This study was conducted to identify and analyse the expression of gametogenesis‐associated genes and proteins in foetal and adult buffalo gonads of both the sexes. Relative quantification of the genes was determined by qPCR and Western blotting. Immunohistochemistry was also performed for various gametogenesis‐associated proteins in foetal and adult gonads of both the sexes. We observed significantly (p < 0.05) increased expression of primordial germ cell‐specific, meiotic as well as genes associated with oocyte maturation and development in foetal ovaries as compared to the adult ones. However, significantly (p < 0.05) increased expression of proteins associated with oocyte maturation like GDF9 and ZP4 was found in adult ovaries, indicating temporal regulation of mRNA translation during oogenesis. Meiotic genes showed significantly (p < 0.05) increased expression in adult testes as compared to foetal testes and ovaries, indicating onset of meiosis at a later stage in spermatogenesis. In general, the expression of primordial germ cell‐associated as well as meiotic genes was higher in adult testes, indicating the increased biological activity in the organ. Immunohistochemistry revealed localized expression of gametogenesis‐associated proteins in ovarian follicles and seminiferous tubules of testes, while the surrounding somatic tissues were devoid of these proteins. The study gives an understanding of the sequential and temporal events of gene expression as well as mRNA translation during male and female gametogenesis. It could also be concluded that follicles and seminiferous tubules are the functional units of the female and male gonads, respectively, and their function could be enhanced by appropriate chemical and genetic intervention of the somatic tissue immediately surrounding them. This assumes importance in the context that buffalo attains sexual maturity at an older age of 2–3 years and have smaller ovaries with lesser number of primordial follicles in comparison with cattle, which is suggested to be the main reason of their poor breeding performance.  相似文献

A study was conducted to examine the distribution pattern of four rhizomatous medicinal and aromatic plant species (MAPs) viz., Angelica glauca, Pleurospermum angelicoides, Rheum emodi and Arne- bia benthamii in different forest stands in Central Himalaya. Results show that A. glauca and P. angelicoides had a higher (50%) frequency at Chipkoan, Garpak and Phagati forest, R. emodi had a higher (60%) frequency at Rishikund, Suki and Himtoli, and A. benthamii had a higher (70%) frequency at Suki and Khambdhar The densities of A. glauca (0.6 plants·m -2 ) and P. angelicoides (0.5 plants·m -2 ) were higher at Chipkoan and Garpak sites than at other micro-sites, while densities of R. emodi (0.8 plants·m -2 ) and A. benthamii (1.0 plants·m -2 ) were higher at Suki and Khambdhar sites. A. glauca had highest total basal covers (TBC) (1.2 cm 2 ·m -2 ) at Chipkoan, P. angelicoides had highest TBC (0.92 cm 2 ·m -2) at Lati kharak site, A. benthamii had the highest TBC (6.48 cm 2 ·m -2 ) atKhambdhar, and R. emodi had highest TBC (4.53 cm 2 ·m -2 ) at Rishikund. For the four studied species, A. glauca showed a contagious distribution, P. angelicoides and R. emodi showed the random and A. benthamii showed the regular type of distribution.  相似文献

The efficacy of a synthetic detergent (Surf Excel) as a potential chemical hybridizing agent in Brassica juncea was studied. Foliar sprays with various concentrations of the detergent caused reductions in plant height, number of branches and leaves per plant, size of leaves, anther size, pollen per flower, ovules per flower, pollen fertility, fruits per plant, fruit size, seeds per fruit, total yield per plant and 100 seed weight as compared with those of untreated plants. The style in the floral buds of plants sprayed with different concentrations of Surf Excel elongated and so the receptive stigma protruded from the buds which facilitated cross‐pollination by honey bees. The plants sprayed once with 2% Surf Excel exhibited an elongated style with a raised receptive stigma and 100% pollen sterility without causing a significant reduction in total yield.  相似文献

A line × tester analysis was carried out in mulberry (Morus spp.) to determine the genetic interaction in the expression of various quantitative characters including leaf yield. Eight clonal varieties were selected, 3 of them were designated as lines (♀) viz. Berhampore-1, China white and MS-5 and 5 of them were called as testers (♂) viz. Mandalaya, Kosen, Assamjati, MS-1 and Kajli. Combining ability studies were conducted on these parents along with their F1 hybrids for the variables laminar index, growth rate, weight of 100 dry leaves, number of primary branches per plant, plant height, nodal distance, leaf-twig ratio, aerial biomass, moisture content, moisture retention capacity and leaf yield. Broad genetic variability was observed among the genotypes. The ratio of General Combining Ability (GCA) and Specific Combining Ability (SCA) indicated the predominance of non-additive genes in mulberry. While China white (female) and MS-1 (male) were the best general combiners among the parents, Berhampore-1 × Kajli was the best cross for leaf yield. Results suggest that selective crossing followed by proper screening may be the best approach for breeding of high yielding varieties in mulberry. The objective of this study was to explore the possibility of producing wild buffalo embryos by interspecies somatic cell nuclear transfer (iSCNT) through handmade cloning using wild buffalo somatic cells and domestic buffalo (Bubalus bubalis) oocytes. Somatic cells derived from the ear skin of wild buffalo were found to express vimentin but not keratin and cytokeratin‐18, indicating that they were of fibroblast origin. The population doubling time of skin fibroblasts from wild buffalo was significantly (p < 0.05) higher, and the cell proliferation rate was significantly (p < 0.05) lower compared with that of skin fibroblasts from domestic buffalo. Neither the cleavage (92.6 ± 2.0% vs 92.8 ± 2.0%) nor the blastocyst rate (42.4 ± 2.4% vs 38.7 ± 2.8%) was significantly different between the intraspecies cloned embryos produced using skin fibroblasts from domestic buffalo and interspecies cloned embryos produced using skin fibroblasts from wild buffalo. However, the total cell number (TCN) was significantly (p < 0.05) lower (192.0 ± 25.6 vs 345.7 ± 42.2), and the apoptotic index was significantly (p < 0.05) higher (15.1 ± 3.1 vs 8.0 ± 1.4) for interspecies than that for intraspecies cloned embryos. Following vitrification in open‐pulled straws (OPS) and warming, although the cryosurvival rate of both types of cloned embryos, as indicated by their re‐expansion rate, was not significantly different (34.8 ± 1.5% vs 47.8 ± 7.8), the apoptotic index was significantly (p < 0.05) higher for vitrified–warmed interspecies than that for corresponding intraspecies cloned embryos (48.9 ± 7.2 vs 23.9 ± 2.8). The global level of H3K18ac was significantly (p < 0.05) lower in interspecies cloned embryos than that in intraspecies cloned embryos. The expression level of HDAC1, DNMT3a and CASPASE3 was significantly (p < 0.05) higher, that of P53 was significantly (p < 0.05) lower in interspecies than in intraspecies embryos, whereas that of DNMT1 was similar between the two types of embryos. In conclusion, these results demonstrate that wild buffalo embryos can be produced by iSCNT.  相似文献

Osmotic adjustment (OA) is considered as an important physiological mechanism of drought adaptation in many crop plants. The present investigation was aimed at assessing the importance of OA in improving productivity under drought. Using two automated rain-out shelters, 26 extra-short-duration pigeonpea [Cajanus cajan (L.) Millsp.] genotypes were grown with irrigation during the growth period or with water deficit imposed from flowering until maturity. Mean leaf Ψs100 (60–92 DAS) under drought correlated significantly (r2=0.72**; n=26) to the mean OA (60–92 DAS) and contributed 72% of the genotypic variation in OA. Significant genotypic variation was observed in the initiation of OA, the duration of OA and the degree of OA. Based on the measured OA at 72, 82, and 92 days after sowing (DAS), genotypes were grouped into five different clusters. Genotypic differences in total dry matter production under drought were positively associated with OA at 72 DAS (r2=0.36**, n=26). Significant positive relationship between OA at 72 DAS and grain yield under drought was found (r2=0.16*; n=26). However, OA towards the end of pod filling phase, i.e. at 92 DAS, had a significant negative relationship with grain yield under drought (r2=0.21*; n=26). Genotypic differences in grain yield under drought was best explained using stepwise multiple regression to account for differences in OA at 72, 82, and 92 DAS (r2=0.41**; n=78). The degree of OA at 72 and 82 DAS contributed positively to the grain yield, whereas OA at 92 DAS contributed negatively to this relationship.  相似文献

Phytic acid, saponin and polyphenol contents in grains of various varieties of black gram (Vigna mungo) Mung bean (Vigna radiata L.) amphidiploids ranged from 697 to 750, 2746 to 2972 and 702 to 783 mg/100 g, respectively. Domestic processing and cooking methods including soaking, ordinary and pressure cooking of soaked and unsoaked seeds, and sprouting significantly lowered phytic acid, saponin and polyphenol contents of the amphidiploid seeds. Soaking for 18 h removed 31 to 37% of the phytic acid; the extent of removal was higher with long periods of soaking. Saponins and polyphenols were relatively less affected. Loss of the antinutrients was greater when soaked instead of unsoaked seeds were cooked. Pressure cooking had a greater effect than ordinary cooking. Antinutrient concentrations declined following sprouting; the longer the period of germination the greater was the reduction.  相似文献
