Fasciolosis is an endemic zoonotic parasitic disease with significant impacts on human health and both animal health and production. Early post-infection impacts on the host remain unclear. The objective of this study was to determine the changes, if any, to levels of endotoxin in cattle plasma in response to early-stage infection with Fasciola hepatica. Thirty-six (36) commercial bred cattle were experimentally infected with approximately 400 viable metacercariae. Plasma lipopolysaccharide (endotoxin) levels were examined on 24 occasions from 0 h before infection to 336 h after infection using the Limulus Amoebocyte Lysate chromogenic end point assay and compared with that of six (6) uninfected control animals. Peak lipopolysaccharide levels in infected animals were reached at 52 h after infection and returned to pre-infection levels at time 144 h after infection. Infected animals had significantly elevated lipopolysaccharide levels between 24 and 120 h after infection when compared to uninfected animals. The mean change in endotoxin units (EU)/mL over time after infection was statistically significant in infected animals. Elevations of lipopolysaccharide occurred in all infected animals suggesting a possible repeatable and titratable endotoxemia conducive to therapeutic agent model development. 相似文献

An important part of agricultural adaptation is the timing of crop sowing dates, affecting yields and the level of risk incurred during a particular season. Cold stress is especially relevant in maize, Zea mays L., so that the timing of planting in the spring is a tactical response to short‐term weather, but is also subject to strategic planning with regard to longer‐term climate. Both factors compare the potential implications of cold stress to the additional yield obtainable through earlier planting. New cultivars suited to growing conditions in Europe and generally increasing spring temperatures have enabled earlier planting, but it is still dependent on short‐term weather during the planting period. In the context of field‐level decision‐making, a panel regression is used to estimate the relationship between weekly local temperature and precipitation and planting dates at specific sites throughout Germany. Next, localised weather data and planting behaviour are linked to yields at the district (Landkreis) level to show the effects of planting date on yield. Based on these relationships optimal planting dates are explored with some associated costs and benefits. Results show a trend towards earlier planting that follows observed increasing spring temperatures and the availability of more cold‐tolerant cultivars but this advance is buffered by the increasing severity of minimum temperatures during a critical period. Earlier planting potentially increases yield but this is offset by additional management costs and risk. A robust and simple depiction of farmer behaviour in climatic, technological and economic context can help to understand trends in crop management and productivity that effect agricultural landscapes. 相似文献

The purpose of this review is to examine the current knowledge of the role of trees in providing shelter for pastures, crops, and livestock, for controlling erosion of soils and improving productivity and sustainability of agricultural production in Australia — and the extent to which this knowledge has been applied.Land degradation — tree loss and associated soil salinity, water and wind erosion, soil acidification, soil structural decline and nutrient degradation — is evidence that our primary production systems are not sustainable. We have sought increased production without proper consideration of the ecological context of that system. About half of Victoria's crop and pasture lands are affected or at risk, and in Western Australia about 25% of the cleared agricultural land is wind-eroded and 60% is potentially susceptible, salinity affects 0.43 m ha and half of the divertible surface water is affected by salinity. Similar problems occur in other States. At least 43 m ha or 13% of our rangelands are seriously degraded by wind erosion caused by overgrazing, often coinciding with drought or a run of drier years.Minimum tillage and stubble management for erosion control in cropping has been a major extension and research activity in Australian agriculture. Severe weather, combined with imperfect adoption of appropriate grazing and crop management systems, shows the weakness of complete reliance on these methods of erosion control. An effective system must accommodate the impact of extreme events, which are the most damaging. However, the complementary use of windbreaks to reduce soil erosion is rare, and their establishment has not been promoted, despite the wide-spread adoption of this technology by other countries.In the cropping and higher rainfall grazing areas, the systematic planting of 10% of the land in a net of shelterbelts/timberbelts/clusters could achieve a 50% windspeed reduction; this would substantially improve livestock and pasture production in the short and long-term. Wind erosion could be dramatically reduced and crop production probably increased by the use of windbreaks. Wheat and oat yield at Rutherglen (Victoria), and lupin yield at Esperance (Western Australia), were increased in the sheltered zone by 22% and 47%, and 30%, respectively.In semi-arid and dry temperate areas, planting of 5% of the land to shelter could reduce windspeed by 30–50% and soil loss by up to 80%. This planting would also contribute substantially to achieving other objectives of sustainable agriculture. Agroforestry — particularly timberbelts applications — will be important in the long-term strategy for achieving revegetation. If some of the trees yield a marketable product then the adoption of the system will be more readily achieved.In the arid (pastoral) areas there is an urgent need to promote the ethic that preservation and improvement of the perennial grass and shrub vegetation is critical for the protection of the soil and maintenance of land capability. Control of animal grazing remains the sole means of preventing erosion in much of this zone. While satellite imagery allows us to assess the condition of leasehold lands, we have failed to achieve stocking policies that will halt the degradation of our rangelands. 相似文献

We measured vertical and horizontal variation in canopy transmittance of photosynthetically active radiation in five Pseudotsuga menziesii (Mirb.) Franco-Tsuga heterophylla (Raf.) Sarg. (Douglas-fir-western hemlock) stands in the central Cascades of southern Washington to determine how stand structure and age affect the forest light environment. The shape of the mean transmittance profile was related to stand height, but height of mean maximum transmittance was progressively lower than maximum tree height in older stands. The vertical rate of attenuation declined with stand age in both the overstory and understory. A classification of vertical light zones based on the mean and variance of transmittance showed a progressive widening of the bright (low variance and high mean) and transition (high variance and rapid vertical change) zones in older stands, whereas the dim zone (low variance and mean) narrowed. The zone of maximum canopy surface area in height profiles, estimated by inversion of transmittance profiles, changed from relatively high in the canopy in most young stands ("top-heavy") to lower in the canopy in older stands ("bottom-heavy"). In the understory, all stands had similar mean transmittances, but the spatial scale of variation increased with stand age and increasing crown size. The angular distribution of openness was similar in all stands, though the older stands were less open at all angles than the younger stands. Understory openness was generally unrelated to transmittance in the canopy above. Whole-canopy leaf area indices, estimated using three methods of inverting light measurements, showed little correspondence across methods. The observed patterns in light environment are consistent with structural changes occurring during stand development, particularly the diversification of crowns, the creation of openings of various sizes and the elaboration of the outer canopy surface. Forest and glasshouse experiments are described in which necrosis of beech bark followed wound inoculation with Nectria isolates during dormancy. An obscure sesquiterpene, rotundone, has been identified as a hitherto unrecognized important aroma impact compound with a strong spicy, peppercorn aroma. Excellent correlations were observed between the concentration of rotundone and the mean 'black pepper' aroma intensity rated by sensory panels for both grape and wine samples, indicating that rotundone is a major contributor to peppery characters in Shiraz grapes and wine (and to a lesser extent in wine of other varieties). Approximately 80% of a sensory panel were very sensitive to the aroma of rotundone (aroma detection threshold levels of 16 ng/L in red wine and 8 ng/L in water). Above these concentrations, these panelists described the spiked samples as more 'peppery' and 'spicy'. However, approximately 20% of panelists could not detect this compound at the highest concentration tested (4000 ng/L), even in water. Thus, the sensory experiences of two consumers enjoying the same glass of Shiraz wine might be very different. Rotundone was found in much higher amounts in other common herbs and spices, especially black and white peppercorns, where it was present at approximately 10000 times the level found in very 'peppery' wine. Rotundone is the first compound found in black or white peppercorns that has a distinctive peppery aroma. Rotundone has an odor activity value in pepper on the order of 50000-250000 and is, on this criterion, by far the most powerful aroma compound yet found in that most important spice. 相似文献

Wheat flour from plants deficient in sulfur has been shown to contain substantially higher levels of free amino acids, particularly asparagine and glutamine, than flour from wheat grown where sulfur nutrition was sufficient. Elevated levels of asparagine resulted in acrylamide levels up to 6 times higher in sulfur-deprived wheat flour, compared with sulfur-sufficient wheat flour, for three varieties of winter wheat. The volatile compounds from flour, heated at 180 degrees C for 20 min, have been compared for these three varieties of wheat grown with and without sulfur fertilizer. Approximately 50 compounds were quantified in the headspace extracts of the heated flour; over 30 compounds were affected by sulfur fertilization, and 15 compounds were affected by variety. Unsaturated aldehydes formed from aldol condensations, Strecker aldehydes, alkylpyrazines, and low molecular weight alkylfurans were found at higher concentrations in the sulfur-deficient flour, whereas low molecular weight pyrroles and thiophenes and sugar breakdown products were found at higher concentrations in the sulfur-sufficient flour. The reasons for these differences and the relationship between acrylamide formation and aroma volatile formation are discussed. 相似文献
