Malignant hyperthermia susceptibility: biochemical basis for pathogenesis and diagnosis.

Malignant hyperthermia (MH) is a hypermetabolic and hypercontractile syndrome triggered by anesthesia or various stressors that cause a sustained increase in sarcoplasmic ionized Ca. Susceptibility is apparently inherited in an autosomal dominant pattern. The primary molecular defect results in hypersensitive ligand-gating of the Ca-release channel of sarcoplasmic reticulum (SR) in skeletal muscle: channel opening is stimulated by abnormally low concentrations of agonist. We attribute MH to a mutation in the gene for the fast twitch muscle isoform of the Ca-channel, resulting in the expression of a cardiac-like isoform in fast muscle. Syndromes with some resemblance to MH can occur due to other genetic or acquired imbalances in Ca-flux across SR that favor net release of Ca. Either defective uptake or release can be detected as increased sensitivity of muscle to the contracture-producing effects of caffeine and halothane. Thus, caffeine and/or halothane contracture tests for MH-susceptibility may give false positives when there is decreased Ca-uptake, such as in muscular dystrophies. Ca-channel hypersensitivity and decreased Ca-uptake activity can be detected by assays using isolated SR. Functional assays using lymphocytes are being assessed as potential replacements for muscle contracture tests. Polymorphism analysis of proteins or nucleic acids for the MH or closely-linked genes has been used to trace the inheritance of MH-susceptibility.     

Influence of footing surface on mounting and other sexual behaviors of estrual Holstein cows

Seven ovariectomized Holstein cows, treated sequentially with progesterone and estradiol benzoate to induce estrus, were used to determine whether cows showed a preference for mounting and displaying other sexual behaviors toward estrual or nonestrual cows tied on dirt or concrete surfaces. Preference tests were conducted in a test area that consisted of equal-sized concrete and dirt surfaces; two cows, designated A and B, were tied on the two surfaces, one on either side. Cows A and B were treated so that on each of four test days both were estrual, only A was estrual, only B was estrual or neither A nor B were estrual. On each test day, five estrual test cows were introduced individually into the test area for two 30-min test periods. During the first test period, tied cow A was on concrete and tied cow B was on dirt, but during the second test period, their positions were switched. Test cows were able to move freely from surface to surface and to interact with tied cow A or B. Estrual test cows spent 21.6 +/- 1.4 min of each 30-min test period on dirt, regardless of the estrual status of the cow tied on concrete or dirt. Mounting activity was 3- to 15-fold greater on dirt than on concrete when there was an estrual cow tied on dirt, regardless of the estrual status of the cow on concrete. Mounting activity was fourfold greater on concrete than on dirt when there was a tied estrual cow on concrete and a tied nonestrual cow on dirt.(ABSTRACT TRUNCATED AT 250 WORDS)     

Causes of death and illness in the native sheep of North Ronaldsay, Orkney. I. Adult sheep

Post-mortem examinations on 71 native sheep found dead on the island of North Ronaldsay, Orkney in four visits between April 1983 and July 1985 were carried out. The sheep in this almost feral flock have access to a small area of unmanaged moorland pasture but are otherwise restricted to the foreshore where they subsist largely on Laminaria spp. and other seaweeds. Young adult animals died largely of heavy parasite burdens combined with inadequate nutrition, while the older sheep often starved because of severe dental disease precipitated by heavy deposits of tartar on the cheek teeth--rarely seen in sheep on a more conventional diet. Other underlying metabolic conditions, such as the extensive mineralization of the kidney medulla in many mature sheep, may be debilitating. The pathological findings suggest that adaptation to the peculiar environmental rigours and dietary restrictions on North Ronaldsay is less complete than has previously been assumed.     

Improving sow productivity through management during gestation, lactation and after weaning

Number of pigs produced per sow per year is dependent upon the number of pigs born live, the number that survive to weaning and the interval between consecutive farrowings for the sow. Feeding and management of the sow during late gestation affects birth weight and amount of energy stored as glycogen and lipid in the piglet. Piglets that are heavier and that have more energy stores have a higher survival rate. Adding fat to the sow's diet during the last month of gestation or altering the sow's metabolism to direct more nutrients to the fetus are methods for increasing piglet birthweight and energy stores. Feeding the sow properly during lactation is important for maximum yield of milk and milk energy, which affects survival of pigs to weaning, and for rebreeding performance of the sow after weaning. Energy intake during lactation can be increased by adding fat to diets, and this is beneficial in situations where feed intake is insufficient to meet the sow's requirements. For example, fat supplementation during lactation is beneficial for primiparous sows and for sows lactating during hot weather. The minimum practical lactation length is about 2 wk for normal rebreeding performance of the sow. Split weaning or separation of the litter from the sow for 6 to 12 h/d will shorten the rebreeding interval or induce estrus during lactation. Administration of pregnant mare's serum gonadotropin, with or without human chorionic gonadotropin, will induce estrus during lactation, and the response is better after the second week of lactation. Similar treatments at weaning will shorten the rebreeding interval.(ABSTRACT TRUNCATED AT 250 WORDS)     

Acute viral hepatitis in California sea lions

Acute viral hepatitis was diagnosed in five California sea lions (Zalophus californianus) stranded along the Los Angeles coast. Light microscopy revealed large nuclear inclusion bodies in hepatocytes. Electron microscopy provided evidence that these inclusion bodies were composed of adenovirus-like virions. Attempts to grow the virus in cell culture systems were unsuccessful.     

Diagnosing appendicular osteosarcoma with ultrasound-guided fine-needle aspiration: 36 cases

OBJECTIVES: To determine the ability to obtain diagnostic cytology samples from appendicular bone lesions using ultrasound-guided needle aspirations. Secondary objectives were to compare cytological evaluations with histopathological results and to determine the utility of staining malignant mesenchymal cells for the presence of alkaline phosphatase. METHODS: Aspirations from 36 aggressive appendicular bone lesions with histological diagnoses were included in the study. Ultrasound was used to guide the needle to the medullary cavity or the adjacent soft tissue mass. The smears stained with Wright-Giemsa and nitroblue tetrazolium chloride/5-bromo-4-chloro-3-indoyl phosphate toluidine salt (NBT/BCIP) were examined. RESULTS: A diagnostic sample was obtained in 32 of the 36 cases. Of the 32 diagnostic samples, cytology indicated sarcoma, with a sensitivity of 97 per cent (confidence interval: 83 to 100 per cent) and a specificity of 100 per cent (confidence interval: 16 to 100 per cent). When a diagnosis of sarcoma was made on cytology, alkaline phosphatase staining indicated osteosarcoma, with a sensitivity of 100 per cent (confidence interval: 87 to 100 per cent). CLINICAL SIGNIFICANCE: The results of this study indicate that ultrasound-guided needle aspiration of aggressive bone lesions is a viable technique for identifying malignant mesenchymal cells and for diagnosing sarcomas. It is cost-effective and minimally invasive. Furthermore, identifying alkaline-phosphatase-negative malignant mesenchymal cells from a bone aspiration may rule out osteosarcoma, whereas alkaline-phosphatase-positive malignant mesenchymal cells are suggestive of osteosarcoma.     

Body condition of sows across parities and relationship to reproductive performance

Weight, heartgirth, backfat and body condition of sows was monitored in a commercial, farrow-to-finish unit during 1 yr. Measurements were obtained during the third, ninth and fifteenth week of gestation and the day after weaning. Litter performance and rebreeding rate also were recorded. Body weight and heartgirth increased (P less than .01) over parities because gestational gains were larger than lactational losses. Backfat tended to be lower in later parities. Body weight, heartgirth and backfat, but not condition score, declined from weaning to the third week of the subsequent gestation in both first and second litter sows. Correlations among measures of body condition were low (less than .45), except the overall correlation between body weight and heartgirth. Number of pigs born alive increased and interval to estrus decreased in later parities. There were no significant relationships between changes in body condition and rebreeding performance of sows. These results suggest that changes in body condition typically observed in sows housed in commercial production units may be too subtle to have an effect on reproductive performance.
