The ring‐tailed coati (Nasua nasua) is a procyonid whose population is in sharp decline. Therefore, studies are needed to better understand the reproduction of this animal. For this reason, this study aimed to evaluate the morphology, morphometry and sperm ultrastructure of ring‐tailed coati sperm. Four captive adult males were used for this study. Slides stained with Bengal Rose were used for the morphometric and morphologic analyses. The length and width of the head were measured, as well as the length of the midpiece and tail and the total length of the sperm. Scanning electron microscopy and transmission electron microscopy were used for the ultrastructural analyses. The most obvious morphological abnormalities observed were coiled tails (6.1 ± 8.7%) and the lack of acrosomes (5.4 ± 4.4%). Regarding the morphometry, the measurements of the head (length × width), midpiece (length) and tail (length) were (mean ± SD) 6.2 ± 0.4 × 8.1 ± 0.6 μm, 14.1 ± 0.5 and 63.9 ± 4.1 μm, respectively, and the total length of the sperm was 86.1 ± 4.3 μm. Through electron microscopy, the presence of electron‐lucent points in the nucleus and the presence of approximately 55 mitochondrial spirals in the midpiece were identified. The data obtained in this study provide detailed information on the sperm characteristics of coatis and may inform future research on germplasm conservation, both for this species and other threatened procyonids.  相似文献

The effect of Himalayan balsam (Impatiens glandulifera) on survival and growth of naturally regenerated silver birch (Betula pendula) and planted Norway spruce (Picea abies) and silver fir (Abies alba) seedlings was studied in a weeding experiment over 3 years. Three different treatments were applied: control, mowing, and hand weeding by pulling out the entire plant. There were no consistent treatment effects on height and diameter of the tree seedlings. The coverage of Rubus fruticosus had a negative impact on diameter increment of Norway spruce and silver fir. As opposed to growth, treatment effects on seedling survival could be found for planted Norway spruce and silver fir. However, it is very likely that these effects, namely higher seedling survival after mowing, have to be attributed to the control of bramble (Rubus fruticosus) rather than to that of Himalayan balsam. It is concluded that Himalayan balsam is not able to seriously affect the growth of already established seedlings.  相似文献

Recent research shows that most soils are more or less water repellent. Already subcritical water repellency may cause incomplete soil wetting and preferential flow. Both processes potentially reduce the residence time of water and solutes in the vadose zone, resulting in an enhanced risk of groundwater contamination. The objective of the present paper is, therefore, to evaluate the impact of reduced soil wettability on the soil water infiltration rate and to investigate the tendency towards preferential flow with the analysis of the immobile water content in the infiltration zone. In november 2002, a field experiment was done in a coniferous forest, 30 km N of Hannover, Germany. Soil hydrophobicity was quantified by measuring the contact angles. The hydraulic conductivity of the podsolic sandy soil was measured depth‐dependent with a double‐ring tension infiltrometer in three soil horizons. To quantify possible preferential‐flow effects, a LiBr‐Tracer was added to the infiltrating water to evaluate the mobile water‐content fraction after infiltration. Additionally, infiltration rates of water were compared with infiltration rates of ethanol which were determined after water infiltration at the same locations. Results show that the actual water repellency of field‐moist soil was mainly subcritical (contact angle <90°). Water infiltration rates were reduced due to subcritical repellency by a factor of 3–170 compared with ethanol infiltration rates (exclusion of wetting effects). This spatially variable infiltration behavior was not clearly reflected neither by the small‐scale contact‐angle measurements nor by the analysis of the average immobile soil water content in the infiltration zone. We conclude that this specific infiltration behavior of water caused by small‐scale wettability effects may temporarily reduce the local connectivity of water‐flow pathways.  相似文献

Wettability parameters determined for individual soils often show a considerable variation depending on the kind of sample (aggregated or homogeneous material) and the method used. To investigate the causes of this variation, we assessed wettability of both intact and crushed aggregates and bulk soil using different methods. Wettability of intact aggregates was characterized by a modified technique where the specific infiltration rates of water and a completely wetting liquid were used to define a repellency index. Contact angles were determined on crushed aggregates and bulk soil using the Wilhelmy plate and capillary rise methods. The repellency index was found to be sensitive to slight differences in wettability and was in good agreement with Wilhelmy plate contact angles. Contact angles measured with the capillary rise method showed a strong deviation from those determined with the Wilhelmy plate method. This can be ascribed to the underlying assumptions of the capillary rise method (i.e. cylindrical and parallel capillaries) resulting in an over‐estimation of contact angle, particularly for the small‐sized particle fraction because of the impact of inertia and pore structure. No significant differences were found between intact and crushed aggregates whereas the bulk soil was slightly more water‐repellent, probably because of a somewhat larger organic carbon content. We conclude that the contact angle determined by the Wilhelmy plate method and the repellency index are appropriate parameters for characterizing soil water repellency because they detected small changes in wettability over a wide range extending from subcritical water repellency to hydrophobicity.  相似文献

The protective impact of aggregation on microbial degradation through separation has been described frequently, especially for biotically formed aggregates. However, to date little information exists on the effects of organic‐matter (OM) quantity and OM quality on physical protection, i.e., reduced degradability by microorganisms caused by physical factors. In the present paper, we hypothesize that soil wettability, which is significantly influenced by OM, may act as a key factor for OM stabilization as it controls the microbial accessibility for water, nutrients, and oxygen in three‐phase systems like soil. Based on this hypothesis, the first objective is to evaluate new findings on the organization of organo‐mineral complexes at the nanoscale as one of the processes creating water‐repellent coatings on mineral surfaces. The second objective is to quantify the degree of alteration of coated surfaces with regard to water repellence. We introduce a recently developed trial that combines FTIR spectra with contact‐angle data as the link between chemical composition of OM and the physical wetting behavior of soil particles. In addition to characterizing the wetting properties of OM coatings, we discuss the implications of water‐repellent surfaces for different physical protection mechanisms of OM. For typical minerals, the OM loading on mineral surfaces is patchy, whereas OM forms nanoscaled micro‐aggregates together with metal oxides and hydroxides and with layered clay minerals. Such small aggregates may efficiently stabilize OM against microbial decomposition. However, despite the patchy structure of OM coating, we observed a relation between the chemical composition of OM and wettability. A higher hydrophobicity of the OM appears to stabilize the organic C in soil, either caused by a specific reduced biodegradability of OM or indirectly caused by increased aggregate stability. In partly saturated nonaggregated soil, the specific distribution of the pore water appears to further affect the mineralization of OM as a function of wettability. We conclude that the wettability of OM, quantified by the contact angle, links the chemical structure of OM with a bundle of physical soil properties and that reduced wettability results in the stabilization of OM in soils.  相似文献

The effect of rotaviruses and enterotoxigenic Escherichia coli administered in various sequences to cesarean-derived, colostrum-deprived calves was studied using light and electron microscopy. The structure of the lymphoid tissue in the ileum, the number of mitoses in the crypts, number of intraepithelial lymphocytes, and enzyme histochemistry (alkaline phosphatase, acid phosphatase, succinic dehydrogenase, beta-galactosidase, and leucinaminopeptidase) of the ileal dome epithelium were evaluated. The area of lymphoid follicles in Peyer's patches of the ileum was investigated morphometrically. Monoinfections with either rotavirus or enterotoxigenic E. coli induced a significant increase in lymphoid follicle area, but did not affect dome epithelial cells. Dual infections did not consistently affect the follicle area, but the number of intraepithelial lymphocytes and the mitotic indices exceeded those of comparable monoinfections. Changes in activity of enzymes in the ileal dome epithelial area were minor.  相似文献

Two strains of E. coli O139:K12 (B):H1 were compared in vitro and in the intestinal environment. Both strains colonized the small intestines of experimentally inoculated pigs and exhibited in vivo a similar relationship to the microvillus border as enterotoxigenic E. coli (ETEC). Strain 107/86 grown on blood agar expressed numerous long flexible non-haemagglutinating fimbriae which were antigenically distinct from the known fimbriae of porcine ETEC. It adhered in vitro to porcine enterocyte brush border fragments. Strain 124/76 grown on blood agar was devoid of fimbriae and did not adhere to brush border fragments. However, fimbriae morphologically and antigenically indistinguishable from those of strain 107/86 were detected in the intestinal environment by direct immunofluorescence and by immuno electron microscopy.  相似文献

Long‐term capillary rise experiments (0 to about 89 000 hour) were performed at 19°C on homogenized and heat‐treated podsolic forest top‐ and subsoil samples. These were packed into columns, the bases of which were then partially immersed, at constant depth, in water reservoirs to simulate a constant water table. Selected columns were equipped with tensiometer and TDR probes. Other columns were removed at prescribed times and divided into 2‐cm horizontal segments whose volumetric water contents were determined. The degree of saturation was then estimated by comparison with the capillary rise in duplicate arrangements of samples immersed in ethanol. It was found that the heat treatments conferred increased water repellency (WR) on the soil, which increased with temperature (significantly so at greater than 60°C). This had a profound effect on the capillary rise characteristics and development of water content in the soil behind the wetting front, indicating an effective, albeit slow, reduction in effective WR. This has implications for hydraulic modelling of soils with significant WR and demonstrates that sub‐surface WR exerts a significant influence on capillary rise from a water table and suggests that commonly used indicators of surface WR using droplet tests may not be useful for such modelling purposes.  相似文献

Objectives

To describe colostrum management practices carried out in northern Victorian dairy herds and to identify weaknesses in these areas that may affect calf health and welfare by comparing the results with the current industry recommendations

Methods

A questionnaire to obtain information about colostrum management and calf‐rearing practices was sent to commercial dairy farming clients of Rochester Veterinary Practice between June and September 2013. The questionnaire consisted of a general herd overview and colostrum harvesting practices.

Results

The response rate was 39% (58/150). Many dairy producers were not meeting the current industry recommendations in the following areas: (1) time of removal calf from the dam, (2) relying on calf suckling colostrum from the dam to achieve adequate passive transfer, (3) failing to supplement calves with colostrum, (4) feeding inadequate volumes of colostrum, (5) delayed colostrum harvesting, (6) pooling of colostrum, (7) failing to objectively assess colostrum quality or relying on visual assessment and (8) storing colostrum for a prolonged periods of time at ambient temperatures.

Conclusion

The results from this survey highlight the need for greater awareness of industry standards for colostrum management and feeding hygiene.
