A rapid and low cost procedure, the carbon immunoassay (CIA) test, was evaluated for the diagnosis of Toxoplasma gondii infections. Using a closely related parasite (Besnoitia jellisoni) as antigen, and homologous or heterologous immune sera, it was demonstrated by light and electron microscopy that CIA is a very reliable and specific test. As it is neither expensive nor time-consuming, it can be recommended for general and routine laboratory use. 相似文献

The presence of a begomovirus in potato plants with yellow mottle symptoms was determined for the first time in Cuba. The
incidence of typical begomovirus-like symptoms in potato plants in some regions of Havana province (Güira de Melena, San José
de las Lajas, Güines and Boyeros) during the growing seasons from 1992 to 1998 was in general low. However, in some cultivars
belonging to the National Program for Potato Genetic Improvement, the incidence reached 100%. Yield losses, determined in
1992 and 1994, ranged as high as 19% to 56.33% depending on the cultivar. Characterization of the causal agent was done by
light microscopy, host range (graft and mechanical transmission), DNA hybridizations, polymerase chain reaction, and restriction
fragment length polymorphism analysis. Nucleotide sequence of the amplified fragments revealed the presence ofTomato mottle Taino virus. The virus was transmittedvia tubers and has been detected in mixed infections withPotato virus X and withPotato leaf roll virus.

During the months of April to August in 1999 and 2002, oral swabs were collected from 146 tortoises (Testudo sp.) in private collections in the United Kingdom and tested by polymerase chain reaction (PCR) for the presence of Mycoplasma agassizii and Chelonian herpesvirus (ChHV). The presence of M. agassizii was confirmed by restriction digestion of the PCR product. A 307-bp fragment of the ChHV UL5 homologue gene was sequenced and found to show most similarity to equine herpesvirus type 1. A prevalence of 15.8 and 8.2% was found for M. agassizii and ChHV, respectively. Comparison of the carriage of both M. agassizii and ChHV in different species of tortoises correlated the presence of M. agassizii with Testudo horsfieldii and ChHV with Testudo marginata and Testudo graeca iberia. An association of ChHV with stomatitis was also found. Mixed infections with both agents were detected. The findings further demonstrate this pathogen-tortoise association and the cross transmission of these infections if different tortoise species are housed together. 相似文献

Urinary tract diseases are among the main reasons for consultation in veterinary clinics and hospitals. It affects animals of any age, breed and gender. Among the diseases that affect this system, urolithiasis is the second largest cause of clinical signs compatible with feline urinary tract disease. The term urolithiasis refers to the presence of uroliths in any region of the urinary tract, but it is more commonly seen in the bladder and urethra. Uroliths are classified based on the type of mineral present in their composition, therefore, quantitative and qualitative analyzes are important for a better therapeutic approach. The animals may suffer from the disease and be asymptomatic, or show nonspecific clinical signs, making the diagnosis difficult. The disease should not be seen as a single problem, but as a consequence of various disorders. As dietary, metabolic, genetic and infectious causes, as well as factors that potentiate the chance of development of uroliths such as breed, age, sex, age range, obesity, sedentary lifestyle, geographic region and climate. Thus, the knowledge of the factors that influence the formation of uroliths, as well as the understanding of the pathophysiology, are key elements for better alternatives of therapy and prevention. The recognition of these factors helps to identify susceptible populations, minimizing exposure and increasing the protection factors, which facilitates the diagnosis and treatment of patients with urolithiasis. Pepper huasteco yellow vein virus (PHYVV) is the main virus of pepper crop in Mexico. No resistant cultivars are available and resistance breeding is hampered by the lack of knowledge of heritability (h 2) of PHYVV resistance. This is a continuation of previous studies and the objectives were to analyze the h 2 and the behavior of the resistant trait to PHYVV. Four resistant assays were done with three resistant wild lines (UAS12, UAS13 and UAS10) of Capsicum annuum in the S4, S5, S6 and S7 generation under greenhouse conditions. Plants from all tests were inoculated with PHYVV through Bemisia tabaci. Line UAS12 was the most resistant showing a significantly proportion of resistant plants, less disease symptoms and longer incubation time, followed by the lines UAS13 and UAS10 in all assays. Distribution of symptoms showed a bimodal tendency in all the trials, suggesting that two groups of genes are involved in this resistance trait. The lines UAS12, UAS13 and UAS10 showed the same pattern of response to selection with an average of h 2 of 0.17, 0.06, 0.02 and 0.00 in the S4, S5, S6 and S7, respectively. These results indicate that all lines responded positively to the selection in the S4, S5 and S6, whereas in the S7 there was no response by the possible exhaustion of variation. Line UAS12 is the most promising genotype and the lines UAS13 and UAS10 are genetic resources that can be supplemented to breed the resistance of PHYVV. Quinoa protein concentrate (QPC) was extracted and digested under in vitro gastrointestinal conditions. The protein content of QPC was in the range between 52.40 and 65.01% depending on the assay used. Quinoa proteins were almost completely hydrolyzed by pepsin at pH of 1.2, 2.0, and 3.2. At high pH, only partial hydrolysis was observed. During the duodenal phase, no intact proteins were visible, indicating their susceptibility to the in vitro simulated digestive conditions. Zebrafish larvae model was used to evaluate the in vivo ability of gastrointestinal digests to inhibit lipid peroxidation. Gastric digestion at pH 1.2 showed the highest lipid peroxidation inhibition percentage (75.15%). The lipid peroxidation activity increased after the duodenal phase. The digest obtained at the end of the digestive process showed an inhibition percentage of 82.10%, comparable to that showed when using BHT as positive control (87.13%). 相似文献

The effects of water and salt stress on rate of germination and seedling growth were investigated under laboratory conditions in 46 soya bean genotypes from Central-West region of Brazil to verify how these stresses may limit crop establishment during the initial growth stage and also to identify the most tolerant genotypes to drought and salinity. Mild water and salt stresses were imposed by seed exposure to –0.20 MPa iso-osmotic solutions with polyethylene glycol—PEG 6000 (119.57 g/L) or NaCl (2.357 g/L) for 12 days at 25°C. The germination percentage, seedling length and seedling dry matter were measured, and then, salt or drought tolerance indexes were calculated. The “NS 5909 RG,” “NS 7000 IPRO,” “NS 7338IPRO,” “FPS Solimões RR,” “NS 5151 IPRO,” “SYN 13610 IPRO,” “LG 60177 IPRO,” “NS 6909 IPRO” and “BMX Desafio RR” were identified as the most drought-tolerant genotypes, whereas under salinity conditions, the genotypes “5D 615 RR,” “BMX Desafio RR,” “5D 6215 IPRO” and “BMX Ponta IPRO” were identified as tolerant. The objective of this study was to evaluate the effect of collagenase and two final dimethylformamide (DMF) concentrations (4% and 7%) on alpaca frozen-thawed sperm quality. A total of 25 ejaculates from 5 alpaca were obtained using electroejaculation. Each individual ejaculate was evaluated and then diluted 4:1 in a solution of 1 mg/ml collagenase in HEPES-TALP medium and incubated for 4 min at 37°C. Subsequently, samples were diluted in TRIS-fructose-citric acid-egg yolk and cooled to 5°C. Then, each sample was divided in two aliquots and DMF at final concentration of 4% or 7% was added, equilibrated for 1 hr at 5°C and frozen over liquid nitrogen vapours. A Kruskal–Wallis test was used to evaluate the sperm morphometry, and Completely Random Block designs were used to analyse sperm motility, viability, membrane function and acrosome status. After collagenase incubation, none of the samples showed thread formation, and sperm parameters were preserved. Non-progressive motile sperm were higher (p < .05) in equilibrated samples (4% DMF: 31.8 ± 8.3% and 7% DMF: 36.3 ± 11.8%) compared to raw (10.1 ± 4.3%) and frozen-thawed semen (4% DMF: 9.7 ± 1.8% and 7% DMF: 7.5 ± 3.2%). Sperm membrane function, membrane integrity and intact acrosomes were higher (p < .05) in raw semen (40.1 ± 12.2%, 94.6 ± 3.2% and 91.3 ± 8.1%) compared to frozen-thawed samples (4% DMF: 19.8 ± 4.7%, 53.2 ± 2.7%, 65.7 ± 8.7% and 7% DMF: 20.4 ± 4.5%, 54.1 ± 1.4%, 64.6 ± 9.1%). Length of the sperm head was lower in frozen-thawed samples, being statistically different with 4% DMF compared to pre-freezing samples. The ratio between acrosome and head areas was greater (p < .05) in frozen-thawed samples. Incubation of raw alpaca semen with collagenase decreased the thread formation without affecting sperm quality. Frozen of collagenase treated alpaca semen with 4% or 7% DMF did not preserve the sperm parameters in thawed samples. 相似文献
