Piscirickettsia salmonisis the causative bacterial pathogen of piscirickettsiosis, a salmonid disease that causes notable mortalities in the worldwide aquaculture industry. Published research describes the phenotypic traits, virulence factors, pathogenicity and antibiotic‐resistance potential for various P. salmonisstrains. However, evolutionary and genetic information is scarce for P. salmonis. The present study used multilocus sequence typing (MLST) to gain insight into the population structure and evolution of P. salmonis. Forty‐two Chilean P. salmonisisolates, as well as the type strain LF‐89T, were recovered from diseased Salmo salar, Oncorhynchus kisutchand Oncorhynchus mykissfrom two Chilean Regions. MLST assessed the loci sequences of dnaK, efp, fumC, glyA, murG, rpoD and trpB. Bioinformatics analyses established the genetic diversity among P. salmonis isolates (H = 0.5810). A total of 23 sequence types (ST) were identified, 53.48% of which were represented by ST1, ST5 and ST2. Population structure analysis through polymorphism patterns showed few polymorphic sites (218 nucleotides from 4,010 bp), while dN/dS ratio analysis indicated purifying selection for dnaK, epf, fumC, murG, and rpoD but neutral selection for the trpB loci. The standardized index of association indicated strong linkage disequilibrium, suggesting clonal population structure. However, recombination events were detected in a group of seven isolates. Findings included genogroups homologous to the LF‐89T and EM‐90 strains, as well as a seven‐isolate hybrid genogroup recovered from both assessed regions (three O. mykiss and four S. salar isolates). The presented MLST scheme has comparative potential, with promising applications in studying distinct P. salmonis isolates (e.g., from different hosts, farms, geographical areas) and in understanding the epidemiology of this pathogen. 相似文献

Although pasta is generally not considered for its aromatic properties, some evidence proves that cereal flours release volatile compounds and they might have an effect on the aroma of the transformed products. This work reports on the characterization of the volatile components of semolina and pasta obtained from four durum wheat cultivars (Triticum durum Desf., cvs. PR22D89, Creso, Cappelli, Trinakria). Semolina samples were characterized through polar metabolite profiling and fatty acid analysis to identify potential precursors of the volatile components. The results show significant differences among the samples tested with cv. Trinakria characterized by the highest content of sugars and fatty acids. Volatile composition was investigated both in semolina and in cooked pasta using headspace solid-phase micro-extraction (HS-SPME) and identified by GC–MS. Thirty-five volatile compounds including aldehydes, ketones, alcohols, terpenes, esters, hydrocarbons and a furan were identified. Significant differences were observed between semolina and pasta samples in terms of composition and amount of the volatile compounds. During cooking an increase in aldehyde content, the appearance of ketones and a decrease in alcohol content were observed. Correlations between metabolites and volatiles demonstrate that the flavour of cooked pasta may differ significantly depending on the durum wheat cultivar employed. 相似文献

Our main goal was to investigate if robust chemical fingerprints could be developed for three Argentinean red wines based on organic, inorganic, and isotopic patterns, in relation to the regional soil composition. Soils and wines from three regions (Mendoza, San Juan, and Co?rdoba) and three varieties (Cabernet Sauvignon, Malbec, and Syrah) were collected. The phenolic profile was determined by HPLC-MS/MS and multielemental composition by ICP-MS; (87)Sr/(86)Sr and δ(13)C were determined by TIMS and IRMS, respectively. Chemometrics allowed robust differentiation between regions, wine varieties, and the same variety from different regions. Among phenolic compounds, resveratrol concentration was the most useful marker for wine differentiation, whereas Mg, K/Rb, Ca/Sr, and (87)Sr/(86)Sr were the main inorganic and isotopic parameters selected. Generalized Procrustes analysis (GPA) using two studied matrices (wine and soil) shows consensus between them and clear differences between studied areas. Finally, we applied a canonical correlation analysis, demonstrating significant correlation (r = 0.99; p < 0.001) between soil and wine composition. To our knowledge this is the first report combining independent variables, constructing a fingerprint including elemental composition, isotopic, and polyphenol patterns to differentiate wines, matching part of this fingerprint with the soil provenance. 相似文献

Context
Amphibians are declining worldwide and land use change to agriculture is recognized as a leading cause. Argentina is undergoing an agriculturalization process with rapid changes in landscape structure.Objectives
We evaluated anuran response to landscape composition and configuration in two landscapes of east-central Argentina with different degrees of agriculturalization. We identified sensitive species and evaluated landscape influence on communities and individual species at two spatial scales.Methods
We compared anuran richness, frequency of occurrence, and activity between landscapes using call surveys data from 120 sampling points from 2007 to 2009. We evaluated anuran responses to landscape structure variables estimated within 250 and 500-m radius buffers using canonical correspondence analysis and multimodel inference from a set of candidate models.Results
Anuran richness was lower in the landscape with greater level of agriculturalization with reduced amount of forest cover and stream length. This pattern was driven by the lower occurrence and calling activity of seven out of the sixteen recorded species. Four species responded positively to the amount of forest cover and stream habitat. Three species responded positively to forest cohesion and negatively to rural housing. Two responded negatively to crop area and diversity of cover classes.Conclusions
Anurans within agricultural landscapes of east-central Argentina are responding to landscape structure. Responses varied depending on species and study scale. Life-history traits contribute to responses differences. Our study offers a better understanding of landscape effects on anurans and can be used for land management in other areas experiencing a similar agriculturalization process.6.
The carotenoid concentration of durum wheat is a criterion for the assessment of semolina quality, and it is of particular importance in determining the color of pasta. Thus, the development of a rapid screening method for kernel and semolina color has facilitated increasing the yellow color in durum wheat cultivars. However, the distribution of the pigment might vary within the ear of the wheat. The micromethod for the determination of yellow pigment concentration (YPC) of single wheat kernels now allows the discovery of whether this variation is the same for cultivars with different carotenoid concentrations. All of the cultivars investigated showed similar trends in the arrangements of the kernels within the ears, with the central positions of the ears being the most stable for YPC. Indeed, the best combination of higher YPC and larger kernel size is seen for the basal‐central region of the ear, which can be used for the selection during wheat‐breeding programs of cultivars with a more intense yellow color. 相似文献

- Freshwater ecosystems are under a constant risk of being irreversibly damaged by human pressures that threaten their biodiversity, the sustainability of ecosystem services (ESs), and human well‐being. Despite the implementation of various environmental regulations, the challenges of safeguarding freshwater assets have so far not been tackled successfully.
- A promising way forward to stop the loss of freshwater biodiversity and to sustain freshwater‐based ESs is by implementing ecosystem‐based management (EBM), an environmental planning and adaptive management approach that jointly considers social and ecological needs. Responsible for considerable recent success in sustainably managing and conserving marine ecosystems, EBM has not yet been championed for fresh waters.
- A major reason for the delayed uptake of EBM in fresh waters is likely to be its complexity, requiring planners to be familiar with the latest developments in a range of different research areas. EBM would therefore benefit from becoming more tangible to receive attention on the ground.
- To facilitate uptake, eight core research areas for EBM and their innovations are introduced, and the way in which they feed into the workflow that guides the EBM planning stage is explained.
- The workflow links biodiversity distributions with ES supply‐and‐demand modelling and SMART (specific, measurable, attainable, relevant, and timely) target planning, including scenario‐ and cross‐realm perspectives, the prioritization of management alternatives, spatial prioritization of biodiversity conservation and ES areas, and the quantification of uncertainties. Given the extensive resources, time, and technical capacity required to implement the full workflow, a light and an ultralight version of the workflow are also provided.
- Applied in concert, the eight well‐known research areas allow for better planning and operationalizing, and eventually for implementing EBM in freshwater ecosystems. EBM has great potential to increase public acceptance by introducing the consideration of human needs and aspirations into typically biodiversity‐driven conservation and management approaches. This will ultimately improve the integrity of freshwater ecosystems.

Fusarium head blight (FHB) caused by Fusarium graminearum (FG) is a destructive disease impacting barley worldwide. The disease reduces the grain yield and contaminates grains with mycotoxins, such as the trichothecene deoxynivalenol (DON). Although the infection mainly affects the grain yield, little is known about its impact on grain structural and biochemical properties. Yet, such information is instrumental to characterize the facets of resistance in the grains. After artificial inoculation of six barley cultivars with FG in a 2 years field test, different levels of symptoms on spikes, of colonisation of grains and of DON content were observed. The infections caused a reduction in grain weight and an average decrease of 10% of the β-glucan content in grains, indicating alterations of grain filling, composition and structure. According to our results, we postulate the presence of two distinct resistance mechanisms in the grain, tolerance to grain filling despite infection as well as the inhibition of mycotoxin accumulation. Differently to wheat, in barley, type IV resistance (tolerance of the grain to infection) is directly linked with type III resistance (resistance against kernel infection). The resistance against toxin accumulation (named type V resistance in wheat) appeared to be independent to all other resistance types. Generally, the resistance was significantly influenced by the environment and by genotype x environment interactions explaining the generally weak stability of resistance in barley. Interestingly, a significant and inverse relationship between DON contamination and β-glucan content in grains suggests that high β-glucan content in grains contributes to type V resistance. 相似文献
