Melanin is a widely distributed and striking dark-colored pigment produced by countless living organisms. Although a wide range of bioactivities have been recognized, there are still major constraints in using melanin for biotechnological applications such as its fragmentary known chemical structure and its insolubility in inorganic and organic solvents. In this study, a bacterial culture of Streptomyces cavourensis SV 21 produced two distinct forms of melanin: (1) a particulate, insoluble form as well as (2) a rarely observed water-soluble form. The here presented novel, acid-free purification protocol of purified particulate melanin (PPM) and purified dissolved melanin (PDM) represents the basis for an in-depth comparison of their physicochemical and biological properties, which were compared to the traditional acid-based precipitation of melanin (AM) and to a synthetic melanin standard (SM). Our data show that the differences in solubility between PDM and PPM in aqueous solutions may be a result of different adjoining cation species, since the soluble PDM polymer is largely composed of Mg2+ ions and the insoluble PPM is dominated by Ca2+ ions. Furthermore, AM shared most properties with SM, which is likely attributed to a similar, acid-based production protocol. The here presented gentler approach of purifying melanin facilitates a new perspective of an intact form of soluble and insoluble melanin that is less chemical altered and thus closer to its original biological form.  相似文献

Triplicate groups of one hundred Tra catfish (8 g?±?0.2) were fed seven test diets containing increasing levels of AFB1 (0, 50, 100, 250, 500, and 1000 μg AFB1 kg?1). Additionally Mycofix® Secure was added at 1.5% to one diet containing 500 μg AFB1 kg?1. Results showed that Tra catfish are sensitive to AFB1. Reduction in weight gain (P?<?0.05) was observed for fish fed 50 μg AFB1 kg?1 and declined further with increasing levels of AFB1 in the diets. Fish fed diets contaminated with 500 and 1000 μg AFB1 kg?1 showed increased (P?>?0.05) hepatosomatic index (HIS), while an increase in adipose somatic index (ASI) was observed in fish fed 50 μg AFB1 kg?1 and above when compared to the control and Mycofix® diets. After 12 weeks, blood serum analysis revealed higher alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels in fish fed the 50, 100, and 250 μg AFB1 kg?1 suggesting occurrence of liver damage. Disease resistance of fish exposed to Edwardsiella ictaluri was also compromised by the presence of AFB1 in the feed and was directly related to the contamination level. Seven days after Edwardsiella ictaluri exposure, survival rates were 50, 41.7, 31.7, and 8.3% for fish fed control, 50, 100, and 250 μg AFB1 kg?1, respectively. This trial shows that AFB1 at a level of 50 μg AFB1 kg?1 and above can affect fish performance and disease resistance. Application of an effective mycotoxin management in the feed seems to be useful to prevent the negative effects of AFB1.  相似文献

A new social forestry program has been implemented in Java to overcome encroachment of state forests. In this program, the state and local communities jointly manage the state forests and share the benefits of increased forest resource stock and flow as a result of the management. This article aims to investigate the complete incentive structure of the social forestry program and how the incentive structure changes community member participation in forest management. Property rights transfers and economic analyses were employed to analyze the incentive structure. We conclude that although incomplete, the transfer of rights from the state to the community members is a useful approach toward establishing an effective incentive structure and provides a net gain to the community members. The extent of community participation, however, depends not only on the incentive structure but also on the social capital that exists in the community.  相似文献

Complexes of starch and monoacyl-sn-glycerophosphatidylcholine (GPC) containing various acyl (myristoyl, palmitoyl, and stearoyl) chains were subjected to hydrolysis with glucoamylase (EC 3.2.1.3). The enzyme hydrolyzed approximately 40% of starch control and 20-28% of starch-GPC complexes. Among the GPCs examined, 1- and 2-monomyristoyl-sn-GPC showed the highest resistance to enzyme hydrolysis, and the hydrolysis rate of starch-GPCs was greater with longer chains. Enzymatic hydrolysis strongly affected the thermal properties of the starch. After enzymatic hydrolysis of starch-GPC complexes for 24 h, their thermograms had broader peaks with lower enthalpies than the corresponding starch without enzyme; however, the starch-GPC complexes showed little change. The surface of starch-GPC granules was less eroded. These results showed that the increasing amount of starch-GPC complexes could be more resistant to hydrolysis.  相似文献

A series of treatments were designed to evaluate the efficacy of feeding commercial multispecies probiotics feeding with enzymes and fermentation process on the growth parameters and culture environment of Pacific white shrimp, Litopenaeus vannamei (Boone, 1931), in an intensive culture system. Commercial multispecies probiotics and enzymes (PEs) were continuously applied in three different doses, namely (i) 0.2, (ii) 0.4, and (iii) 0.6 mg L−1 and designated as 0.2, 0.4, and 0.6 PE during the first 30 days of intensive culture of Pacific white shrimp, L. vannamei (Boone, 1931). The probiotics were continually applied every alternate day, while the enzymes were added every sixth day throughout the trial period. The PE dose for all treated tanks was increased by 0.2 ppm after 30 days of culture and another 0.2 ppm after day 60. Meanwhile, fermentation technique, which has become the common method applied in Indonesia in the control treatment, was added on the same day with the PE group with increasing dosage, following the same trend with the 0.6 PE group. Results showed that the group receiving 0.6 PE showed higher final biomass, higher mean weight, and protein retention efficiency as well as a lower feed conversion ratio compared with the control treatment. Abundance of Vibrio spp. remained below 103 cfu mL−1 throughout the trial. Water quality indicators TAN, NO2 N and NO3 N peaked in weeks 3–5 and then declined after that until the end of the culture period in all tanks. This decline was significantly faster in PE-treated tanks. Multispecies PEs have potential applications in controlling Vibrio spp., maintaining proper water quality condition, and enhancing the growth of shrimp in intensive culture system.  相似文献

Development of transferred xenogeneic vole embryos in mouse uteri   总被引:1,自引:0,他引:1  
An experimental model to study interspecific pregnancy using voles, Microtus arvalis, and green fluorescent protein gene‐induced transgenic mice is presented. Xenogeneic blastocysts from the vole were transferred into the uteri of pseudopregnant mice along with allogeneic blastocysts from green fluorescent protein gene‐induced transgenic mice. The uteri containing xeno‐allo combined transfers were examined from day 6 to 13 of gestation. Although the vole embryos implanted, the uteri containing vole embryos were smaller compared with those having allogeneic mouse embryos. On day 8, the uteri containing vole embryos hemorrhaged internally and no vole embryo was found in the pregnant uterus after day 11. Allogeneic mouse embryos developed normally despite the presence and abortion of the vole embryos. In uteri implanted with vole embryos, decidua were formed and numerous blood vessels were distributed around the embryo. Maternal blood cells infiltrated into the celomic cavity of the vole embryo through the discontinuous region of trophoblast. Periodic acid‐Schiff‐positive granulated metrial gland cells were remarkably increased in the decidual sites. These findings suggest that a disorder of embryo–maternal interaction might induce the appearance of numerous granulated metrial gland cells and rejection of the embryos.  相似文献

In the daily and final landfill cover barrier system, the hydraulic properties of compacted soil liners and the strength of soil can be adversely affected by desiccation cracking, resulting in the loss of effectiveness and integrity of the containment system as a barrier. Recently, there is an interest of using fiber additive to overcome the desiccation cracking problem. In this study, the desiccation crack test was conducted to investigate the effect of fiber additive on suppressing desiccation cracks in compacted Akaboku soils. Polypropylene (C3H6) fiber was used as an additive material for soil sample. The percentages of fiber used were varied as 0.0%, 0.2%, 0.4%, 0.6%, 0.8%, 1.0% and 1.2% (by dry weight of samples). The soil specimens were compacted under the conditions of maximum dry density and optimum water content. The surficial cracking area was measured to determine the crack intensity factor (CIF) of the soil samples. The desiccation crack test results indicated that the percentage of volume change of the compacted soil specimen decreased with addition of fiber. The change in the soil surface area decreased with increasing in the fiber content (FC), and consequently, the volumetric shrinkage strain decreased. The CIF for the soil without fiber (FC?=?0.0%) were significantly higher than the soil with fiber additive. The CIF of soil at FC?=?0.0% decreased from 2.75% to 0.6% for the soil at FC?=?0.2%. It was also found that the maximum crack depth reaches almost 50% of the thickness of the soil without fiber additive. This study suggests the potential application of the fiber additives to soils as an available method to suppress desiccation cracks encountered in landfill cover barriers.  相似文献

Two feeding experiments were conducted to confirm methionine requirement in practical diets of juvenile Nile tilapia, Oreochromis niloticus. Test diets used in both experiments contained 414 kcal gross energy, 28 g protein, and 5 g lipid per 100 g diet. In the first experiment, seven diets were made using cottonseed meal (CSM), dehulled solvent‐extracted soybean meal (DSESM), and gelatin as intact protein sources. Methionine was added to five of these diets at 0.03 or 0.06% increments to produce methionine levels ranging from 0.33 to 0.57% of the diet. Each diet was fed to four replicate groups of male juvenile Nile tilapia (5.62 ± 0.13 g) in a recirculation system for 8 wk. Broken‐line regression analysis of weight gain indicated that methionine requirement of juvenile Nile tilapia was 0.49% of the diet or 1.75% of dietary protein at cystine level of 0.45% of the diet. The second experiment was designed based on methionine requirement determined in the first experiment and also contained seven test diets. The first six diets contained CSM and DSESM as protein sources. Methionine was added to five of these diets at an increasing rate of 0.06% to produce methionine levels ranging from 0.49 to 0.79% of the diet. In the last diet (Diet 7), a portion of DSESM was replaced by gelatin to reduce methionine level to 0.33% of the diet in order to test whether methionine is limited. Each diet was also fed to four replicate groups of male juvenile Nile tilapia (2.32 ± 0.06 g) in a recirculation system for 9 wk. At the termination of the second experiment, there were no significant differences in terms of weight gain, survival, and feed efficiency ratio (FER) among the fish fed the first six diets. However, weight gain and FER of the fish fed these diets were significantly better than those fed Diet 7, confirming the methionine requirement value as has been determined in the first experiment.  相似文献
