Leachates, particularly those from mature landfills, are difficult to treat by biological processes because of their high toxicity and low biodegradability. Therefore, the development of new treatment technology is necessary. The treatment of landfill leachate by peroxicoagulation and solar peroxicoagulation using a batch electrolytic reactor with a Fe cathode and a Cu anode is proposed. The tested operational variables included pH (2.8 and 8.2), current density (11 and 16 mA cm?2), treatment time (5, 10, 15, 20, 25, and 30 min), and presence of solar ultraviolet (UV) light and were collected using a compound parabolic collector. The optimum conditions were a pH, current density, and treatment time of 2.8, 16 mA cm?2, and 10 min, respectively. The presence of UV did not have a significant effect. The chemical oxygen demand and biochemical oxygen demand removed were 62.3% and 55.5%, respectively. The results of UV-visible absorption, fluorescence, and Fourier transform infrared spectroscopy measurements confirm the oxidation process. 相似文献
Antithrombotic activity of brewers’ spent grain peptides before and after simulated gastrointestinal digestion and their effects on blood coagulation pathways were evaluated. Two hydrolysates were produced using sequential enzymatic systems: alkaline protease + Flavourzyme (AF) and neutral protease + Flavourzyme (PF). Simulation of gastrointestinal digestion of AF and PF hydrolysates was made using porcine pepsin and pancreatin enzymes, obtaining the corresponding digested samples: AFD and PFD, respectively. Peptides were fractionated by ultrafiltration using a 1 kDa cut-off membrane. Hydrolysates had peptides with medium and low molecular weights (2100 and 500 Da, respectively), and Glu, Asp, Leu, Ala, and Phe were the most abundant amino acids. Gastrointestinal digested hydrolysates presented high proportion of small peptides (~500 Da), and higher amount of Val, Tyr, and Phe than hydrolysates. Mass spectrum (HDMS Q-TOF) of AFD-ultrafiltered fraction <1 kDa exhibited peptides from 500 to 1000 Da, which are not present in AF. PFD showed the generation of new peptides from 430 to 1070 Da. All samples showed thrombin inhibitory activity. However, no effect was observed on prothrombin time. Peptides <1 kDa from hydrolysates and digested samples delayed thrombin and thromboplastin time respect to the control (~63%). Also the samples showed thrombin inhibitory activity at common pathway level. Thus, brewers’ spent grain peptides exerted their antithrombotic activity by inhibiting the intrinsic and common pathways of blood coagulation. This is the first report to demonstrate that brewers’ spent grain peptides are able to delay clotting time after simulated gastrointestinal digestion. 相似文献

The use of spent coffee grounds (SCG) as organic amendment is a triple solution: a reuse of this bio-residue (6 millions of tons per year), an increase in soil organic carbon (SCG contain 82% of carbohydrates and 13% of proteins) and a decrease in CO2 emissions into the atmosphere. Thus, we investigate the effect of SCG on soil and plants in an in vitro assay. The variables considered were SCG dose (2.5 and 10%), two agricultural soils (Calcisol and Luvisol), and four cultivation times (15, 30, 45 and 60 days). The chemical and physicochemical soil properties, SEM images and growth parameters were analyzed. The highest dose of SCG increased organic carbon, total N and available K and P by 286%, 188%, 45%, and 9%, respectively, while decreasing lettuce growth by 233% compared to control. The SEM study revealed that SCG particles are incorporated into soil aggregates and microorganisms grow over them, which is also corroborated by a 10-times increase of soil respiration rate. The behavior of both soils differs for some SCG effects. In conclusion, SCG have a great potential to increase soil fertility and further investigations are needed in order to improve the use of SCG as amendment. 相似文献

The objectives were i) to assess indicators for potential nitrogen (N) mineralization and ii) to analyze their relationships for predicting winter wheat (Triticum aestivum L.) growth parameters (yield and N uptake, Nup) in Mollisols of the semi-arid and semi-humid region of the Argentine Pampas. Thirty-six farmer fields were sampled at 0–20 cm. Several N mineralization indicators, wheat grain yield and Nup at physiological maturity stage were assessed. A principal component (PC) analysis was performed using correlated factors to grain yield and Nup. The cluster analysis showed two main groups: high fertility and low fertility soils. In high fertility soils, combining PCs in multiple regression models enhanced the wheat yield and Nup prediction significantly with a high R2 (adj R2 = 0.71–0.83). The main factors that explained the wheat parameters were associated with water availability and N mineralization indicator, but they differ according to soil fertility.

Lima bean (Phaseolus lunatus L.) is an important food source in Brazil, especially in the northeast region, where its production and consumption are high. The goals of the present study were to estimate natural outcrossing rates and genetic diversity levels of Lima bean from Brazil, using ten microsatellite loci to obtain information for their conservation and breeding. Fourteen accessions were selected from an experiment in field with open-pollinated and with the presence of pollinating insects. Twelve seeds of each of the 14 selected accessions were grown in screenhouse for tissue harvest and DNA extraction. The multilocus model was used to determine the reproductive system. The outcrossing rate was 38.1 % (tm = 0.381; ts = 0.078), and the results indicated a mixed mating system with a predominance of selfing (1 ? tm = 61.9 %). The biparental inbreeding rate was high (tm ? ts = 0.303) and the multilocus correlated paternity was quite high (rp(m) = 0.889), indicating that the progeny was mostly composed of full sibs. The average effective number of pollen donors per maternal plant (Nep) was low (1.12), and the fixation index for maternal genotypes (Fm) was 0.945, indicating that most genitors resulted from inbreeding. The studied families presented considerable genetic variability: A = 6.10; %P = 30; He = 0.60 and Ho = 0.077. Total diversity was high (HT = 0.596), and a portion was distributed within families (HS = 0.058). In addition, diversity was higher between families (DST = 0.538), and genetic differentiation was high (GST = 0.902). The results presented here can be used in the implementation of Lima bean conservation and breeding programs in Brazil. 相似文献
Lupinus albus seeds contain conglutin gamma (Cγ) protein, which exerts a hypoglycemic effect and positively modifies proteins involved in glucose homeostasis. Cγ could potentially be used to manage patients with impaired glucose metabolism, but there remains a need to evaluate its effects on hepatic glucose production. The present study aimed to analyze G6pc, Fbp1, and Pck1 gene expressions in two experimental animal models of impaired glucose metabolism. We also evaluated hepatic and renal tissue integrity following Cγ treatment. To generate an insulin resistance model, male Wistar rats were provided 30% sucrose solution ad libitum for 20 weeks. To generate a type 2 diabetes model (STZ), five-day-old rats were intraperitoneally injected with streptozotocin (150 mg/kg). Each animal model was randomized into three subgroups that received the following oral treatments daily for one week: 0.9% w/v NaCl (vehicle; IR-Ctrl and STZ-Ctrl); metformin 300 mg/kg (IR-Met and STZ-Met); and Cγ 150 mg/kg (IR-Cγ and STZ-Cγ). Biochemical parameters were assessed pre- and post-treatment using colorimetric or enzymatic methods. We also performed histological analysis of hepatic and renal tissue. G6pc, Fbp1, and Pck1 gene expressions were quantified using real-time PCR. No histological changes were observed in any group. Post-treatment G6pc gene expression was decreased in the IR-Cγ and STZ-Cγ groups. Post-treatment Fbp1 and Pck1 gene expressions were reduced in the IR-Cγ group but increased in STZ-Cγ animals. Overall, these findings suggest that Cγ is involved in reducing hepatic glucose production, mainly through G6pc inhibition in impaired glucose metabolism disorders. 相似文献
The palm ruff, Seriolella violacea (Cojinoba), is a potential new species for Chilean aquaculture. To approach Cojinoba larviculture, an experimental Artemia enrichment emulsion, containing docosahexaenoic acid (DHA)/eicosapentaenoic acid (EPA) = 2.5, supplemented with vitamin E, astaxanthin, and β‐glucan, was evaluated in both Artemia and Cojinoba larvae, 30–50 d.a.h. This study tested an experimental enrichment emulsion versus a commercial emulsion, with an integral approach of multicompound emulsions. After 23 h enrichment, experimental emulsion (EE)‐enriched nauplii reached DHA and EPA concentrations of 23.8 and 18.7 mg/g dry weight (dwt), respectively, while in Cojinoba larvae they were 18.4 and 19.7 mg/g dwt. Control emulsion (CE)‐enriched nauplii exhibited lower DHA and EPA (6.1 and 7.7 mg/g dwt), while only DHA decreased in the control larvae (12.6 mg/g dwt). Vitamin E was higher in EE‐enriched nauplii (29.2 mg/100 g dwt) than in the control (8.4 mg/100 g dwt). Larvae fed EE‐enriched Artemia exhibited 8% increase in survival and 19% in growth compared with the control. Astaxanthin was detected only in larvae fed EE‐enriched nauplii. The tumor necrosis factor‐α concentration was not significantly different between larvae fed EE‐ and CE‐enriched nauplii. EE looks promising as an Artemia enrichment and experimental diet to assess palm ruff larval requirements, and has a positive impact on fish larvae performance. 相似文献
