A selected group of pharmaceutical compounds were evaluated for the ability to inhibit the biochemical activity of fibrinoligase (coagulation factor XIIIa*) in pooled equine plasma. Criteria for the pharmaceuticals selected were based on the mechanism of the transglutamination biochemical reaction mediated by coagulation factor XIIa*. These criteria were complemented by recognition of the molecular configuration and chemical composition of amino acid residue side chains involved in the process of covalent fibrin monomer polymerization (cross-linking, transglutamination) mediated by this enzyme. Each pharmaceutical was evaluated individually and in combination with other potential coagulation factor XIIIa* inhibitors in an effort to detect additive and synergistic phenomenon. In this context, pharmaceuticals with a carbonylamide (eg, cefuroxime, Girard's reagent-P, prolinamide) were applied in concert with compounds with a terminal amine (eg, D-arginine, L-lysine) functional group. In concept, this method theoretically served to competitively simulate glutamine and lysine amino acid residues within strands of fibrin monomer substrate involved in phase I (carbonylamide) and phase II (terminal amine) of the transglutamination reaction (covalent fibrin monomer cross-linking). Halogen-dinitro and ethylene compounds were also evaluated because of their reported ability to inactivate enzyme systems dependent on an intact sulfhydryl group located at their biochemically active site (eg, cystine amino acid residue). Our lack of understanding of the immune microenvironment in canine osteosarcoma (cOSA) has limited the identification of potential immunotherapeutic targets. In particular, our ability to utilize readily available tissue from a dog's primary tumour to predict the type and extent of immune response in their pulmonary metastatic lesions is unknown. We, therefore, collected 21 matched pairs of primary tumours and pulmonary metastatic lesions from dogs with OSA and performed immunohistochemistry to quantify T‐lymphocyte (CD3), FOXP3+ cell, B‐lymphocyte (Pax‐5), and CD204+ macrophage infiltration. We found that T‐lymphocytes and FOXP3+ infiltrates in primary tumours positively correlated with that of metastatic lesions (ρ = 0.512, P = 0.038 and ρ = 0.698, P = 0.007, respectively), while a strong trend existed for CD204+ infiltrates (ρ = 0.404, P = 0.087). We also observed T‐ and B‐lymphocytes, and CD204+ macrophages to be significantly higher in a dog's pulmonary metastasis compared to their primary tumour (P = 0.018, P = 0.018, P = 0.016, respectively), while FOXP3+ cells were only significantly higher in metastases when all primary tumour and metastasis lesions were compared without pairing (P = 0.036). Together, these findings suggest that the metastatic immune microenvironment may be influenced by that of the primary cOSA, and that primary tumour immune biomarkers could potentially be applied to predict immunotherapeutic responses in gross metastatic disease. We, therefore, provide a rationale for the treatment of cOSA pulmonary metastases with immunotherapeutics that enhance the anti‐tumour activity of these immune cells, particularly in dogs with moderate to high immune cell infiltration in their primary tumours. 相似文献

Effects of dexamethasone, levamisole, or combined dexamethasone-levamisole administration on polymorphonuclear neutrophil (PMN) function in healthy, adult female goats were studied. Goats were assigned to treated (n = 6) and control (n = 6) groups. In experiment 1, treated goats were given levamisole (6 mg/kg of body weight, IM). In experiment 2, treated goats were given 0.1 mg of dexamethasone/kg, IV, for 3 consecutive days, 1 mg of dexamethasone/kg, IV, for 6 consecutive days, and 6 mg of levamisole/kg, IM, with a 4th injection of 1 mg of dexamethasone/kg. All injections were administered 12 hours before blood collection. The PMN were evaluated for random migration and chemotaxis under agarose, ingestion of Staphylococcus aureus, cytochrome C reduction, iodination, and antibody-dependent cell-mediated cytotoxicity. Levamisole alone did not alter the function of caprine PMN. Both doses of dexamethasone caused increased random migration and decreased cytochrome C reduction and iodination. Dexamethasone resulted in no changes in chemotaxis, S aureus ingestion, and antibody-dependent cell-mediated cytotoxicity. Random migration and cytochrome C reduction returned toward base line in cells from dexamethasone and levamisole-treated goats. Although iodination activity in cells from dexamethasone-treated goats remained significantly (P less than 0.05) lower than those of controls after levamisole administration, a rebound toward base-line activity occurred. 相似文献

The riverscape perspective recognizes the heterogeneous habitat types within the stream corridor as a single, integrated ecological
unit operating across spatial scales. Although there is ample evidence that the riverscape notion is appropriate in understanding
the physical phenomena of stream corridors, significantly less attention has focused on its ecological ramifications. To this
end, we surveyed riverscape habitat variables and bird community characteristics in the Champlain Valley of Vermont, USA.
From the data collected, we used information theoretic methodology (AICc) to model relationships between bird community attributes and key habitat variables across the riverscape. Our models with
the greatest support suggest that riverine bird communities respond to a suite of characteristics; representing a variety
of riverscape habitats at the in-stream, floodplain, and riparian levels. Channel slope, drainage area, percent conifers,
and in-stream habitat condition were among the most influential variables. We found that piscivores are potentially important
indicators of riverscape condition, responding to a host of variables across the riverscape. Our results endorse a holistic
approach to assessing and managing the mosaic of patches in the riverscape and suggest that a riverscape approach has significant

BACKGROUND: Recent reports indicate increased amounts of mRNA from inflammation-related genes in the prodromal stage of laminitis. HYPOTHESIS: Cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) undergo distinct patterns of expression in equine laminae in the developmental stage (DEV) and acute clinical stage (LAM) of laminitis. ANIMALS: Horses selected from an outbred population were placed into 1 of 4 groups: DEV (n = 5), CON-3h (control group for DEV, n = 5), LAM (n = 5) and CON-10h (control group for LAM, n = 5). METHODS: Laminar and skin samples were obtained from (1) animals either undergoing leukopenia (DEV) or the onset of clinical signs of laminitis (LAM) after black walnut extract (BWE) administration and (2) animals either 3 (CON-3h) or 10 (CON-10h) hours after administration of water. Real-time quantitative polymerase chain reaction (RT-qPCR), immunoblotting, and immunohistochemical analysis were performed for COX-1 and COX-2. RESULTS: Upon immunohistochemical analysis of all 4 groups, COX-2 was expressed by most viable epithelial cells in both laminae and skin. COX-1 exhibited similar epithelial expression to COX-2 in skin epidermis, but was expressed exclusively in the basal layer of laminar epidermis. COX-1 protein was not detectable in dermal vasculature of equine skin or laminae, whereas COX-2 was present in endothelial and vascular smooth muscle cells of dermal vasculature in both skin and laminae in all groups. A marked increase in laminar COX-2 protein concentrations was detected on immunoblotting in the DEV group, although a lesser increase was observed in the LAM group. CONCLUSIONS AND CLINICAL IMPORTANCE: COX-2 protein expression is markedly increased in the resident laminar cell types in the developmental stage of BWE-induced laminitis. 相似文献

Certification and principles, criteria and indicators (PCI) describe desired ends for sustainable forest management (SFM) but do not address potential means to achieve those ends. As a result, forest owners and managers participating in certification and employing PCI as tools to achieving SFM may be doing so inefficiently: achieving results by trial-and-error rather than by targeted management practices; dispersing resources away from priority objectives; and passively monitoring outcomes rather than actively establishing quantitative goals. In this literature review, we propose six concepts to guide SFM implementation. These concepts include: Best Management Practices (BMPs)/Reduced Impact Logging (RIL), biodiversity conservation, forest protection, multi-scale planning, participatory forestry, and sustained forest production. We place these concepts within an iterative decision-making framework of planning, implementation, and assessment, and provide brief definitions of and practices delimited by each concept. A case study describing SFM in the neo-tropics illustrates a potential application of our six concepts. Overall our paper offers an approach that will help forest owners and managers implement the ambiguous SFM concept. 相似文献

ABSTRACT As more forest entities worldwide consider pursuing Forest Stewardship Council (FSC) certification, a critical question remains on whether stand-level management impacts differ between certified and uncertified forests. To begin to answer this question, we measured forest structure on three FSC-certified stands, three uncertified stands, and six adjacent unharvested reference stands (12 stands total) composed primarily of sugar maple (Acer saccharum) on non-industrial private properties in central Vermont, USA. The certified and uncertified partial harvests reduced total tree biomass and live tree carbon storage by one-third compared to reconstructed pre-harvest conditions. Both treatments also contained significantly lower densities of saplings and some mid-size trees compared to non-harvested references due to similar impacts from harvesting. The net present value of merchantable sugar maple over 10 year projections was consistently lower on certified than uncertified stands, but this difference was insignificant at discount rates from 4–8%. The certified stands contained significantly greater total residual volumes of coarse woody debris (standing and downed) than uncertified stands, although the debris was smaller than that found in unmanaged mature forests. Overall, our data suggest that FSC-certified harvested stands in northern hardwood forests have similar sugar maple timber value, aboveground live tree carbon storage value, similar live tree structure, and greater residual coarse woody debris than uncertified harvested stands. 相似文献
