During the last decade, Piscine orthoreovirus was identified as the main causative agent of heart and skeletal muscle inflammation (HSMI) in Atlantic Salmon, Norway. A recent study showed that PRV-1 sequences from salmonid collected in North Atlantic Pacific Coast (NAPC) grouped separately from the Norwegian sequences found in Atlantic Salmon diagnosed with HSMI. Currently, the routine assay used to screen for PRV-1 in NAPC water and worldwide cannot differentiate between the two groups of PRV-1. Therefore, this study aimed at developing a real-time polymerase chain reaction (RT-qPCR) assay to target the PRV-1 genome segments specific for variants associated with HSMI. The assay was optimized and tested against 71 tissue samples collected from different regions including Norway, Chile and both coast of Canada and different hosts farmed Atlantic Salmon, wild Coho Salmon and escaped Atlantic Salmon collected in British Columbia, West Coast of Canada. This assay has the potential to be used for screening salmonids and non-salmonids that may carry PRV-1 potentially causing HSMI.  相似文献

Reconciling food security, economic development and biodiversity conservation is a key challenge, especially in the face of the demographic transition characterizing many countries in the world. Fisheries and marine ecosystems constitute a difficult application of this bio‐economic challenge. Many experts and scientists advocate an ecosystem approach to manage marine socio‐ecosystems for their sustainability and resilience. However, the ways by which to operationalize ecosystem‐based fisheries management (EBFM) remain poorly specified. We propose a specific methodological framework—viability modelling—to do so. We show how viability modelling can be applied using four contrasted case‐studies: two small‐scale fisheries in South America and Pacific and two larger‐scale fisheries in Europe and Australia. The four fisheries are analysed using the same modelling framework, structured around a set of common methods, indicators and scenarios. The calibrated models are dynamic, multispecies and multifleet and account for various sources of uncertainty. A multicriteria evaluation is used to assess the scenarios’ outcomes over a long time horizon with different constraints based on ecological, social and economic reference points. Results show to what extent the bio‐economic and ecosystem risks associated with the adoption of status quo strategies are relatively high and challenge the implementation of EBFM. In contrast, strategies called ecoviability or co‐viability strategies, that aim at satisfying the viability constraints, reduce significantly these ecological and economic risks and promote EBFM. The gains associated with those ecoviability strategies, however, decrease with the intensity of regulations imposed on these fisheries.  相似文献

Intensive vegetable production in greenhouses has rapidly expanded in China since the 1990s and increased to 1.3 million ha of farmland by 2016, which is the highest in the world. We conducted an 11‐year greenhouse vegetable production experiment from 2002 to 2013 to observe soil organic carbon (SOC) dynamics under three management systems, i.e., conventional (CON), integrated (ING), and intensive organic (ORG) farming. Soil samples (0–20 and 20–40 cm depth) were collected in 2002 and 2013 and separated into four particle‐size fractions, i.e., coarse sand (> 250 µm), fine sand (250–53 µm), silt (53–2 µm), and clay (< 2 µm). The SOC contents and δ13C values of the whole soil and the four particle‐size fractions were analyzed. After 11 years of vegetable farming, ORG and ING significantly increased SOC stocks (0–20 cm) by 4008 ± 36.6 and 2880 ± 365 kg C ha?1 y?1, respectively, 8.1‐ and 5.8‐times that of CON (494 ± 42.6 kg C ha?1 y?1). The SOC stock increase in ORG at 20–40 cm depth was 245 ± 66.4 kg C ha?1 y?1, significantly higher than in ING (66 ± 13.4 kg C ha?1 y?1) and CON (109 ± 44.8 kg C ha?1 y?1). Analyses of 13C revealed a significant increase in newly produced SOC in both soil layers in ORG. However, the carbon conversion efficiency (CE: increased organic carbon in soil divided by organic carbon input) was lower in ORG (14.4%–21.7%) than in ING (18.2%–27.4%). Among the four particle‐sizes in the 0–20 cm layer, the silt fraction exhibited the largest proportion of increase in SOC content (57.8% and 55.4% of the SOC increase in ORG and ING, respectively). A similar trend was detected in the 20–40 cm soil layer. Over all, intensive organic (ORG) vegetable production increases soil organic carbon but with a lower carbon conversion efficiency than integrated (ING) management.  相似文献

Savanna woodlands in Sudan host great biodiversity, provide a plethora of ecosystem goods and services to local communities, and sustain numerous ecological functions. Although the importance of the Acacia trees in these areas is well known, up-to-date information about these woodlands' diversity is limited and changes in their woody vegetation composition, density, diversity and relative frequency are not monitored over time. This study explored tree diversity and stand stage structure in Nuara Reserved Forest, a typical savanna woodland ecosystem in southeastern Sudan. A total of 638 circular sample plots (1000 m2 for each) were established using a systematic sampling grid method. The distance between plots was 200 m. In each plot, all living trees with diameter at breast height (DBH) ≥5.00 cm were identified and counted, and their DBH values were recorded. From these data, tree composition, diversity, density and stage structure were assessed. There were 12,259 individual trees representing four species (Acacia seyal, Balanites aegyptiaca, Acacia Senegal and Acacia mellifera) that belong to two families. The dominant species was Acacia seyal. Average tree density was 191 trees/hm2 and the Shannon-Weiner index for trees diversity was 0.204. Overall, young trees comprised 86.30% of the forest. The state of tree richness and density in the study area was low compared to other similar environments in the region and around the world. We recommended adoption of a proper management system that includes monitoring of woody vegetation diversity in this forest, and management actions to enhance tree diversity and sustain ecosystem services to local communities. In addition to care for the dominant Acacia seyal stands, more attention and conservation should be devoted to reestablishing Acacia senegal and Acacia mellifera trees because of their high ecological and economic values for local communities.  相似文献

Sustainability of traditionally cultivated rice in the rice-wheat cropping zone (RWCZ) of Pakistan is dwindling due to the high cost of production, declining water resources and escalating labour availability. Thus, farmers and researchers are compelled to find promising alternatives to traditional transplanted rice (TPR). A field study was conducted in Punjab, Pakistan, in 2017 and 2018 to explore the trade-offs between water saving and paddy yield, water productivity and economics of two aromatic rice varieties under dry direct seeded rice (DDSR) and TPR. The experiment was comprised of three irrigation regimes on the basis of soil moisture tension (SMT) viz., continuous flooded (>−10 kPa SMT), alternate wetting and drying (AWD) (−20 kPa SMT) and aerobic rice (−40 kPa SMT), maintained under TPR and DDSR systems. Two aromatic rice verities: Basmati-515 and Chenab Basmati-2016 were used during both years of study. In both years, DDSR produced higher yields (13–18%) and reduced the total water inputs (8–12%) in comparison to TPR. In comparison to traditional continuous flooded (CF), AWD under DDSR reduced total water input by 27–29% and improved the leaf area index (LAI), tillering, yield (7–9%), and water productivity (44–50%). The performance of AWD with regard to water savings and increased productivity was much higher in DDSR system as compared to AWD in TPR system. Cultivation of DDSR with aerobic irrigation improved water savings (49–55%) and water productivity (22–30%) at the expense of paddy yield reduction (36–39%) and spikelet sterility. With regard to variety, the highest paddy yield (6.6 and 6.7 t ha−1) was recorded in DDSR using Chenab Basmati-2016 under AWD irrigation threshold that attributed to high tiller density and LAI. The economic analysis showed DDSR as more beneficial rice establishment method than TPR with a high benefit-cost ratio (BCR) when the crop was irrigated with AWD irrigation threshold. Our results highlighted that with the use of short duration varieties, DDSR cultivation in conjunction with AWD irrigation can be more beneficial for higher productivity and crop yield.  相似文献

The present investigation on chemical constituents of the soft coral Sarcophyton cherbonnieri resulted in the isolation of seven new cembranoids, cherbonolides F–L (1–7). The chemical structures of 1–7 were determined by spectroscopic methods, including infrared, one- and two-dimensional (1D and 2D) NMR (COSY, HSQC, HMBC, and NOESY), MS experiments, and a chemical reduction of hydroperoxide by triphenylphosphine. The anti-inflammatory activities of 1–7 against neutrophil proinflammatory responses were evaluated by measuring their inhibitory ability toward N-formyl-methionyl-leucyl-phenylalanine/cytochalasin B (fMLF/CB)-induced superoxide anion generation and elastase release in primary human neutrophils. The results showed that all isolates exhibited moderate activities, while cherbonolide G (2) and cherbonolide H (3) displayed a more active effect than others on the inhibition of elastase release (48.2% ± 6.2%) and superoxide anion generation (44.5% ± 4.6%) at 30 µM, respectively.  相似文献

Six diets were formulated to investigate the success of fish meal (FM) replacement by plant proteins; diet 1 reflected a commercial feed (8% FM), diet 3 contained 4% FM, and diet 5 was devoid of FM. Whereas, diet 2, diet 4, and diet 6 reflected diet 1, diet 3, diet 5, respectively, and supplemented with essential amino acid (EAA). At the end of 8‐week trial, there was no significant difference in survival rate. Significantly higher final weight, weight gain rate, and specific growth rate were recorded in the group fed diet 2 compared with the other treatments (except diet 4) (p < 0.05). Feed conversion ratio of fish fed diet 2 was significantly lower than those fed diets 1, 3, and 5 (p < 0.05). The lowest feed intake and highest protein efficiency rate were found in fish fed diet 2 (p < 0.05). There was no significant difference in whole body compositions between treatments. Plasma aspartate transaminases, alanine aminotransferase, and glucose were significantly affected by dietary treatments (p < 0.05), while plasma protein and albumin contents were not influenced by the treatments. The relative expression of target of rapamycin (TOR) and phosphatidylinositol 3‐kinase, regulatory subunit 1 (alpha) (PIK3R1) in fish fed diet 3 (4% FM) were significantly down‐regulated compared with those fed diet 6 for TOR and diets 4 and 6 for PIK3R1 (p < 0.05). Insulin receptor substrate 1 (IRS‐1) and janus kinase 3 (JAK3) expressions were fluctuated, with the higher levels in fish fed diets 4 and 6. In conclusion, the findings of this study indicate that plant protein mixture supplemented with EAA could be used to substitute FM in practical diet for Megalobrama amblycephala.  相似文献

A few studies have illustrated the effects of sodium salt derived from alginic acid on different fish species. However, little is known about the effect of sodium alginate on catfish (Clarias gariepinus). Therefore, this study was performed to assess the use of low molecular weight sodium alginate ( LMWSA) in C. gariepinus. A total of 180 apparently healthy C. gariepinus with a mean body weight of 45 g were randomly divided into three equal groups (D1, D2 and D3). D1 the control group received a control diet, while D2 and D3 received 1% and 3% LMWSA, respectively, for 8 weeks. A challenge test against Aeromonas hydrophila was performed on 15 randomly selected catfish for 15 days. At the end of the experiment, catfish in D3 that received a diet of 3% LMWSA showed significant increases in the final body weight, weight gain and thermal‐unit growth coefficient compared with those in D2 and D1. There was a significant decrease in the erythrogram in D1 after the 4‐day pathogen challenge. A leucogram revealed leucocytosis, heterophilia and lymphocytosis in catfish in D2 and D3 compared with those in D1. After the 4‐day challenge, the following changes took place: lysozyme, nitric oxide, phagocytic activity and the respiratory burst were significantly elevated in catfish that received LMWSA and were more pronounced in D3 than in D1. The mortalities of catfish have been stopped after pathogen challenge from 8‐day in D1 and D2 where at 6‐day in D3. Thus, administration of 1% and 3% LMWSA enhances the growth, immune response and resistance of C. gariepinus against A. hydrophila.  相似文献
