This review on current biotechnological methods in forestry for in vitro tissue cultures to define the effect of stress conditions on trees,concentrates on somatic embryogenesis.Callus tissue,the key product of somatic embryogenesis,grows over a tree wound under ex vitro conditions.Callus tissue can be used in research in areas such as pathogenic susceptibility at the embryonic level,effect of heavy metals,influence of low temperatures(cryopreservation),production of secondary metabolites and transformation of plants.Callus of arborescent plants can be induced in vitro by fungal elicitors to produce secondary metabolites for pharmaceutical and cosmetic industries and are strongly repellant to herbivores and can thus act to protect forests.Analyses of dual cultures demonstrated that callus tissue exposed to a pathogenic fungus responds by synthesizing low-molecular-mass proteins belonging to an immune protein class.Cryopreservation of embryonic callus tissue also has broad applications,e.g.,for valuable plant genotypes in gene banks.Without strategies to protect forests against stressfactors,forest ecosystems will degrade to the detriment of all life,including humans.In vitro biotechnological research using callus tissue contributes to progress in forestry and the disciplines of ecology,physiology,phytopathology,culture and selection of plants.  相似文献

Pseudodidymella fagi, described from infected leaves of Fagus crenata in Japan, has recently been spreading in Europe, where it causes leaf blotch of European beech Fagus sylvatica. In the years 2008–2016, an outbreak of P. fagi was observed on European beech in Switzerland, Germany and Austria, and it is reported here for the first time from Slovakia where it was detected in 2017. The identification of the pathogen is supported by the morphological analyses of material observed in vivo and in vitro and molecular analyses based on ITS sequences. Detailed morphological characterization of cultures based on European material, on F. sylvatica, is provided here for the first time.  相似文献

The aim of the study was to determine the possibility of experimental media (agglomerate elastomers EPP) application as biological media bed, which serves the purpose of water purification in recirculating aquaculture systems (RAS). RAS enables mass‐production of fish in small volume of water in a limited area. This involves the possibility of multiple usage of water during culture. However, for that purpose of maintaining proper physico‐chemical parameters, water purification from products of metabolism, especially toxic nitrogen compounds, is required. One of the simplest and most effective ways to achieve it is combining application of two types of water filtration: mechanical and biological. It is needed to study new media for biological bed with proper filling is able to purify water from toxic nitrogen compounds.  相似文献

Key message

Quercus robur seedling mass was affected more by planting density than by taproot pruning. Root pruning enhanced stem biomass at the expense of roots in later growth stages. Alteration of biomass allocation due to nursery practices may result in greater susceptibility to injury and death of the seedlings under unfavorable environmental conditions.

Context

Plants adjust their growth and modulate the resource allocation in response to applied treatments and environmental conditions.

Aims

The aim was to examine how taproot pruning in seedlings grown at different densities affected long-term growth of Quercus robur.

Methods

Seedlings, sown as acorns at two planting densities, with or without pruned roots were harvested in the second, fourth, and fifth years of growth. The effect of root pruning on biomass allocation was determined by measuring leaf, stem, and root mass fractions; carbohydrate concentrations in the roots; and C/N ratios. Specific leaf area and root length were also determined to assess morphological adaptations to growth conditions.

Results

Total seedling mass was affected more by planting density than by taproot pruning. After 4 years of growth, root mass fractions were lower and stem mass fractions were greater in seedlings planted at a higher density. Five-year old root-pruned seedlings also had a lower root mass fraction and higher stem mass fractions than unpruned seedlings. Specific root length was not affected by root pruning or planting density.

Conclusion

Decrease of relative root biomass with simultaneous increase of stem biomass may be a long-term consequence of taproot pruning of Q. robur, and the effects may manifest years after the seedling stage.

Purpose

Soils that develop on the dumps in historical arsenic mining sites contain high concentrations of As thus constituting a serious environmental risk. This study was aimed to examine the changes in arsenic solubility in mine soils as induced by organic matter introduced with forest litter.

Materials and methods

Four large samples of initially developed soils were collected from the dumps remaining in former mining sites and were incubated for 90 days at various moistures: 80% of maximum water holding capacity and 100% (flooded conditions), with and without addition of beech forest litter (BL), 50 g/kg. Soils contained up to 5.0% As. Soil pore water was collected periodically with MacroRhizon suction samplers and examined on As, Mn, and Fe concentrations, pH, Eh, and dissolved organic carbon (DOC). The properties of dissolved organic matter were characterized by UV-VIS spectroscopic parameters A4/A6 and SUVA254.

Results and discussion

Application of BL resulted in an intensive release of As from soils, particularly at 100% moisture. As concentrations in soil pore water increased strongly during the first 2 or 4 weeks of incubation and then started to decrease in all cases, except for one flooded soil. As was released particularly intensively from carbonate-containing soils. The mechanisms of As mobilization, including reductive dissolution of Mn and Fe oxides and the competition with DOC for sorption sites on the oxides, were discussed as related to soil properties. Pore water concentrations of DOC were increasing at the beginning of incubation and started to decrease after two or four weeks. Spectroscopic parameters of dissolved organic matter in ZS soils indicated increasing aromaticity and progress of humification.

Conclusions

Forest litter introduced to mine dump soils causes a mobilization of As into soil pore water. This effect, particularly strong in carbonate-rich soils, is apparently related to high concentrations of DOC and usually declines with time, which may be explained by the progress in humification. The relationships between DOC properties and As speciation in soil pore water should be dissected for better interpretation of experimental results.

Bovine tuberculosis is a contagious and zoonotic disease of animals and humans. In Europe, the number of reported cases of tuberculosis has decreased. Equidae are relatively rarely infected even in endemic areas. The presented report describes a case of chronic Mycobacterium bovis tuberculosis in a 30-year-old female donkey. The donkey initially presented with persistent lymphadenopathy; however, as the disease progressed, weight loss became apparent. To the authors' knowledge, this is the second confirmed case of tuberculosis in a donkey in Europe.  相似文献

Forest transformation from coniferous monocultures to mixed stands is being promoted worldwide, including the introduction of fast-growing broadleaved tree species within native stands. Here, we studied how enrichment of temperate European Scots pine (Pinus sylvestris) forest by North-American northern red oak Quercus rubra impacted macronutrient concentrations in two long-lived and dominant components of the forest understory: bilberry Vaccinium myrtillus and lingonberry V. vitis-idaea. Study sites were located in forest complexes (central Poland) which occupy continuously reforested lands (hereafter ancient forests) as well as post-agricultural lands (recent forests), all suitable for mesic pine forests. Samples of bilberry and lingonberry leaves, stems, and fruits were collected in pine stands and in adjacent Scots pine-red oak stands, in both ancient and recent forests. Concentrations of macronutrients (C, N, P, K, Ca, S, and Mg) in aboveground biomass components were analysed using standardized chemical procedures. The study revealed intra- and interspecific (bilberry vs. lingonberry) differences in concentrations of all nutrients in leaves, stems, and fruits, except for invariable C concentrations. Macronutrient accumulations in plants were decreased by land-use discontinuity and favoured by enrichment of tree stands by Q. rubra. The estimated macronutrient pools were much higher for V. myrtillus than V. vitis-idaea in all forest types studied. They were lower in forests enriched with Q. rubra, both ancient (up to 25.5% for bilberry and 99.9% for lingonberry) and recent (46.9% and 99.9%, respectively), as well as in recent pine forest (46.6% and 81.1%, respectively) than in ancient pine forest. Higher K and S pools (39.3% and 6.5%, respectively) noted for bilberry in an ancient forest with Q. rubra were exceptions. Despite more effective accumulations of elements at the species level, macronutrient pools of Vaccinium myrtillus and V. vitis-idaea decreased significantly in the presence of introduced Q. rubra due to negative impacts of this broadleaved tree on bilberry and lingonberry cover and biomass. Therefore, the limitation of alien Q. rubra planting in sites of mesic pine forest with the abundant occurrence of V. myrtillus and/or V. vitis-idaea is recommended.
