Three-year-old, non-lactating and non-pregnant Merino ewes, raised on pasture under a program of strategic treatment with anthelmintic and found to be extremely resistant to "trickle" infection with Haemonchus contortus, were given single-dose infections with either H. contortus or Trichostrongylus colubriformis or both species together. The purpose was to ascertain the intensity of protective immunity against the 2 parasites in sheep with immunity acquired from a presumably slight exposure to infection. To provide a criterion, some infected ewes were immunosuppressed with corticosteroid, dexamethasone. Untreated ewes were extremely resistant to challenge infection with either 15,000 or 150,000 H. contortus or 15,000 T. colubriformis. Surprisingly, when mixed infection was given, egg counts for H. contortus were significantly elevated compared with infection by that species alone. Antibody to antigens from infective larval and adult H. contortus was measured in serum by enzyme-linked-immunosorbent assay (ELISA) during the course of infection. Serum titres against larval antigens were significantly depressed when infections with either H. contortus or T. colubriformis were permitted by immunosuppression with dexamethasone, whereas those against adult antigen were depressed when infection with T. colubriformis was permitted.  相似文献

BACKGROUND

Monitoring is an integral component of integrated pest management (IPM) programmes used to inform crop management decisions. Vine weevil, Otiorhynchus sulcatus F. (Coleoptera: Curculionidae), continues to cause economically significant losses in horticultural crops due to an inability to reliably detect the presence of this species before crop damage occurs. To improve vine weevil monitoring we investigated the behavioural responses of adult vine weevils to visual (monitoring tool shade/colour, height and diameter as well as the effect of monitoring tool and plant density) and olfactory (host plant and conspecifics) cues under glasshouse conditions.

RESULTS

Monitoring tool shade, height and diameter all influenced monitoring tool efficacy, with individuals exhibiting a preference for black, tall and wide monitoring tools. The total number of individuals recorded in monitoring tools increased with monitoring tool density. By contrast, plant density did not influence the number of individuals recorded in monitoring tools. Yew-baited monitoring tools retained a larger number of individuals compared to unbaited ones. Similarly, more vine weevils were recorded in monitoring tools baited with yew and conspecifics than in unbaited monitoring tools or those baited with only yew. Baiting monitoring tools with conspecifics alone did not enhance the number of vine weevils recorded in monitoring tools.

CONCLUSIONS

Our study confirms that visual and olfactory cues influence vine weevil behaviour. This provides information on key factors that influence vine weevil monitoring tool efficacy and can be used to inform the development of a new monitoring tool for this pest. © 2023 The Authors. Food quality aspects of farmed turbot (Psetta maxima) were compared following two methods of slaughter: the current commercial method, by immersion in an ice slurry, which is then dewatered after approximately 20 min, or by first humanely, electrically stunning the fish using a prototype commercial stunner, before immersion in an ice slurry, which is dewatered after 20 min. Quality was assessed for up to 10 days of storage on ice following slaughter. No differences were found between the slaughter methods in terms of an overall carcass quality: overall appearance, haemorrhage, damage, burst gall bladder, staining of the body cavity by leakage from the gut or damage to the spine. No detectable difference was found between the treatments using the industry standard freshness scoring system, the Quality Index Method. Both groups of fish were classified as ‘Fresh’ after 10 days of storage on ice. Using objective measurements of colour, no differences between fish from either treatment were found in fillet colour. Changes in flesh pH were similar in electrically stunned and traditionally killed fish with a mean pH (±SE) at 2 h post‐mortem of 6.80±0.027 declining to 6.44±0.032 at 24 h post‐mortem. Humane electrical stunning of turbot at slaughter neither detectably improved nor decreased product quality as measured between 1 and 10 days of storage on ice.  相似文献

The leaf area index (LAI) of boreal forest can be estimated using reflected radiation recorded by satellite sensors. Measurements of visible and near infrared radiation are commonly used in the normalized difference vegetation index (NDVI) to estimate LAI. However, research, mainly in tropical forest, has demonstrated that LAI is related more closely to radiation of middle infrared wavelengths than of visible wavelengths. This paper derives a vegetation index, VI3, based on radiation from vegetation recorded at near and middle infrared wavelengths. For a boreal forest canopy, the relationship between VI3 and LAI was observed to be much stronger than that between NDVI and LAI. In addition, the LAI estimated using VI3 accounted for about 76% of the variation in field estimates of LAI, compared with about 46% when using the NDVI. We conclude that information provided by middle infrared radiation should be considered when estimating the leaf area index of boreal forest.  相似文献

Soil surface CO(2) flux (F(s)) is the dominant respiratory flux in many temperate forest ecosystems. Snowpacks increase this dominance by insulating the soil against the low temperature to which aboveground components are exposed. However, measurement of F(s) in winter may be impeded by snow cover. Likewise, developing annual F(s) models is complicated by seasonal variation in root and microbial metabolism. We compared three methods of measuring sub-snow F(s): (1) dynamic chamber measurements at the upper snowpack surface (F(snow)), (2) dynamic chamber measurements at the soil surface via snowpits (F(soil)), and (3) static estimates based on measured concentrations of carbon dioxide ([CO(2)]) and conductance properties of the snowpack (F(diffusional)). Methods were compared at a mid-elevation forest in northeastern Washington, a mid-elevation forest in northern Idaho, and a high-elevation forest and neighboring meadow in Wyoming. The methods that minimized snowpack disturbance, F(diffusional) and F(snow), yielded similar estimates of F(s). In contrast, F(soil) yielded rates two to three times higher than F(snow) at the forested sites, and seven times higher at the subalpine meadow. The ratio F(soil)/F(snow) increased with increasing snow depth when compared across all sites. Snow removal appears to induce elevated soil flux as a result of lateral CO(2) diffusion into the pit. We chose F(snow) as our preferred method and used it to estimate annual CO(2) fluxes. The snowpack was present for 36% of the year at this site, during which time 132 g C m(-2), or 17% of the annual flux, occurred. We conclude that snowpack CO(2) flux is quantitatively important in annual carbon budgets for these forests and that the static and dynamic methods yield similar and reasonable estimates of the flux, as long as snowpack disturbance is minimized.  相似文献

Field-trapping experiments with synthetic 2,7-nonadiyl dibutyrate, the female-produced sex pheromone of the orange wheat blossom midge, Sitodiplosis mosellana (Géhin), demonstrated that pheromone traps were highly attractive to males and caught very few non-target organisms. Different formulations of pheromone were tested to identify the optimum release rate and dispenser type for use in pheromone traps in the UK. Key findings were that racemic pheromone was as effective as enantiomerically pure (2S,7R)-2,7-nonadiyl dibutyrate, that release rates higher than 0.5 microg day(-1) were not necessary and that the optimal formulation was a 1 mg pheromone loading in a rubber septum. Pheromone traps gave a reliable indication of peak midge emergence, onset of flight and abundance of midges throughout the season. A strong correlation between maximum trap catch and crop infestation levels was obtained.  相似文献

Food irradiation has been acommercially viable postharvest technology fornearly 50 years (the actual idea of usingionizing radiation to extend the shelf-life offoods is over a century old), yet it has beenused only occasionally and sporadically.Interviews with reporters and the sources theyused at a Louisiana newspaper and a Floridanewspaper uncovered three cultural spherespresent in the debate over this post harvesttechnology – food, science/technology, andjournalism. Each of these spheres were pointsof contention for reporters and sources, andthis has had an affect on the adoption of thetechnology among those involved in the foodindustry. Interviews with both reporters andthe individuals they relied on as sourceselucidate how different issues encompassedwithin different cultural spheres have beenlinked to this post harvest technology, andhave been used to help shape this debate. Thesecultural spheres offer various groups power tocontrol relations with reporters, though thatpower can be usurped by others, including thereporters themselves. Interviews with reportersand their sourses may help us understand howthe values attached to cultural spheres aremobilized by various groups to make sense of acontroversial topic, and how those groups gainentrance to public arenas and are then able tomaintain their presence.  相似文献

If they could be easily exfoliated, layered materials would become a diverse source of two-dimensional crystals whose properties would be useful in applications ranging from electronics to energy storage. We show that layered compounds such as MoS(2), WS(2), MoSe(2), MoTe(2), TaSe(2), NbSe(2), NiTe(2), BN, and Bi(2)Te(3) can be efficiently dispersed in common solvents and can be deposited as individual flakes or formed into films. Electron microscopy strongly suggests that the material is exfoliated into individual layers. By blending this material with suspensions of other nanomaterials or polymer solutions, we can prepare hybrid dispersions or composites, which can be cast into films. We show that WS(2) and MoS(2) effectively reinforce polymers, whereas WS(2)/carbon nanotube hybrid films have high conductivity, leading to promising thermoelectric properties.  相似文献
