Suppression subtractive hybridization(SSH) was performed to isolate the differentially expressed cDNA fragments on maize under low-zinc and zinc-deficiency conditions.In the forward-subtracted library,the cDNA of maize seedlings treated with low-zinc was used as tester and the cDNA of maize seedlings treated with zinc-deficiency was used as driver.In the reverse-subtracted library,the cDNA of maize seedlings treated with zinc-deficiency was used as tester and the cDNA of maize seedlings treated with low-zinc was used as driver.The cDNA fragments differentially expressed under low-zinc and zinc-deficiency conditions were collected in the forward and reverse subtracted library,respectively.Fifteen clones were randomly picked to sequence in each library.The sequencing results showed that the genes induced under low-zinc and zinc-deficiency conditions were different.The differentially expressed genes could be used to explain the different appearances when maize seedlings were treated with low-zinc and zinc-deficiency. 相似文献

用溶液培养的方法研究了缺锌、低锌和正常供锌对玉米生长发育的影响.结果表明:缺锌和低锌使玉米的株高和茎叶干重均显著降低,低锌比缺锌降低的更多;缺锌和低锌对根干重的影响远小于对地上部的影响,所以,使根冠比增大,缺锌和低锌对玉米生长发育的这种影响与其影响玉米体内的锌含量有关,而且这种影响因玉米的品种不同而有差异. 相似文献

利用抑制差减杂交(Suppression subtractive hybridization,SSH)技术分离玉米低锌与正常供锌条件下表达有差异的cDNA片段.以低锌培养的玉米幼苗为试验方,正常供锌的玉米幼苗为对照方,建立正向差减文库;以正常供锌的玉米幼苗为试验方,以低锌诱导培养的玉米幼苗为对照方,建立反向差减文库.正、反向文库分别收集低锌和正常供锌条件下特异表达的cDNA片段.在2个文库中,各随机挑选30个克隆进行测序.序列分析表明,在正向差减文库中,分离得到的基因多与抗病防御、信号转导、细胞生长与分裂等方面相关;在反向差减文库中,分离得到的基因多与细胞组织发生及代谢等方面有关. 相似文献

以甘草幼苗为材料,采用营养液培养的方法,用0、8×10-6、4×10-5、2×10-4、10-3 mmol/L 5个不同浓度的加镧营养液培养甘草幼苗,通过比较生长量、药用成分积累量以及MDA质量摩尔浓度、游离氨基酸、可溶性蛋白质质量分数等研究不同浓度镧对甘草产量及品质的影响。结果表明,不同处理中,10-3 mmol/L镧浓度过大,导致植株死亡;8×10-6 mmol/L能有效促进甘草的生长,地上地下的产量分别高出不加镧处理的70.3%和46.6%,提高整株甘草黄酮和甘草多糖的积累量;叶片中MDA质量摩尔浓度比不加镧处理降低22.5%,有效地减弱脂质过氧化强度及膜系统的伤害。 相似文献

用溶液培养的方法研究了玉米幼苗对短暂供锌的反应及缺锌后再供锌的恢复效果.结果表明:10~12小时的正常供锌后再缺锌培养,对玉米幼苗的危害比一直缺锌的还大;缺锌培养使玉米幼苗出现缺锌症状后再正常供锌,可使之恢复,低锌使玉米出现的缺锌症状比缺锌培养的更难以恢复,证明低锌比缺锌对玉米造成的危害更大,缺锌使玉米的有机酸分泌增加,低锌增加的更多. 相似文献
