The aim of this study was to find the biochemical markers of blackleg, evoked by the fungus Leptosphaeria maculans in seedlings of oilseed winter rape. Analyses of antioxidant enzymes activity and reactive oxygen species detection were performed 24, 48 and 72 h after inoculation. Moreover, measurements of the total pool of phenolic compounds, trans-cinnamic acid and ferulic acid were carried out. The results proved the virulence of Ph L5 isolate towards the rape seedlings. An increase in total phenolic content, including trans-cinnamic acid, was observed 24 h after inoculation. High activity of phenylalanine ammonia-lyase is probably another symptom of the defense mechanisms, despite the fact that it was not accompanied by elevated phenolics level, which were probably rapidly exhausted during Phoma lingam inoculation. A drop in phenolics content was balanced by higher activity of SOD, CAT and non-specific peroxidases. Enhanced activity of these enzymes was a response to the reactive oxygen species overproduction. ROS accumulation in indirectly inoculated first true leaves after inoculation may suggest a systemic defense response.  相似文献

Scots pine (Pinus sylvestris L.) and European beech (Fagus sylvatica L.) dominate many of the European forest stands. Also, mixtures of European beech and Scots pine more or less occur over all European countries, but have been scarcely investigated. The area occupied by each species is of high relevance, especially for growth evaluation and comparison of different species in mixed and monospecific stands. Thus, we studied different methods to describe species proportions and their definition as proportion by area. 25 triplets consisting of mixed and monospecific stands were established across Europe ranging from Lithuania to Spain in northern to southern direction and from Bulgaria to Belgium in eastern to western direction. On stand level, the conclusive method for estimating the species proportion as a fraction of the stand area relates the observed density (tree number or basal area) to its potential. This stand-level estimation makes use of the potential from comparable neighboring monospecific stands or from maximum density lines derived from other data, e.g. forest inventories or permanent observations plots. At tree level, the fraction of the stand area occupied by a species can be derived from the proportions of their crown projection area or of their leaf area. The estimates of the potentials obtained from neighboring monospecific stands, especially in older stands, were poorer than those from the maximum density line depending on the Martonne aridity index. Therefore, the stand-level method in combination with the Martonne aridity index for potential densities can be highly recommended. The species’ proportions estimated with this method are best approximated by the proportions of the species’ leaf areas. In forest practice, the most commonly applied method is an ocular estimation of the proportions by crown projection area. Even though the proportions of pine were calculated here by measuring crown projection areas in the field, we found this method to underestimate the proportion by 25% compared to the stand-level approach.  相似文献

A series of per-O-methylated flavonoid di- and tri-glycosides, linked with 1-2 and/or 1-6 glycosidic bonds between sugar rings that were isolated from different plant materials were analyzed. It was demonstrated that the fragmentation behavior of permethylated flavonoid glycosides is dependent on the glycosidic bond placement between sugars. Y(n) type fragment ions, created after glycosidic bond cleavage with oxygen retention on sugar at the reducing end for permethylated compounds, were observed in the normal and linked-scan mass spectra recorded for alpha(1-2) bonded conjugates of flavonoid di- and tri-glycosides. Moreover, for alpha (1-6) linked glycosides, Y fragments created after rearrangement and elimination of internal sugar residues were observed in addition to Y(n) type ions, but these fragment ions were not registered in normal desorption ionization spectra. This second type of fragmentation was also reported previously in collision-induced dissociation tandem mass spectrometry (CID MS/MS) spectra of some oligosaccharides and flavonoid glycosides, but their presence was independent of the glycosidic bonds placement between sugar rings.  相似文献

Relationship between somaclonal variation and type of culture in cucumber   总被引:3,自引:0,他引:3  
Highly inbred B line of cucumber was used to compare the effect of four types of in vitro culture on somaclonal variation. The plants were regenerated from the following types of culture: twelve- and eighteen-month-old liquid culture of meristematic clumps (LMC12(18)), ten-month-old embryogenic cytokinin-dependent suspension (CDS), eighteen-month-old embryogenic cytokinin-dependent suspension in medium with modified NH+ 4/NO3 - ratio (CDS 1.7), twelve-month-old embryogenic auxin-dependent suspension (ADS), thirty six-month-old embryogenic auxin-dependent suspension in medium with modified NH+ 4/NO3 - ratio (ADS 1.7) and recurrent leaf callus regeneration (RLC) – repeated 5 times. The differences in the incidence of the following properties were observed: the ploidy of R0 plants, the segregation of new morphological traits in R1 and the germination ability of R1 seeds. R1 families with the segregation of new phenotypes were most numerous in CDS (62.5%) and LMC18 (57.9%), next in CDS1.7 (35.7%), while the smallest number was found in LMC12 (11.1%) and RLC (3.4%).Tetraploid and mixoploid plants occurred in ADS1.7 and ADS (100%) whereas CDS and RLC were observed to contain only tetraploids, respectively 33.3% and 55.2%. There were no changes of ploidy after LMC12, LMC18 and CDS1.7. Among new phenotypes there were such that have not been described so far in cucumber: ginkgolike leaf (gll), yellow-green chlorophyll mutants (y-gc), serrate margin of corolla in male and female flowers (smc). Leakage of jet fuel from an oil skimmer has resulted in soil and groundwater contamination at the fire fighting training site of the Oslo international airport. The site is located on an unconfined sandy aquifer formed by the Gardermoen ice contact delta. A strong relationship between hydrogeological settings and spatial distribution of the hydrocarbons has been observed. Leaching hydrocarbons constitute a source of dissolved BTEX and naphthalene. Concentration of the dissolved hydrocarbons, electron acceptors and metabolic by-products has revealed concomitant changes in space and in time, in response to groundwater fluctuations. Correlation between high concentrations of hydrocarbons, elevated alkalinity and depleted electron acceptors supports the hypothesis that intrinsic bioremediation has been taking place at the site. Naphthalene, however, has been found persistent under anoxic conditions. Calculations of biodegradation potential indicates that Fe(III) reduction, methanogenesis, nitrate and sulphate reduction are the main factors controlling biodegradation.  相似文献
