), polygalacturonase (PGase), pectin-lyase (PLase), antioxidant activity, guaiacol peroxidase (G-POD), polyphenol oxidase (PPO), and catalase (CAT) were elucidated in the fruits of experimental sets and they were compared with that of controls. The coatings showed a significant effect on the development of quality variables, with the additional effect of geraniol and thymol as a function of the polysaccharide matrix. The coatings inhibited decay incidence, reduced weight loss, delayed changes in the contents of anthocyanin, TSS, and total soluble phenolics; inhibited the increase in G-POD and PPO activities; and retarded the reduction in CAT activity. Compared to the controls, all of the coatings had positive effects on the inhibition of cell wall degrading enzymes (PGase and PLase) and among all the tested coatings, T5, chitosan (1%) + starch (1%) + sorbitol (0.5%) + tween (0.05%) + thymol (0.02%) was the best. This formulation showed also the highest antimicrobial activity and the greatest effect on other physiochemical parameters and it can be suggested to use it as a useful coating agent for extending the shelf-life and maintaining quality of strawberry fruit.  相似文献

The in vivo and in vitro mechanistic anti-inflammatory actions of cucurbitacin E (CE) (Citrullus lanatus var. citroides) were examined. The results showed that LPS/INF-γ increased NO production in RAW264.7 macrophages, whereas L-NAME and CE curtailed it. CE did not reveal any cytotoxicity on RAW264.7 and WRL-68 cells. CE inhibited both COX enzymes with more selectivity toward COX-2. Intraperitoneal injection of CE significantly suppressed carrageenan-induced rat's paw edema. ORAC and FRAP assays showed that CE is not a potent ROS scavenger. It could be concluded that CE is potentially useful in treating inflammation through the inhibition of COX and RNS but not ROS.  相似文献

Zizyphus oxyphylla Edgew is used in Pakistan as a folk medicinal remedy for inflammatory conditions, pains especially rheumatic pain, fevers, allergy and diabetes. The aim of the current study was to scientifically validate the folk use of Z. oxyphylla Edgew by using the isolated compound in vitro and in vivo levels. Kushecarpin A was isolated from ethyl acetate fraction of the plant crude extract. Molecular docking simulations predicted Kushecarpin A as a potential new lipoxygenase (LOX) inhibitor. Kushecarpin A showed significant lipoxygenase inhibition (IC50: 7.2 ± 0.02 μM) thus validated computational prediction. It also exhibited significant and highly significant inhibition (p < 0.05 and p < 0.01) of carrageenan-induced hind paw oedema at the doses of 5, 10 and 20 mg/kg. Kushecarpin A seems to be a potentially new anti-inflammatory compound responsible for anti-inflammatory activities of Z. oxyphylla Edgew. In vitro and in vivo anti-inflammatory inflammatory activities were found in good agreement with the folk medicinal use of Z. oxyphylla Edgew in inflammatory disorders.  相似文献

The conversion and degradation of malonylglucosides were kinetically characterized under elevated pH/heat conditions. Malonylgenistin and malonyldaidzin were heated at 60, 80, and 100 degrees C and pH values of 8.5, 9, and 9.5. A simple kinetic model was developed, which adequately predicted the conversion and degradation reactions. The conversion and degradation rates increased as temperature and pH increased. The rates of conversion of both malonylglucosides into their respective beta-glucosides were comparable under all pH/heat treatments. However, at 100 degrees C, the rates of degradation of malonyldaidzin were approximately double those of malonylgenistin, under all pH treatments. When malonlydaidzin was heated at 100 degrees C and pH 9.5, degradation of the produced daidzin occurred. Therefore, an alternative kinetic model was developed to better predict the conversion and degradation of malonyldaidzin occurring at 100 degrees C and pH 9.5. The models developed provide soy food manufacturers with guidelines for better control of the profile and level of isoflavones..  相似文献

Abstract.— Amino acid profiles of freshwater prawn Macrobrachium rosenbergii were determined during early larval stages (I-IX) to provide some baseline biochemical information of the growing larvae. The larvae obtained from several females were stocked into three 250-L tanks at a density of 30 larvae/L. The feeding regime consisted of newly hatched Artemia nauplii and egg custard containing 5% cod liver oil. For each developmental stage, larval samples from each experimental tank were pooled, freeze dried and after acid hydrolysis, the quantification of amino acids was done by a reverse phase high performance liquid chromatography ( N = 2). The highest represented of the total amino acids were for glutamic acid and phenylalanine (with cystine) with ranges from 13.4–16.6 and 9.7–11.5%, respectively, whereas tryptophan (1.4–1.6%). methionine (1.4–2.7%) and histidine (2.9–4.2%) were relatively lower. The levels of the following essential amino acids did not significantly change during larval development: arginine, leucine, phenylalanine (plus cystine), threonine, tryptophan and valine. Despite statistically significant changes that were observed in levels of some amino acids, amino acid composition appeared to be relatively unchanged during the larval growth. The results may suggest that the amino acid requirements of the freshwater prawn is relatively constant during larval life and can be satistied by a suitable protein source that resembles the larval amino acid profile.  相似文献

Malaysian Mahseer (Tor tambroides) has a good prospect for aquaculture because of its high market demand. However, there is a scarce information on gut microbiota associated with Malaysian Mahseer unlike other fish species. Therefore, we constructed and compared gut microbiota in different developmental stages (larval, juvenile, fingerling, yearling, and adult) using culture dependent and PCR‐DGGE fingerprinting technique for better understanding of gut microbiota composition associated with T. tambroides. Culturable gut microbiota composition in all developmental stages were composed of β‐ and γ‐Proteobacteria, and Bacilli. Biodiversity analysis of culturable gut microbiota showed that larval, juvenile, and adult stages have higher diversity than fingerling and yearling stages. Ward's linkage cluster analysis showed that culturable gut microbiota composition in larval and juvenile stages were close to adult stages, whereas fingerling and yearling stage composed same cluster. PCR‐DGGE fingerprinting technique showed that unculturable gut microbiota were constituted by α‐and γ‐Proteobacteria, and Actinobacteria. Ward's linkage cluster analysis showed that unculturable gut microbiota composition in both larval and juvenile stages were distinct from other developmental stages. Our results revealed that gut microbiota composition were varied in different developmental stages of Malaysian Mahseer and continuous shifts of gut microbiota from larval to adult stages.  相似文献

The efficacy of antimicrobial treatment of cotton fabrics depends on various parameters of the coating process, such as the chemical nature and concentration of the antimicrobial agent, the composition of the crosslinking formulation, and the curing temperature. The inclusion complex of triclosan with β-cyclodextrin (βCD) was synthesized and characterized by FTIR, XRD, NMR, Raman, SEM, and TGA. The minimum inhibitory concentration and minimum bactericidal concentration of the complex against Klebsiella pneumoniae and Staphylococcus aureus were compared to those of its precursor. A multifactorial study included an evaluation of the effects of triclosan complexation with β-cyclodextrin, a comparison between the glyoxal and tetracarboxylic acid as crosslinkers, an investigation of the effect of crosslinker and catalyst concentrations, and a comparison of curing at 120°C and 180°C. The cotton was characterized by FTIR-ATR, the micrographs of treated samples were obtained by SEM and the weight add-on was calculated. The bactericidal properties were determined according to AATCC-147. The correlation between the coating process parameters and the antimicrobial efficacy was determined. The optimal combination leading to the highest weight add-on and the antimicrobial coating that was most durable to multiple detergent washes at an elevated temperature was the use of complexed triclosan grafted onto the cotton in the presence of tetracarboxylic acid, followed by curing at 180°C. The curing temperatures were 120°C (P=0.002) and 180°C (P=0.008), catalysts were 1 % and 2 % aluminium sulfate and sodium hypophosphite (P<0.001), and the crosslinkers were 5 % and 10 % glyoxal and butanetetracarboxylic acid (P<0.001); these parameters significantly enhanced the antimicrobial properties of the treated fabrics. The study showed that βCD did not have antimicrobial activity, while the βCD/triclosan-treated textile exhibited potential antimicrobial properties. Overall, the bactericidal activity of fabrics can be enhanced by using βCD/triclosan with 10 % butanetetracarboxylic acid as a cross-linker and 5 % sodium hypophosphite as a catalyst at a curing temperature of 180°C.  相似文献
