The expression of growth factors was evaluated immunohistochemically in normal and cystic ovaries of sows. The immunohistochemically stained area (IHCSA) was quantified by image analysis to analyse the expression of these proteins in the follicular wall of secondary, tertiary and cystic follicles. IGF‐I immunoreactivity was strong in the granulosa cell layer (GC), moderate in the theca interna (TI) and mild in the theca externa (TE) of the normal follicles. There was severe reduction of the labelling to IGF‐I in the GC of the follicular and luteinized cysts. In the normal follicles, the reactivity for IGF‐II was very similar to pattern noted in IGF‐I. There was reduction of the IHCSAs in the GC of the follicular and luteinized cysts, but the decrease was not significant. The staining of the IGF‐II in the TI and TE of the cysts was increased, in comparison with normal follicles. The IHCSAs for VEGF were higher in the GC and TE of the normal follicles in contrast to TI, but this difference was noted only in the tertiary follicle. The VEGF reactivity increased in the GC of the cysts, in relation to normal follicles. The results of the current study show that the formation of ovarian cysts in sows is associated with alterations in the immunohistochemical expression of some growth factors. 相似文献

Transverse drying shrinkage was measured at microscopic and mesoscopic levels in poplar wood characterised by an increasing
growth strain (GS), from normal to tension wood. Results show that: (a) the drying shrinkage, measured as a relative thickness
decrease, was significantly higher for G-layer (GL) than for the other layers (OL), GL shrinkage was not significantly correlated
with GS, and OL shrinkage was negatively correlated with GS. (b) In gelatinous fibre (G-fibre), lumen size increased during
drying and this increase was positively related with GS, but in normal wood fibre, lumen size decreased during drying. These
findings suggest that GL shrank outwards (i.e., its internal perimeter increases), so that its shrinkage weakly affected the
total cell shrinkage and the mesoscopic shrinkage was controlled by the OL shrinkage which shrank inwards (i.e., its external
perimeter decreases). (c) Measurements done on 7 × 7 mm2 thin sections evidenced a negative correlation between transverse
shrinkage and GS, significant in T direction but weak in R direction. These observations at both levels allow to discuss the

The purpose of this study was to estimate the expression and relative amounts of estrogen (ER) and progesterone receptors (PR) and their isoforms as well as heat shock protein 70 (HSP70) in ovaries of rats with induced cystic ovarian disease (COD). Primary, secondary, tertiary, atretic and cystic follicles were evaluated by immunohistochemistry and total ovarian proteins were analyzed by Western blot. In the granulosa layer, growing and cystic follicles in the treated group have a higher expression of ERα than growing follicles of control individuals. In the theca interna layer, tertiary follicles presented a significantly higher expression of ERα in the treated group. An increase in total ERα protein was detected in the treated group. Granulosa cells of all growing, atretic and cystic follicles show a lower expression of ERβ in animals with COD, and the total protein expression of ERβ was lower in this group. The expression of PR was lower in the granulosa cell layer of tertiary and cystic follicles in treated animals, and theca interna layer had less intense immunostaining in this group. Although there were no differences in the expression of PR-B by Western blotting, the expression of PR-A was higher and the expression of PR-C was smaller in the treated group. An intense HSP70 immunostaining was observed in the cells of cystic follicles. By Western blotting, higher protein expression of HSP70 was detected in the ovarian samples of the control group than those of the treated ones. Ovaries of animals with COD exhibited an altered steroid receptor expression and subtype balance as compared with control animals, and an increase in HSP70 immunoexpression. 相似文献

Soil pH, soil organic matter (SOM), dissolved organic carbon (DOC) and total trace metal concentration (M(tot)) control the solubility of metals in the soil. Several regression models have included these soil chemical variables for the prediction of metal solubility and free metal ion (FMI) concentrations in contaminated soils. We hypothesize that models developed on contaminated soils (after optimization of the coefficients) can be used on samples from uncontaminated sites. Soil samples were collected from unpolluted agricultural and forest soils located in Eastern Croatia and extracted with water to determine the concentrations of Cd, Cu, Pb and Zn. We used these data to test the applicability of three regression models on existing conditions under different land uses. The same predictors issued in the three models and the same regression coefficients were utilized in the present study. The results showed a good correlation between the observed and predicted values of metal solubility. However, the models overestimate the total solution concentration (M(sol)) and the concentrations of free metal ions (FMI) in solution, and therefore the same regression coefficients were optimized to fit our own observations. This was found to be very successful. The results showed that pH and DOC played a very important role in controlling metal solubility, while SOM and CEC were somewhat less significant. The impact of total soil concentration of metals (M(tot)) was rather minor. However, we feel that to carry out good predictions of M(sol) and FMI, the M(tot) is needed in such regression models. 相似文献

Addition of granulosa cells (GC) (1.10(6)/ml) affected maturation of non-ovulatory oocytes of cattle and caused delay in nuclear maturation (60.8 versus 35.3 or 37.1 percent). In vitro use of pre-incubated GC in the process of maturation led to higher rates of fertilisation (49.4 versus 25.2 or 21.0 percent) and of cleavage (29.6 versus 13.3 or 9.1 percent). No explanation has yet been found for different oocyte behaviours in response to use of fresh or pre-incubated GC. Transfer of seven embryos from maturation with GC as well as in vitro fertilisation and development to four recipients resulted in the birth of one female calf. 相似文献

The main idea of this article is to present various perspectives in order to analyze the recent crisis concerning the agriculture-based rural societies in the developed capitalist communities. In all of these countries there is a production crisis, resulting in too much food. But this is also an ideological crisis, because the consumer thinks that the food is produced at too high a price. And it is a political crisis as well because a major part of the voters think subsidies and trade barriers are too high. The paper argues that beneath the present agricultural and rural policy crisis lies the failure of three great projects of our time: 1) The project of natural science; 2) The project of liberal capitalism; and 3) The project of scientific socialism. The failure of these three projects has to do with the breakdown of the positivist idea of modernization. Modernization theory was partly wrong because it overlooked the persistence of locally based life modes. Those life modes must be understood before a sustainable rural development is found. The article reviews some contemporary social science perspectives that have recently been developed to grasp the fundamental changes of today's rural societies. Based on those perspectives and primarily the life mode perspective, five key elements that are essential to analyze if we want to understand future development are isolated: food production, resources, space, social diversity, and culture.Reidar Almås is a Professor of Sociology at the University of Trondheim, Norway, where he directs the Center for Rural Research. This article raises the issue of the extent to which a single nation can develop a “national agricultural policy,” pursuing internal goals in agrarian development, goals that vary significantly from those of other industrialized countries. What are the conflicts arising from such a policy and how do these conflicts interfere with the general agricultural crisis of these countries? The Norwegian case is explored as an example of a blend of social-democratic and center-populist agricultural policies. The decision in 1975 by the Norwegian Parliament to establish the income level of farmers at the same level as workers is seen as an experiment. As agricultural policy, this new departure represented a deviation from the earlier policies of rationalization. The Social Democrats developed their new position as a result of internal and external pressure. A series of reforms financed by the new state incomes from oil production were undertaken, of which income equalization and a farm relief service were the most important. Most Norwegian farmers speak of the reform years as “the good years of escalation.” Equity was reached in 1982, but nevertheless several problems emerged: 1) the escalation of subsidized incomes led to overproduction and decreasing incomes from the market (after 1982), 2) the large budgetary transfers to agriculture caused a legitimation crisis, fueling the constituency of the right-wing Progress Party, 3) the large subsidies to agriculture contributed to the fiscal crisis of the state, 4) the problems mentioned above, contributed to the general crisis of the Norwegian Social Democracy which finds itself with dwindling support. The likely outcome of the present Norwegian farm crisis is the implementation of an agricultural policy that will protect the most vulnerable sector and open the more competitive sector to market forces. 相似文献

Development of new semen cryopreservation techniques improving sperm survival and ensuring availability of viable spermatozoa for a prolonged time‐period after AI is promising tools to reduce sensitivity of timing of AI and enhance overall fertility. The SpermVital® technology utilizes immobilization of bull spermatozoa in a solid network of alginate gel prior to freezing, which will provide a gradual release of spermatozoa after AI. The objective of this study was to compare post‐thaw sperm quality and in vitro sperm survival over time of Norwegian Red bull semen processed by the SpermVital® (SV) technology, the first commercialized production line of SpermVital® (C) and by conventional procedure applying Biladyl® extender (B). Post‐thaw sperm motility was not significantly different between SV, C and B semen (p > .05). However, sperm viability and acrosome intactness were higher for SV than C and B semen (p < .05). Small differences in DNA quality were observed (p < .05). Sperm viability after storage in uterus ex vivo was higher for SV than for C semen (p < .05). Furthermore, sperm survival in vitro over time at physiological temperature was significantly higher for SV semen than C semen as well as B semen during the incubation period of 48 hr (p < .05). In conclusion, the SpermVital® technology is improved and is more efficient in conserving post‐thaw sperm quality and results in higher sperm viability over time in vitro for SV than for C and B semen. 相似文献

Pigs were transported from several breeding facilities at the age of 10-12 weeks and regrouped in a fattening farm, specialized in breeding pigs for subsequent slaughter. Blood samples were obtained from the animals just before transport and daily for 17 days after installation in the fattening farm. On each occasion a group of ten animals (170 animals in total) was sampled. The levels of interferon-alpha (IFN-alpha) in serum were measured as antiviral activity in a cytopathic effect inhibition assay. Beginning at day 4 after installation, a significant proportion of sera contained IFN-alpha, with the highest incidence of IFN-alpha positive animals (25%) and IFN-alpha titers on days 5-10. This indicates a high frequency of viral infections in the animals. The in vitro ability of peripheral blood mononuclear leukocytes (PBMCs) to produce IFN-alpha after stimulation by glutaraldehyde-fixed pseudorabies virus-infected PK15 cells and their proliferative response to the T-cell mitogen leukoagglutinin (LA) was also monitored. There was a significant, but moderate decrease in the ability of PBMCs to produce IFN-alpha during the observation period. In contrast, the response to the mitogen LA decreased markedly during the first 5 days, and thereafter remained at the same low level. The proliferative response to LA was significantly lower for PBMCs from serum of IFN-alpha-positive than from IFN-alpha-negative animals. These impaired PBMC responses could indicate a stress-induced immune depression, possibly contributing to the high incidence of viral infections. 相似文献
