Effects of a topical anaesthetic formulation and systemic carprofen, given singly or in combination, on the cortisol and behavioural responses of Merino lambs to castration

Objective   To determine the effectiveness of a topical anaesthetic formulation (Tri-Solfen) with or without the administration of a non-steroidal anti-inflammatory drug (carprofen) on the pain and distress response associated with ring or surgical castration of ram lambs.
Procedures   Merino ram lambs (n = 78) were allocated to 10 treatment groups: 4 groups of knife-castrated lambs and 4 groups of ring-castrated lambs received carprofen (4 mg/kg SC) and Tri-Solfen; 2 control groups (sham) received carprofen at 0 or 4 mg/kg SC. Measurements included plasma cortisol and haptoglobin concentrations, haematology, and behaviour, including posture.
Results   Knife-castrated lambs had higher peak cortisol and integrated cortisol responses for the first 6 h after treatment and greater concentration s of circulating acute phase proteins than ring-castrated lambs, both of which were significantly different from the sham controls. Tri-Solfen applied to the knife castration wound significantly reduced both the peak plasma cortisol concentration and the integrated cortisol response for the first 6 h and improved lying behaviour in the first 12 h. Carprofen reduced the cortisol response to knife castration at 30 min, but elevated the cortisol responses at 24 and 48 h. Carprofen nearly halved the number of acute pain behaviours associated with ring castration. There were no significant additive or synergistic effects from combining the analgesic treatments. Tri-Solfen applied to the tail wound provided no detectible benefits during ring castration + tail docking.
Conclusions   The physiological and behavioural responses suggest that ring castration has less impact on the lamb than knife castration. The specific analgesic treatments can provide modest amelioration of the pain and discomfort associated with castration. Alternative doses or application methods may enhance their efficacy.     

An assessment of the nature conservation value of Majorca

In spite of increasing numbers of tourists each year, Majorca retains a variety of natural and semi-natural vegetation types. These are principally in the limestone sierra and around the coast. Of the fairly numerous species of endemic plants which occur, some are frequent. The Balearic Islands are also an important staging-post for migrant birds, and the wetlands of Majorca are the most valuable habitat from the ornithological standpoint. This paper summarizes the major vegetation types of Majorca and discusses the contribution made to the conservation value of the island by its 21 species of vascular plants that are recognized as endemic to the Balearic Islands and in 13 cases are exclusively Mallorcan. Finally, the conservation threats arising from urbanization, from indirect effects of development, and from changes in agricultural practice, are briefly discussed.     

Experimental infection of normal and immunosuppressed pigs with Pseudomonas pseudomallei

A single dose of 5 x 10(8) bacilli of Pseudomonas pseudomallei by intratracheal injection resulted in acute (21 cases) or chronic (19 cases) melioidosis in 40 of 48 pigs. Fifteen (10 acute and 5 chronic) had been immunosuppressed by cyclophosphamide before inoculation. The major clinical signs were initial fever, marked neutrophilia and, in the acute cases, respiratory distress. There were no signs of the nasal and ocular discharge, paresis or diarrhoea seen in acute cases in south-east Asia. The cyclophosphamide treatment caused a significant decrease in the neutrophil count by 7 d after inoculation in all 15 immunosuppressed pigs, and all were culture positive at necropsy. Eight of the 33 non-treated pigs were culture negative at necropsy. Pigs overcoming the initial phase of infection had more abscess-like nodules that were bacteriologically sterile at necropsy than the pigs with acute cases of melioidosis. P. pseudomallei was isolated predominantly from the spleen, lungs and the injection site. Although only one strain was used in this study, it is likely that Australian strains of P. pseudomallei are not as virulent as the south-east Asian isolates.     

Virginia barrier island configuration: a reappraisal

The 12 Virginia barrier islands are divided into three groups on the basis of historical retreat rates and characteristics. The growth and reversal of inlet offsets is a long-term event that results from the operation of known coastal processes. Both geomorphic evidence and wave refraction analyses indicate that shoreline irregularity will diminish with time. There is no basis for the prediction (or expectation) that cape-like features will evolve along this shoreline.     

Structure of the nucleotide activation switch in glycogen phosphorylase a

Adenosine monophosphate is required for the activation of glycogen phosphorylase b and for release of the inhibition of phosphorylase a by glucose. Two molecules of adenosine monophosphate (AMP) bind to symmetry related sites at the subunit interface of the phosphorylase dimer. Adenosine triphosphate (ATP) binds to the same site, but does not promote catalytic activity. The structure of glucose-inhibited phosphorylase a bound to AMP and also of the complex formed with glucose and ATP is described. Crystallographic refinement of these complexes reveals that structural changes are associated with AMP but not ATP binding. The origin of these effects can be traced to different effector binding modes exhibited by AMP and ATP, respectively. The conformational changes associated with AMP binding traverse multiple paths in the enzyme and link the effector and catalytic sites.     

Microspectrophotometry of photoreceptor organelles from eyes of the prawn Palaemonetes

Microspectrophotometric measurements of individual dark-adapted rhabdoms of the prawn Palaemonetes vulgaris reveal the presence of two light-sensitive pigments. A pigment with maximum absorbancy at 555 nanometers is converted by light to a long-lived intermediate with wavelength of maximum absorbancy at 496 nanometers. A second pigment with wavelength of maximum absorbancy at 496 nanometers bleaches in the light, seemingly without forming detectable products at wavelengths longer than 375 nanometers. Both pigments occur in each layer of microvilli.     

Domain separation in the activation of glycogen phosphorylase a

The crystal structure of glycogen phosphorylase a complexed with its substrates, orthophosphate and maltopentaose, has been determined and refined at a resolution of 2.8 angstroms. With oligosaccaride bound at the glycogen storage site, the phosphate ion binds at the catalytic site and causes the regulatory and catalytic domains to separate with the loss of stabilizing interactions between them. Homotropic cooperativity between the active sites of the allosteric dimer results from rearrangements in isologous contacts between symmetry-related helices in the subunit interface. The conformational changes in the core of the interface are correlated with those observed on covalent activation by phosphorylation at Ser14 (phosphorylase b----a).
