Numerous culture-based diagnostics are available on the Australian and international markets for on-farm detection of bacterial pathogens in milk. Use of such diagnostics may provide an opportunity to improve the prudent use of antimicrobials in udder health management. Farms are low-resource settings in terms of diagnostic microbiology capacity. The World Health Organisation has identified criteria for the evaluation of diagnostic tests in low resource settings based on Accuracy, Sensitivity, Specificity, User-friendliness, being Rapid or Robust, Equipment-free and being Deliverable (ASSURED). Here, we review how those criteria can be interpreted in the context of microbiological diagnosis of mastitis pathogens, and how on-farm diagnostics that are currently available in Australia perform relative to ASSURED criteria. This evaluation identifies multiple trade-offs, both with regard to scientific criteria and with regards to convenience criteria. More importantly, the purpose of testing may differ between farms, and test performance should be evaluated relative to its intended use. The ability of on-farm mastitis diagnostics to inform mastitis treatment decision-making in a timely and cost-effective manner depends not just on test characteristics but also on farm-specific pathogen prevalence, and on the farm enterprise's priorities and the farm manager's potential courses of action. With most assay evaluations to date conducted in professional laboratories, there is a surprising dearth of information on how well any of the diagnostic tests perform on-farm and, indeed, of the on-farm decision-making processes that they aim to inform.  相似文献

The aim of the current study was to determine the possible effects of progestagen oestrous synchronization on vascular endothelial growth factor (VEGF) expression during sheep luteogenesis and the peri‐implantation period and the relationship with luteal function. At days 9, 11, 13, 15, 17 and 21 of pregnancy, the ovaries from 30 progestagen treated and 30 ewes cycling after cloprostenol injection were evaluated by ultrasonography and, thereafter, collected and processed for immunohistochemical evaluation of VEGF; blood samples were drawn for evaluating plasma progesterone. The progestagen‐treated group showed smaller corpora lutea than cloprostenol‐treated and lower progesterone secretion. The expression of VEGF in the luteal cells increased with time in the cloprostenol group, but not in the progestagen‐treated group, which even showed a decrease between days 11 and 13. In progestagen‐treated sheep, VEGF expression in granulosa‐derived parenchymal lobule capillaries was correlated with the size of the luteal tissue, larger corpora lutea had higher expression, and tended to have a higher progesterone secretion. In conclusion, the current study indicates the existence of deleterious effects from exogenous progestagen treatments on progesterone secretion from induced corpora lutea, which correlate with alterations in the expression of VEGF in the luteal tissue and, this, presumably in the processes of neoangiogenesis and luteogenesis.  相似文献

This study was designed to evaluate the effect of single layer centrifugation (SLC) and subsequent cold storage on stallion sperm capacitation‐like status and acrosome reaction. Three stallions were included in the study, with three ejaculates per stallion. The samples were examined 4, 24 and 72 h after collection, extension and SLC, with storage at 6°C. Sperm capacitation‐like status was investigated using the fluorescent dye chlortetracycline (CTC). There was no difference in capacitation‐like status between colloid‐selected and non‐selected spermatozoa. Sperm motility decreased significantly during cold storage, whereas the proportion of apparently capacitated spermatozoa increased. There was no change in the proportion of acrosome‐reacted spermatozoa. In conclusion, SLC through Androcoll?‐E does not adversely affect the capacitation‐like status of stallion spermatozoa, although it did increase with time during cold storage.  相似文献

Tillage affects the soil physical and chemical environment in which soil microorganisms live, thereby affecting their number, diversity and activity. However, soil disturbance generally has the greatest impact on biological properties, including both free and symbiotic fungal populations. Interest in more ecologically sustainable agricultural systems is rising with increasing recognition that agricultural intensification can adversely affect environmental quality. This paper discusses the effect of tillage system on some soil characteristics, such as pH, C, N and S levels, total and Olsen-P contents including some P forms associated with organic matter, glomalin contents and arbuscular mycorrhizae (AM) parameters, such as root colonization, spore number and total and active hyphal length. Measurements were in the sixth year of an on-going tillage-rotation experiment conducted on an Ultisol under no-till (NT), reduced tillage (RT) and conventional tillage with stubble mixed into the soil (CTS) or stubble burnt (CTB). Soil was sampled at two dates; after wheat (Triticum aestivum) harvest (autumn) and 6 months after subsequent grassland seeding (spring). Higher C, N, S, total P and fulvic acid-P concentrations and pH occurred under NT and RT than under CTS and CTB after wheat harvest. However, results at the second sampling were not consistent. AM spore number and active hyphal length were highest under NT having the greatest incidence on AM root colonization and P concentration in shoots of the pasture. Glomalin concentration was higher under NT and RT than under CTS and CTB but no differences in calculated glomalin to total C (ca. 5%) were found. It is concluded that a less disruptive effect of NT influences positively all soil characteristics and also increases P acquisition by the following crop in the rotation system.  相似文献

Objective The aims of this work were to (1) develop a low-cost equine movement tracking collar based on readily available components, (2) conduct preliminary studies assessing the effects of both paddock size and internal fence design on the movements of domestic horses, with and without foals at foot, and (3) describe distances moved by mares and their foals. Additional monitoring of free-ranging feral horses was conducted to allow preliminary comparisons with the movement of confined domestic horses. Procedures A lightweight global positioning system (GPS) data logger modified from a personal/vehicle tracker and mounted on a collar was used to monitor the movement of domestic horses in a range of paddock sizes and internal fence designs for 6.5-day periods. Results In the paddocks used (0.8–16 ha), groups of domestic horses exhibited a logarithmic response in mean daily distance travelled as a function of increasing paddock size, tending asymptotically towards approximately 7.5 km/day. The distance moved by newborn foals was similar to their dams, with total distance travelled also dependent on paddock size. Without altering available paddock area, paddock design, with the exception of a spiral design, did not significantly affect mean daily distance travelled. Feral horses (17.9 km/day) travelled substantially greater mean daily distances than domestic horses (7.2 km/day in 16-ha paddock), even when allowing for larger paddock size. Conclusions Horses kept in stables or small yards and paddocks are quite sedentary in comparison with their feral relatives. For a given paddock area, most designs did not significantly affect mean daily distance travelled.  相似文献

The objective of this study was to define the patterns of organogenesis and foetal haemodynamics during the normal gestation of healthy agoutis (Dasyprocta prymnolopha) kept in captivity. Thirty pregnant agoutis that ranged in size from small to medium and weighed between 2.5 and 3 kg underwent B‐mode and Doppler ultrasonography for the biometric evaluation of the foetal organs. The foetal aortic blood flow proved to be predominantly systolic, and the measured flow velocity was 78.89 ± 2.95 cm/s, with a maximum pressure gradient of 2.12 ± 0.27 mmHg. The liver was characterized by its large volume, occupying the entire cranial aspect of the abdominal cavity, and it was associated cranially with the diaphragm and caudally with the stomach. The flow velocity in the portal vein was estimated to equal 12.17 ± 2.37 cm/s, with a resistivity index of 0.82 ± 0.05. The gallbladder was centrally located and protruded cranially towards the diaphragm. The spleen was visualized as an elongated structure with tapered cranial and caudal extremities, and the foetal kidneys were visualized bilaterally in the retroperitoneal region, with the right kidney positioned slightly more cranially than the left. The morphological characterization and hemodynamic analysis of the foetal organs of black‐rumped agoutis via B‐mode and Doppler ultrasonography allow determination of the vascular network and of reference values for the blood flow required for perfusing the anatomical elements essential for maintaining the viability of foetuses at different gestational ages.  相似文献

Single layer centrifugation (SLC) has been shown to select the most robust spermatozoa from the ejaculate in several species. Here the effects of SLC prior to freezing on various parameters of frozen‐thawed bovine sperm quality are reported. Semen from 8 bulls was layered on top of a species‐specific colloid, Bovicoll. After centrifugation for 20 min at 300 g, the resulting sperm pellet was resuspended in OPTIXcell® (IMV Technologies, l′Aigle, France); the SLC‐selected sperm samples and uncentrifuged controls were frozen. On thawing, all sperm samples were analysed for membrane integrity, production of reactive oxygen species, mitochondrial membrane potential (MMP) and chromatin integrity. The SLC‐treated samples had a higher percentage of live, superoxide‐positive spermatozoa than uncentrifuged samples (27.9 ± 5.1% versus 21.7 ± 6.7%; p = .03). They had a higher proportion of spermatozoa with high mitochondrial membrane potential than uncentrifuged samples (55.9 ± 8.2% versus 40.5 ± 15.1%; p = .03) and also a lower proportion of spermatozoa with low mitochondrial membrane potential than non‐treated samples (42.0 ± 8.5% versus 55.9 ± 14.4%; p = .04). No significant effects of treatment were found for membrane integrity or chromatin integrity. The effect of bull was significant on the proportions of dead, superoxide‐positive spermatozoa and live, hydrogen peroxide‐negative spermatozoa, as well as on membrane integrity, but it was not significant for mitochondrial membrane potential or chromatin integrity. These results suggest that SLC selects the most metabolically active bull spermatozoa from the rest of the population in normal ejaculates; the pattern of reactive oxygen species production may be different in SLC‐selected spermatozoa compared to unselected samples.  相似文献

The 2013 EU ban of three neonicotinoids used in seed coating of pollinator attractive crops was put in place because of concern about declining wild pollinator populations and numbers of honeybee colonies. It was also concluded that there is an urgent need for good field data to fill knowledge gaps. In the meantime such data have been generated. Based on recent literature we question the existence of recent pollinator declines and their possible link with the use of neonicotinoids. Because of temporal non‐coincidence we conclude that declines of wild pollinators and of honeybees are not likely caused by neonicotinoids. Even if bee decline does occur and if there is a causal relationship with the use of neonicotinoids, we argue that it is not possible on such short term to evaluate the effects of the 2013 ban. In order to supply future debate with realistic (field) data and to discourage extrapolating the effects of studies using overdoses that are not of environmental relevance, we propose – in addition to field studies performed by the chemical industry – to use the ‘semi‐field worst case’ treated artificial diet studies approach to free flying colonies in the field. This kind of study may provide realistic estimates for risk and be useful to study realistic interactions with non‐pesticide stressors. © 2017 The Authors. Thinner pod walls might contribute to increased yield potential and adaptation of lupins. Fourteen autumn‐sown genotypes of Lupinus albus L. were evaluated over 2 years at four sites in southern Chile. Pod wall proportion (PWP), pod wall specific weight (WSW) and five other pod‐related characters were measured to estimate their genetic variation, heritability across years and sites, and genotypic and phenotypic correlations among the characters. Ranges for PWP and pod WSW were 22.9−43.1% and 16.4−37.4 mg/cm2, respectively. PWP in cultivated L. albus was similar to that reported in L. angustifolius; however, specific weight was substantially lower. Highly significant genotypic effects were found for both characters. Broad sense heritabilities were high for PWP (0.63) and moderate for pod WSW (0.46). These characters were significantly correlated, more so genotypically (rg = 0.67) than phenotypically (rph = 0.36). Selection for low PWP should lead to lower pod wall thickness or density, both of which contribute to pod WSW. Nevertheless, selection for pod WSW could achieve further reductions, particularly if practised among low PWP materials.  相似文献

Intensive tillage for annual crop production may be affecting soil health and quality. However, tillage intensity effects on biological activities of volcanic-derived soils have not been systematically investigated. We evaluated the effects of three different tillage practices on some biological activities of an Ultisol from southern Chile during the third year of a wheat–lupin–wheat crop sequence. Treatments were: no tillage with stubble burning (NTB), no tillage without stubble burning (NT) and conventional tillage with disk-harrowing and stubble burning (CT). Biological activities were evaluated in winter and summer at 0–200 mm and at three soil depths (0–50, 50–100 and 100–200 mm) in winter. Total organic C and N were significantly higher under no-tillage systems than CT. In general, NT increased C and N of microbial biomass in comparison with CT, especially in winter. Microbial biomass C was closely associated with microbial biomass N (r = 0.986, P < 0.05); acid phosphomonoesterase (r = 0.999, P < 0.05); β-glucosidase (r = 0.978, P < 0.05), and others. Changes in biological activities occurred mainly in the upper soil layer (0–50 mm depth) in spite of the short duration of the experiment. Biological activities could be used as practical biological indicators to apply the more appropriate management systems for increasing soil sustainability or productivity.  相似文献
