Nitrogen (N) is an important nutrient for re-vegetation during ecosystem restoration, but the effects of cover restoration on soil N transformations are not fully understood. This study was conducted to investigate N transformations in soils with different cover restoration ages in Eastern China.
Materials and methods
Soil samples were collected from four degraded and subsequently restored lands with restoration ages of 7, 17, 23, and 35 years along with an adjacent control of degraded land. A 15N tracing technique was used to quantify gross N transformation rates.
Results and discussion
Compared with degraded land, soil organic carbon (SOC) and total N (TN) increased by 1.60–3.97 and 2.49–5.36 times in restoration land. Cover restoration increased ammonium and nitrate immobilization, and dissimilatory nitrate reduction to ammonium (DNRA) by 0.56–0.96, 0.34–2.10, and 0.79–3.45 times, respectively, indicating that restoration was beneficial for N retention. There were positive correlations between SOC content and ammonium and nitrate immobilization and DNRA, indicating that the increase in soil N retention capacity may be ascribed to increasing SOC concentrations. The stimulating effect of SOC on ammonium immobilization was greater than its effect on organic N mineralization, so while SOC and TN increased, inorganic N supply did not increase. Autotrophic and heterotrophic nitrification increased with increasing SOC and TN concentrations. Notably, heterotrophic nitrification was an important source of NO3??N production, accounting for 47–67% of NO3??N production among all restoration ages.
Conclusions
The capacity of N retention was improved by cover restoration, leading to an increase in soil organic carbon and total N over time, but inorganic N supply capacity did not change with cover restoration age.
Johne’s disease (paratuberculosis) is an economically important disease of cattle worldwide. The disease is caused by Mycobacterium avium subsp. paratuberculosis (MAP), a fastidious gram-positive bacterium. PCR is increasingly used in diagnostic laboratories for the detection of MAP in fecal samples given the rapid test turnaround time and sensitivity and specificity comparable to fecal culture. However, efficient extraction of DNA for sensitive detection of MAP by PCR is affected by the complex lipid-rich cell wall of MAP and the presence of PCR inhibitors in feces. We evaluated a high-throughput nucleic acid extraction method (MagMAX core nucleic acid purification kit with mechanical lysis module) in conjunction with an hspX gene PCR for the detection of MAP from bovine fecal samples, which resulted in correct identification of all negative (13 of 13) and positive (35 of 35) proficiency test samples obtained from the National Veterinary Services Laboratories. Formal public sector soybean breeding in Africa spans over four decades, and it was initiated by the International Institute of Tropical Agricultural (IITA). As the demand of soybean continues to outstrip production, strategic projects such the Tropical Legume (TL) were initiated, in which the main goal was to enhance the productivity of soybean in the farmers’ fields in Sub‐Saharan Africa. One of the strategies to enhance the productivity of soybean in the farmers’ fields is through developing and deploying improved soybean varieties in the target countries. Through the TL I and TL II projects, a number of varieties were released in the target countries, Kenya, Nigeria, Malawi and Mozambique by employing participatory variety selection (PVS). This review provides highlights of the achievements made by IITA breeding programme and insights of what needs to be done to enhance yield improvement for soybean in Africa using demand‐driven breeding approaches. 相似文献
In order to improve the biological control agent (BCA) efficacy, stress factors threatening the viability of microorganisms during spray application need to be determined. The effect of spray mixture temperature and exposure time on Trichoderma harzianum T 22 and Bacillus amyloliquefaciens QST713 viability were tested. Concurrently the combined effect of mechanical and thermal stress effect on BCA viability were tested at two initial spray mixture temperatures (14 and 25 °C) by simulating a spray application using airblast sprayers featured by different tank capacity and a spray liquid circuit (without and with hydraulic agitation system). To assess the BCA microorganism viability, spray mixture samples were collected at time intervals along trials and plated to count the colony forming units (CFU).
Results
The critical temperature threshold that inhibited BCA viability was 35 °C with 30 min of exposure. The sprayer type, the initial temperature of the spray mixture and the temperature increment during the trials significantly decreased the number of CFU recovered. When simulating a spray application, the spray mixture temperature increase rate was determined mainly by the residual amount of spray mixture in the tank. Even if the tank capacity does not substantially affect the final temperature reached by the spray mixture, the higher residual spray mixture in bigger tanks can expose the BCAs for a longer time to critical temperatures.
Epigenetics provides a molecular mechanism of inheritance that is not solely dependent on DNA sequence and that can account for non-Mendelian inheritance patterns. Epigenetic changes underlie many normal developmental processes, and can lead to disease development as wel. While epigenetic effects have been studied in wel-characterized rodent models, less research has been done using agricultural y important domestic animal species. This review wil present the results of current epigenetic research using farm animal models(cattle, pigs, sheep and chickens). Much of the work has focused on the epigenetic effects that environmental exposures to toxicants, nutrients and infectious agents has on either the exposed animals themselves or on their direct offspring. Only one porcine study examined epigenetic transgenerational effects; namely the effect diet micronutrients fed to male pigs has on liver DNA methylation and muscle mass in grand-offspring(F2 generation). Healthy viable offspring are very important in the farm and husbandry industry and epigenetic differences can be associated with production traits. Therefore further epigenetic research into domestic animal health and how exposure to toxicants or nutritional changes affects future generations is imperative. 相似文献
To report our experience with thoracoscopic lung lobectomy in dogs with primary lung tumors and compare the long‐term outcome between dogs with primary lung tumors treated with lobectomy via thoracoscopy or thoracotomy.
Study Design
Retrospective case series; case–control study.
Animals
Thirteen dogs with primary lung tumors treated with thoracoscopic lung lobectomy; case‐matched cohort of 9 dogs with primary lung tumors treated with lobectomy via thoracotomy.
Methods
Medical records from dogs that underwent thoracoscopic lung lobectomy at a single institution were reviewed. Only cases with primary lung tumors were included. Surgical complications and conversion rate were reported. Cases without conversion and with clean margins on histologic examination were matched on prognostic criteria to cases with primary lung tumors treated with lobectomy via thoracotomy. Survival was compared between thoracoscopy and thoracotomy.
Results
Thirteen dogs with primary lung tumors that measured 2.2–7 cm underwent thoracoscopic lung lobectomy. Nine dogs were diagnosed with carcinoma and 4 with histiocytic sarcoma. There were 3 conversions to open thoracotomy because of poor visualization. There were no differences in short‐term outcome between dogs that had their procedures completed via thoracoscopy and those that were converted to thoracotomy. Nine dogs with clean margins were matched with 9 dogs treated via thoracotomy. Mean (standard deviation) followup was 367 (327) days for dogs undergoing thoracoscopy and 603 (612) days for dogs undergoing thoracotomy. The 2‐year survival rate was 44% for thoracoscopy and 56% for thoracotomy (P = .942).
Conclusion
Selected primary lung tumors in dogs can be safely resected with thoracoscopy and not affect long‐term outcome. 相似文献
