Soybean (Glycine max (L) Merr.) seed is an important source of oil for human consumption. Increasing the percentage of oleic acid in soybean seed oil is an important breeding objective because increasing the oleic acid content improves the oxidative stability of the oil. Extensive literature shows that temperature during seed-fill is positively correlated with the content of oleic acid in soybeans. In addition, it was shown that a maturity QTL was linked to an oleic acid QTL. The Mississippi Delta in the USA is a hot environment where soybean harvest begins in August, which is the hottest part of the season. The purpose of this research was to determine the possibility of developing both early- and late-maturing lines with consistent >?50% oleic acid content in Mississippi. We selected early and late segregants from three genetically different breeding populations also segregating for mid-oleic acid derived from crosses to germplasm N98-4445A, a non-transgenic freely available line with >?50% oleic acid. The selected lines were grown in 2 years in three trials at Stoneville, MS. Results indicated that no late-maturing lines (MG V) met the targeted mid-oleic acid level, whereas MG III and early MG IV lines with oleic acid over 50% were obtained. No maturity-alone effect on oleic acid content was observed, due to the bias of the strong negative correlation between maturity date and mean temperature during seed-fill. This study demonstrated that breeders can effectively develop early soybeans with oleic acid levels greater than 50% for the midsouthern USA.  相似文献

Previous research has demonstrated that glyphosate can affect nitrogen fixation or nitrogen assimilation in soybean. This 2-year field study investigated the effects of glyphosate application of 1.12 and 3.36 kg of ae ha(-1) on nitrogen metabolism and seed composition in glyphosate-resistant (GR) soybean. There was no effect of glyphosate application on nitrogen fixation as measured by acetylene reduction assay, soybean yield, or seed nitrogen content. However, there were significant effects of glyphosate application on nitrogen assimilation, as measured by in vivo nitrate reductase activity (NRA) in leaves, roots, and nodules, especially at high rate. Transiently lower leaf nitrogen or (15)N natural abundance in high glyphosate application soybean supports the inhibition of NRA. With the higher glyphosate application level protein was significantly higher (10.3%) in treated soybean compared to untreated soybean. Inversely, total oil and linolenic acid were lowest at the high glyphosate application rate, but oleic acid was greatest (22%) in treated soybean. These results suggest that nitrate assimilation in GR soybean was more affected than nitrogen fixation by glyphosate application and that glyphosate application may alter nitrogen and carbon metabolism.  相似文献

This paper deals with the influence of different levels of three pectins, low-methylated pectin (LMP), high-methylated pectin (HMP), and low-methylated and amidated pectin (LMA), on the in vitro gastric hydrolysis of beta-lactoglobulin (beta-lg). Proteolysis by pepsin consisted of a 2-h progressive reduction of pH. A turbidity measurement of beta-lg-pectin mixtures was carried out during the proteolysis. The influence of pectins on pepsin enzymatic activity was also evaluated. beta-Lg was resistant to peptic digestion. The presence of each of the three pectins at a concentration of 50 wt % increased the N release at all pH values considered, despite a significant inhibition of the pepsin enzymatic activity with the pectins. The turbidity of beta-lg solutions during proteolysis was reduced by the addition of pectins, because of the formation of electrostatic complexes between this protein and pectins. The increase of N release could be a false positive result due to the difficulty of precipitating protein by trichloroacetic acid because of the formation of electrostatic complexes demonstrated by the decrease of turbidity.  相似文献

In the midsouth USA, soybean is produced either under irrigated or non-irrigated conditions. The objective of this experiment was to show the utility of supplemental irrigation as an alternative to full-season and non-irrigation to achieve high yield and high seed composition. The effects of irrigation and cultivar differences on soybean yield and seed composition were conducted. Two cultivars (Dwight and Freedom) and three irrigation regimes (full-season irrigation, FS; reproductive stage/supplemental irrigation, RI; and non-irrigation, NI) were used. Protein percentage was higher in Dwight under FS and RI than NI. In Freedom, protein percentage was higher under NI than under FS and RI. Under NI, Freedom had higher protein percentage than Dwight, especially in 2004, but lower oil in 2003 and 2004. Cultivars showed significant differences in fatty acids. Yield in Freedom under FS and RI was not significantly different. Nitrogen fixation was substantially higher under NI conditions. The results indicate that irrigation management and cultivar selection significantly affect seed composition and yield. Protein increase in Freedom under non-irrigated conditions may benefit producers for high protein seed under dry-land conditions. Supplemental irrigation at the reproductive stage may be a possible alternative for full season irrigation for the cultivar Freedom. Nontarget injury from glyphosate drift is a concern among growers using non-glyphosate-resistant (non-GR) cultivars. The effects of glyphosate drift on nitrate assimilation and nitrogen fixation potential, nodule mass, and yield of non-GR soybean were assessed in a field trial at Stoneville, MS. A non-GR soybean cultivar 'Delta Pine 4748S' was treated with glyphosate at 12.5% of use rate of 0.84 kg of active ingredient/ha at 3 (V2), 6 (V7), and 8 (R2, full bloom) weeks after planting (WAP) soybean to simulate glyphosate drift. Untreated soybean was used as a control. Soybeans were sampled weekly for 2 weeks after each glyphosate treatment to assess nitrate assimilation and N2 fixation potential. Nitrate assimilation was assessed using in vivo nitrate reductase assay in leaves, stems, roots, and nodules. Nitrogen fixation potential was assessed by measuring nitrogenase activity using the acetylene reduction assay (ARA). Nitrogen content of leaves, shoots, and seed and soybean yield were also determined. In the first sampling date (4 WAP), glyphosate drift caused a significant decrease in NRA in leaves (60%), stems (77%), and nodules (50%), with no decrease in roots. At later growth stages, NRA in leaves was more sensitive to glyphosate drift than stems and roots. Nitrogenase activity was reduced 36-58% by glyphosate treatment at 3 or 6 WAP. However, glyphosate treatment at 8 WAP had no effect on nitrogenase activity. Nitrogen content was affected by glyphosate application only in shoots after the first application. No yield, seed nitrogen, protein, or oil concentration differences were detected. These results suggest that nitrate assimilation and nitrogen fixation potential were significantly reduced by glyphosate drift, with the greatest sensitivity early in vegetative growth. Soybean has the ability to recover from the physiological stress caused by glyphosate drift.  相似文献

The incidence of tuberculosis has been increasing substantially on a worldwide basis over the past decade, but no tuberculosis-specific drugs have been discovered in 40 years. We identified a diarylquinoline, R207910, that potently inhibits both drug-sensitive and drug-resistant Mycobacterium tuberculosis in vitro (minimum inhibitory concentration 0.06 mug/ml). In mice, R207910 exceeded the bactericidal activities of isoniazid and rifampin by at least 1 log unit. Substitution of drugs included in the World Health Organization's first-line tuberculosis treatment regimen (rifampin, isoniazid, and pyrazinamide) with R207910 accelerated bactericidal activity, leading to complete culture conversion after 2 months of treatment in some combinations. A single dose of R207910 inhibited mycobacterial growth for 1 week. Plasma levels associated with efficacy in mice were well tolerated in healthy human volunteers. Mutants selected in vitro suggest that the drug targets the proton pump of adenosine triphosphate (ATP) synthase.  相似文献

Soybean is one of the most important crops worldwide for its protein and oil as well as the health beneficial phytoestrogens or isoflavone. This study reports a relatively dense single nucleotide polymorphism (SNP)‐based genetic map based on ‘Hamilton’ by ‘Spencer’ recombinant inbred line population and quantitative trait loci (QTL) for seed isoflavone contents. The genetic map is composed of 1502 SNP markers and covers about 1423.72 cM of the soybean genome. Two QTL for seed isoflavone contents have been identified in this population. One major QTL that controlled both daidzein (qDZ1) and total isoflavone contents (qTI1) was found on LG C2 (Chr 6). And a second QTL for glycitein content (qGT1) was identified on the LG G (Chr 18). These two QTL in addition to others identified in soybean could be used in soybean breeding to optimize isoflavone content. This newly assembled soybean linkage map is a useful tool to identify and map QTL for important agronomic traits and enhance the identification of the genes involved in these traits.  相似文献
