The change in the Common Agricultural Policy (CAP) of the European Union from product to producer support, including requirements for ‘good agricultural and environmental conditions’ and ‘greening’, is excellent. However, these requirements are now defined in rather general terms. Questions can be raised about suitable indicators, and there is a recognized need for effective management recommendations to support farmers in achieving the required ‘good’ conditions. These recommendations are bound to be quite different for different soils in different countries. A study of Dutch clay soils was based on a storyline describing current problems and management options for improvement, which were quantified using a soil–water–crop simulation model. Indicators were defined for agricultural conditions and suggestions made for the use of the model in a predictive mode to help farmers improve their soil management. Environmental conditions were judged by current environmental guidelines for water and air. When modelling, implicit assumptions that soils are homogeneous were shown to be unrealistic for these clay soils, requiring development of innovative methods and procedures, presenting a challenge for soil research.  相似文献

The saturated hydraulic conductivities (Ksat) of 22 spodic horizons with and without a thin iron pan, were measured in situ with a new technique, using large, carved-out columns, encased in gypsum. Measured infiltration rates and pressure heads above and below the spodic horizons allowed calculation of Ksat, which averaged 8 cm d?1. Flow rates averaged 32 cm d?1, however, due to a hydraulic head gradient across the spodic horizon of 4 cm cm?1. Occurrence of a thin iron pan in the spodic horizon did not affect its Ksat-value. The measured high flow rates exclude the occurrence of perched watertables. Lateral flow of water, forming surface ponds in local depressions, was due to surface runoff, rather than to lateral movement of perched water: surface ponding of water occurred also in soils in which the spodic horizon had been mixed by tillage.  相似文献

Aspergillus niger forms aerial hyphae and conidiophores after a period of vegetative growth. The hyphae within the mycelium of A. niger are divided by septa. The central pore in these septa allows for cytoplasmic streaming. Here, we studied inter- and intra-compartmental streaming of the reporter protein GFP in A. niger. Expression of the gene encoding nuclear targeted GFP from the gpdA or glaA promoter resulted in strong fluorescence of nuclei within the vegetative hyphae and weak fluorescence in nuclei within the aerial structures. These data and nuclear run on experiments showed that gpdA and glaA are higher expressed in the vegetative mycelium when compared to aerial hyphae, conidiophores and conidia. Notably, gpdA or glaA driven expression of the gene encoding cytosolic GFP resulted in strongly fluorescent vegetative hyphae and aerial structures. Apparently, GFP streams from vegetative hyphae into aerial structures. This was confirmed by monitoring fluorescence of photo-activatable GFP (PA-GFP). In contrast, PA-GFP did not stream from aerial structures to vegetative hyphae. Streaming of PA-GFP within vegetative hyphae or within aerial structures of A. niger occurred at a rate of 10–15 μm s-1. Taken together, these results not only show that GFP streams from the vegetative mycelium to aerial structures but it also indicates that its encoding RNA is not streaming. Absence of RNA streaming would explain why distinct RNA profiles were found in aerial structures and the vegetative mycelium by nuclear run on analysis and micro-array analysis.Key words: aerial hypha, Aspergillus, conidia, conidiophore, cytoplasmic streaming, development, fungus, vegetative mycelium  相似文献

Controlled clinical trials to a standardised protocol were conducted into the effect of a water-soluble antibiotic on proliferative enteropathy and its causative agent (Lawsonia intracellularis) on commercial pig farms at six sites in four European countries. Clinical signs of the disease and L intracellularis-specific polymerase chain reaction (PCR)-positive pigs were detected in pens of six- to 12-week-old pigs (weighing 5 to 55 kg) immediately before each trial. Matched pens of randomised pigs were either left unmedicated (32 to 59 pigs per trial), or medicated orally with 10 mg/kg of a water-soluble combination of lincomycin and spectinomycin powder (21 and 42 mg, respectively, of antibiotic activity per litre) for either seven days (33 to 61 pigs per trial), or 14 days (33 to 61 pigs per trial), delivered via the drinking water. Investigators did not know which pens received which treatment In most of the affected pigs in each trial, diarrhoea due to L intracellularis resolved within three to seven days after the medication began, whereas most unmedicated pigs remained diarrhoeic for at least 10 days. On average the medicated pigs gained more weight than the unmedicated pigs over the 21-day trial period (P=0.01). In two trials, the absence of L intracellularis after the treatment ended was confirmed by the PCR.  相似文献

Porcine parvovirus was propagated in PK-15 cells cultured in roller bottles or on microcarrier beads. After inactivation, the virus was used as antigen in the preparation of vaccines. The immunogenic potency and safety of the vaccines were evaluated in specific pathogen free pregnant gilts and guinea pigs. Experimental challenge tests determined the efficacy of the vaccine in preventing porcine parvovirus transplacental infection. Neither viral antigens nor specific antibodies were detected in fetuses from vaccinated gilts. In contrast, fetal death and, or, mummification occurred when unvaccinated gilts were infected. Both virus and, or, antibodies were also detected in fetuses from these unvaccinated gilts. Serum conversion after vaccination was assayed by microserum neutralisation using guinea pig erythrocytes as cell indicators and by haemagglutination inhibition tests. Viral antigens in fetal tissues were detected using ELISA, the immunobeads technique, the haemagglutination test and by virus isolation.  相似文献

Many environmental studies on the protection of European soil and water resources make use of soil water simulation models. A major obstacle to the wider application of these models is the lack of easily accessible and representative soil hydraulic properties. In order to overcome this apparent lack of data, a project was initiated to bring together the available hydraulic data which resided within different institutions in Europe into one central database. This information was then used to derive a set of pedotransfer functions applicable to studies at a European scale. These pedotransfer functions predict the hydraulic properties from parameters collected during soil surveys and can be a good alternative for costly and time-consuming direct measurement of these properties. A total of 20 institutions from 12 European countries collaborated in establishing the database of draulic operties of uropean oils (HYPRES). This database has a flexible relational structure capable of holding a wide diversity of both soil pedological and hydraulic data. As these data were contributed by 20 different institutions it was necessary to standardise both the particle-size and the hydraulic data. A novel similarity interpolation procedure was successfully used to achieve standardization of particle-sizes according to the FAO clay, silt and sand particle-size ranges. Standardization of hydraulic data was achieved by fitting the Mualem-van Genuchten model parameters to the individual θ(h) and K(h) hydraulic properties stored in HYPRES. The HYPRES database contains information on a total of 5521 soil horizons (including replicates). Of these, 4030 horizons had sufficient data to be used in the derivation of pedotransfer functions. Information on both water retention and hydraulic conductivity was available for 1136 horizons whereas 2894 horizons had only information on water retention. Each soil horizon was allocated to one of 11 possible soil textural/pedological classes derived from the six FAO texture classes (five mineral and one organic) and the two pedological classes (topsoil and subsoil) recognised within the 1:1 000 000 scale Soil Geographical Data Base of Europe. Next, both class and continuous pedotransfer functions were developed. By using the class pedotransfer functions in combination with the 1:1 000 000 scale Soil Map of Europe, the spatial distribution of soil water availability within Europe was derived.  相似文献

Interrelationships between peat and water were studied using a hydropedological modelling approach for adjacent relatively intact and degraded peatland in Central Kalimantan, Indonesia. The easy to observe degree of peat humification provided good guidance for the assignment of more difficult to measure saturated hydraulic conductivities to the acrotelm–catotelm hydrological system. Ideally, to prevent subsidence and fire, groundwater levels should be maintained between 40 cm below and 100 cm above the peat surface. Calculated groundwater levels for different years and for different months within a single year showed that these levels can drop deeper than the critical threshold of 40 cm below the peat surface whilst flooding of more than 100 cm above the surface was also observed. In July 1997, a dry El Niño year, areas for which deep groundwater levels were calculated coincided with areas that were on fire as detected from radar images. The relatively intact peatland showed resilience towards disturbance of its hydrological integrity whereas the degraded peatland was susceptible to fire. Hydropedological modelling identified areas with good restoration potential based on predicted flooding depth and duration. Groundwater level prediction maps can be used in fire hazard warning systems as well as in land utilization and restoration planning. These maps are also attractive tools to move from the dominant uni-sectoral approach in peatland resource management toward a much more promising multi-sectoral approach involving various forestry, agriculture and environment agencies. It is demonstrated that the combination of hydrology and pedology is essential for wise use of valuable but threatened tropical peatland ecosystems.  相似文献
