Using sensory information for the prediction of future events is essential for survival. Midbrain dopamine neurons are activated by environmental cues that predict rewards, but the cellular mechanisms that underlie this phenomenon remain elusive. We used in vivo voltammetry and in vitro patch-clamp electrophysiology to show that both dopamine release to reward predictive cues and enhanced synaptic strength onto dopamine neurons develop over the course of cue-reward learning. Increased synaptic strength was not observed after stable behavioral responding. Thus, enhanced synaptic strength onto dopamine neurons may act to facilitate the transformation of neutral environmental stimuli to salient reward-predictive cues.  相似文献

COX-2-derived prostacyclin confers atheroprotection on female mice   总被引:1,自引:0,他引:1  
Female gender affords relative protection from cardiovascular disease until the menopause. We report that estrogen acts on estrogen receptor subtype alpha to up-regulate the production of atheroprotective prostacyclin, PGI2, by activation of cyclooxygenase 2 (COX-2). This mechanism restrained both oxidant stress and platelet activation that contribute to atherogenesis in female mice. Deletion of the PGI2 receptor removed the atheroprotective effect of estrogen in ovariectomized female mice. This suggests that chronic treatment of patients with selective inhibitors of COX-2 could undermine protection from cardiovascular disease in premenopausal females.  相似文献

Role of prostacyclin in the cardiovascular response to thromboxane A2   总被引:1,自引:0,他引:1  
Thromboxane (Tx) A2 is a vasoconstrictor and platelet agonist. Aspirin affords cardioprotection through inhibition of TxA2 formation by platelet cyclooxygenase (COX-1). Prostacyclin (PGI2) is a vasodilator that inhibits platelet function. Here we show that injury-induced vascular proliferation and platelet activation are enhanced in mice that are genetically deficient in the PGI2 receptor (IP) but are depressed in mice genetically deficient in the TxA2 receptor (TP) or treated with a TP antagonist. The augmented response to vascular injury was abolished in mice deficient in both receptors. Thus, PGI2 modulates platelet-vascular interactions in vivo and specifically limits the response to TxA2. This interplay may help explain the adverse cardiovascular effects associated with selective COX-2 inhibitors, which, unlike aspirin and nonsteroidal anti-inflammatory drugs (NSAIDs), inhibit PGI2 but not TxA2.  相似文献

Objective: To determine the effect of gastrointestinal (GI) decontamination on the incidence of prolonged prothrombin time (PT) in dogs after anticoagulant rodenticide ingestion. Design: Retrospective study. Setting: Urban emergency room. Animals: One hundred and fifty‐one client‐owned dogs. Measurements: Dogs presented to the emergency room within 6 hours of ingestion of an anticoagulant rodenticide and had a PT measured within 2–6 days of toxicant ingestion before initiating vitamin K therapy were included. Dogs were categorized as treated or untreated based on the institution of vitamin K therapy following PT testing. The signalment, body weight, type of rodenticide ingested, time elapsed between ingestion and initial presentation, method(s) of GI decontamination, and the times elapsed between both toxicant ingestion and initial hospital presentation until determination of PT were recorded. The PT results were recorded as well as any treatment received following the recheck examination. Any reported incidents of bleeding or untoward effects between exposure and reexamination were recorded. Main results: Of 151 dogs, only 11 dogs (8.3%) developed prolonged PT requiring vitamin K supplementation. None of the 11 dogs with prolonged PTs exhibited signs of bleeding or required transfusion therapy. No differences in age, weight, or time elapsed between treated and untreated patients were found. Conclusions: The incidence of prolonged PT is low in dogs receiving GI decontamination within 6 hours of anticoagulant rodenticide ingestion. Delaying vitamin K therapy until a PT has been assessed 48–72 hours after initial exposure appears to be safe and sensitive in dogs following anticoagulant rodenticide ingestion.  相似文献

Abnormal extrusion of canine teeth is often noted in middle-aged and geriatric domestic cats. The same age group of cats also is commonly affected by tooth resorption (TR). This study explored the relationship between these two phenomena of unknown etiology. Using digital radiography, the distance between the alveolar margin (AM) and cementoenamel junction (CEJ), referred to as the AM-CEJ distance, was measured in clinically and radiographically healthy maxillary canine teeth of 24 TR-affected and 29 TR-free cats. The mean AM-CEJ distance of maxillary canine teeth of cats with and cats without TR was 2.68-mm and 2.22-mm, respectively. An analysis of covariance adjusting for age revealed a significant correlation (p = 0.02) between tooth extrusion and TR. Extrusion of the maxillary canine teeth became clinically apparent when an AM-CEJ distance of 2.5-mm or greater was evident in the absence of horizontal or vertical alveolar bone loss. Based on this criterion, 15 of 24 cats with TR (63.0 %) exhibited extrusion of maxillary canine teeth, compared to 9 of 29 cats without TR (31.0 %). Four extruded and five non-extruded maxillary canine teeth were evaluated histologically. Cementum of extruded teeth was significantly thicker compared to that of non-extruded teeth. Four of 4 canine teeth with extrusion (100 %) showed histological evidence of resorption, compared to 1 of 5 canine teeth without extrusion (20.0 %). These results suggest that tooth extrusion is linked to or may be caused by similar factors responsible for the development of TR.  相似文献

Hypovolemia and hypoperfusion are common life-threatening problems in animals presented to the emergency veterinarian. Assessment of physical findings and rapid recognition and treatment of abnormal tissue perfusion are crucial in optimizing outcome. The clinician should be familiar with the disease being treated and the types of fluids that are available. Development of a fluid therapy plan to correct life-threatening abnormalities and patient monitoring during treatment play an important role in patient outcome.  相似文献

The composition of the greenline plant community is linked to the stability of riparian ecosystems. Cool season exotic grasses are invading native plant communities across the northern Great Plains, potentially compromising streambank stability and increasing the risk of erosion within riparian ecosystems. To determine how the species composition of the greenline community impacts stream type and the risk of streambank erosion, thirty five reaches across five watersheds were sampled to determine the dominant greenline vegetation. At each reach, a cross-section was sampled to determine stream type, greenline vegetation, and risk of streambank erosion. Channel types were delineated using Rosgen’s classification of natural rivers. Canopy cover and composition was assessed using the line point intercept method along a 30.5 m transect in the greenline community. Plants recorded were grouped by their wetland indicator status for the central Great Plains. The Bank Erosion Hazard Index (BEHI) was used to assess the streams risk of erosion by calculating the difference between the bank height and bank full height, average plant rooting depth and density, bank angle degree, and the dominant texture of the bank material. Bank height ratio (BHR) was assessed as a measure of streambank stability and floodplain connectivity. A Nonmetric Multidimensional Scaling ordination was performed to analyze plant community influences. Analysis of the data determined that the most stable stream types (E and C channels), lower BEHI scores, and stable bank height ratios were associated with high amounts of litter and facultative wet species. In comparison, unstable F channels were associated with early successional species and bare ground. Sites with the higher BEHI scores were associated with greenlines comprised of upland and facultative upland and saline tolerant species. Late successional facultative wetland species were found to provide the most protection to intermittent streambanks.  相似文献
