ABSTRACT

The formation of phosphorus (P) compounds including iron-P, aluminum-P and calcium-P in highly weathered tropical soils can be altered upon biochar addition. We investigated the effect of corn cob biochar (CC) and rice husk biochar (RH) pyrolyzed at three temperatures (300°C, 450°C and 650°C) on phosphorus (P) fractions of three contrasting soils. A 90d incubation study was conducted by mixing biochar with soil at a rate of 1% w/w and at 70% field capacity. Sequential P fraction was performed on biochar, soil and soil-biochar mixtures. Increase in most labile P (resin-Pi, NaHCO3-Pi) and organic P fraction (NaHCO3-Po + NaOH-Po) in CC and RH biochars were inversely related to increasing temperature. HCl-Pi and residual P increased with increasing temperature. Interaction of CC and RH with soils resulted in an increase in most labile P as well as moderately labile P (NaOH-Pi) fractions in the soils. CC increased most labile P in the soils more than RH. The increase in most labile P fraction in soils was more significant at relatively lower temperatures (300°C and 450°C) than 650°C. However, the increase in HCl-Pi and residual P of the soils was more predominant at high temperature (650°C). The study suggested that biochar pyrolyzed at 300–450°C could be used to increase P bioavailability in tropical soils.  相似文献

Elm (Ulmus sp.) shoots from in vitro micropropagated plants were tested with Ophiostoma ulmi culture filtrates, at different dilutions to determine the susceptibility of different host clones to Dutch elm disease (DED). ‘Commelin’, a susceptible Dutch hybrid, showed a high wilting index but that of American elm was higher. Although U. pumila × japonica (Sapporo gold 2) was the most tolerant to spore inoculation, its wilting index was as high as that of ‘Commelin’. The absorption capacity and wilting index of the elm shoots showed consistent correlation to isolate effectiveness (as determined by inoculation experiments), to production of toxin (cerato-ulmin) and to filtrate dilution. This method could be used to assess the effectiveness of the different O. ulmi isolates and for the detection of elm genotypes that are highly susceptible to Dutch elm disease.  相似文献

Small hive beetles, Aethina tumida Murray, are parasites and scavengers of honeybee colonies, Apis mellifera L., and have become an invasive species that can cause considerable damage in its new distribution areas. An effective subspecies of Bacillus thuringiensis Berliner (=Bt) would provide an alternative to chemical control of this pest. Therefore, we tested three different Bt strains [B. thuringiensis, var. aizawai (B401®), B. thuringiensis var. kurstaki (Novodor®) and B. thuringiensis var. San Diego tenebrionis (Jackpot®)] and Perizin® (3.2% coumaphos), each applied on combs with a pollen diet fed to pairs of adult beetles. This evaluates the products for the suppression of successful small hive beetle reproduction. While none of the tested Bt strains showed a significant effect on the number of produced wandering larvae, we could confirm the efficacy of coumaphos for the control of small hive beetles. We further show that it is also efficient when applied with a lower concentration as a liquid on the combs. We suggest the continued search for efficient Bt strains naturally infesting small hive beetles in its endemic and new ranges, which may become a part of the integrated management of this pest.  相似文献

Twelve Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) seedlots from coastal British Columbia were assayed for seedborne Fusarium. All of the seedlots were contaminated with Fusarium. Percent of nonstratified seeds from individual seedlots harboring Fusarium ranged from 0.3% to 95.4%. Sixty-seven percent of the seedlots had Fusarium on less than 2% of the seeds. Post-stratification seedborne Fusarium levels were significantly less for running water imbibition compared to standing water imbibition. However, seedling growth at a container nursery was not consistently different for stratified seed imbibed initially in standing or running water. Fusarium disease symptoms were not observed in the nursery environment. The species of Fusarium isolated from seed were F. acuminatum, F. avenaceum, F. lateritium, F. moniliforme, F. oxysporum, F. poae and F. sambucinum. Twelve Fusarium isolates, comprising six species, were assessed for pathogenicity. Disease symptoms were observed after four weeks incubation and Fusarium isolates ranged in virulence from low to high. Fusarium oxysporum isolates were the most pathogenic.  相似文献

Contents of total and individual glucosinolates of mini broccoli cv. Milady and mini cauliflower cv. Clarke were assessed to determine the effect of modified atmosphere packaging on postharvest glucosinolate dynamics of mixed mini Brassica vegetables. Therefore, mixed-packaged mini broccoli and mini cauliflower stored in food trays sealed with two different microperforated biaxial-oriented polypropylene films for up to 7 days at 8 degrees C were analyzed. The results indicate that modified atmosphere at 8% O2 + 14% CO2 was a suitable gaseous combination to maintain aliphatic and indole glucosinolates in mini broccoli for 7 days after an initial decrease at 4 days. In contrast, modified atmosphere at 1% O2 + 21% CO2 resulted in the best retention of indole glucosinolates of mini cauliflower for 7 days and also of aliphatic glucosinolates after an initial decrease at 4 days. Thus, to maintain glucosinolates and external appearance and to prevent off-odor, mini broccoli and mini cauliflower should be packed separately in suitable altered gas composition.  相似文献

Preamble. This series of two papers presents the results of an interdisciplinary research project (ISIS) dealing with bioassay-directed fractionation of marine sediment extracts. Part I presents the extraction and fractionation procedure as well as the results of chemical analysis, including non-target analysis of sediments. Part II describes the results of the biotest battery in relation to chemicals possibly causing parts of the observed effects. A biotest index is used to compare the toxicities of the samples.

Goal, Scope and Background

The ecological relevance of contaminants in mixtures is difficult to assess, because of possible interactions and due to lacking toxicity data for many substances present in environmental samples. Marine sediment extracts, which contain a mixture of environmental contaminants in low concentrations, were the object of this study. The extracts were investigated with a set of different biotests in order to identify the compound or the substance class responsible for the toxicity. For this goal, a combination of biotests, biotest-directed fractionation and chemical analysis has been applied. Further on, a strategy for the development of a biotest index to describe the toxicity of the fractions without a prior ranking of the test results is proposed. This article (Part II) focuses on the biological results of the approach.

Methods

The toxicological potential of organic extracts of sediments from the North Sea and the Baltic Sea was analyzed in a bioassay-directed fractionation procedure with a set of biotests: luciferase reporter gene assays on hormone receptor and Ah receptor, arabinose resistance test, fish embryo test (Danio rerio), comet assay, acetylcholinesterase inhibition test, heat-shock protein 70 induction, oxidative stress and luminescence inhibition test (Vibrio fischeri). The test results provided the basis for the calculation of a biotest index by factor analysis to compare the toxicity of the samples and fractions.

Results and Discussion

Results of 11 biotests on different fractionation levels of the samples were described and discussed with regard to the occurrence of contaminants and their toxic potentials. Polychlorinated biphenyls, polycyclic aromatic hydrocarbons, quinones, brominated indoles and brominated phenols were in the focus of interest. A biotest index was constructed to compare the toxic responses in the samples and to group the biotest results.

Conclusion

The procedure presented in this study is well suited for bioassay-directed fractionation of marine sediment extracts. However, in relatively low contaminated samples, high enrichment factors and sufficient fractionation is necessary to allow identification of low concentrations of contaminants which is required to link effects and possible causes. In the present case, the relation between substances and effects was difficult to uncover due to relatively low concentrations of pollutants compared to the biogenic matrix and to the remaining complexity of the fractions. The results, with respect to the brominated phenols and indoles in the samples, highlight the successful use of bioassay directed fractionation in the case of high concentrations and high toxicity.

Recommendation and Outlook

In general, it has been shown that a marine risk assessment requires focusing on the input of diffuse sources and taking into account the fact of mixture toxicity. Effects resulting from biogenic substances will make the assessment of the influence of anthropogenic substances even more difficult.  相似文献

Cercospora leaf spot (CLS) poses a high economic risk to sugar beet production due to its potential to greatly reduce yield and quality. For successful integrated management of CLS, rapid and accurate identification of the disease is essential. Diagnosis on the basis of typical visual symptoms is often compromised by the inability to differentiate CLS symptoms from similar symptoms caused by other foliar pathogens of varying significance, or from abiotic stress. An automated detection and classification of CLS and other leaf diseases, enabling a reliable basis for decisions in disease control, would be an alternative to visual as well as molecular and serological methods. This paper presents an algorithm based on a RGB‐image database captured with smartphone cameras for the identification of sugar beet leaf diseases. This tool combines image acquisition and segmentation on the smartphone and advanced image data processing on a server, based on texture features using colour, intensity and gradient values. The diseases are classified using a support vector machine with radial basis function kernel. The algorithm is suitable for binary‐class and multi‐class classification approaches, i.e. the separation between diseased and non‐diseased, and the differentiation among leaf diseases and non‐infected tissue. The classification accuracy for the differentiation of CLS, ramularia leaf spot, phoma leaf spot, beet rust and bacterial blight was 82%, better than that of sugar beet experts classifying diseases from images. However, the technology has not been tested by practitioners. This tool can be adapted to other crops and their diseases and may contribute to improved decision‐making in integrated disease control.  相似文献

In honeybee, Apis mellifera, colonies infested with larval and adult small hive beetles (SHB), hive material, and in particular honey, tends to ferment, probably due to SHB-associated yeasts such as the predominant Kodamaea ohmeri. Here, we test the effects of organic acids on K. ohmeri and on SHB-infested honey/pollen combs. Organic acids were applied at standard concentrations used by beekeepers to control other pests. In laboratory tests, the growth of K. ohmeri was significantly inhibited by lactic, formic and acetic acids. Treatments of SHB-infested honey/pollen combs (N = 18 colonies) with acetic acid significantly increased mortality of adult SHB and treatments with formic acid significantly reduced larval infestation. Our data suggest that treatment of honeybee colonies and storage rooms with organic acids could also help in reducing SHB damage.  相似文献
