Productivity and sustainability of rice-rice cropping system depend upon the soil quality which is primarily governed by application of fertilizers and manures. However, such information is limited and hence, the present investigation was carried out in a 9-year-old long-term fertilizer experiment at Bhubaneswar, India. There were seven treatments (control, application of 100% NPK, 150% NPK, 100% NPK + Zn, 100% NPK + FYM, 100% NPK + Zn + B, and 100% NPK + Zn + S) laid out in randomized block design with four replications. Indicators of soil quality (physical, chemical, and biological) were diagnosed from 30 numbers of soil properties measured on the post-wet season soil and soil quality was assessed taking productivity and sustainability of dry season rice as goal functions. Results revealed that the highest productivity and sustainability of dry season rice was found with application of 100% NPK + FYM. This treatment, in general, showed better physical, chemical, and biological properties than rest of the treatments. The highest soil quality index (SQI) was recorded in 100% NPK + FYM (0.941) treatment followed by 150% NPK (0.826) with CEC diagnosed as the only key indicator for rice productivity. For yield sustainability in dry season, reserve K and total N were important contributing 89% and 11%, respectively to the SQI. Therefore, these soil properties could be used to monitor soil quality in wet season. Application of FYM along with 100% NPK could sustain the productivity of dry season rice by improving soil properties under subtropical rice-rice system.  相似文献

The Golgi-localized, gamma-ear-containing, adenosine diphosphate ribosylation factor-binding proteins (GGAs) are multidomain proteins that bind mannose 6-phosphate receptors (MPRs) in the Golgi and have an essential role in lysosomal enzyme sorting. Here the GGAs and the coat protein adaptor protein-1 (AP-1) were shown to colocalize in clathrin-coated buds of the trans-Golgi networks of mouse L cells and human HeLa cells. Binding studies revealed a direct interaction between the hinge domains of the GGAs and the gamma-ear domain of AP-1. Further, AP-1 contained bound casein kinase-2 that phosphorylated GGA1 and GGA3, thereby causing autoinhibition. This could induce the directed transfer of the MPRs from GGAs to AP-1. MPRs that are defective in binding to GGAs are poorly incorporated into AP-1-containing clathrin-coated vesicles. Thus, the GGAs and AP-1 interact to package MPRs into AP-1-containing coated vesicles.  相似文献

Sperm cells perform precise chemotactic and thermotactic movement which is crucial for fertilization. However, the key molecules involved in detection of different chemical and physical stimuli which guide the sperm during navigation are not well understood. Ca2+-signalling mediated by ion channels seem to play important role in motility and other fertility parameters. In this work, we explored the endogenous localization pattern of TRPV channels in the mature spermatozoa of avian species. Using sperm from white pekin duck (Anas platyrhynchos) as the representative avian model, we demonstrate that duck sperm endogenously express the thermosensitive channels TRPV1, TRPV2, TRPV3, TRPV4, and highly Ca2+-selective channels TRPV5 and TRPV6 in specific yet differential locations. All of these TRPV channels are enriched in the sperm tail, indicating their relevance in sperm motility. Interestingly, the TRPV3 and TRPV4 channels are present in the mitochondrial region. Calcium selective TRPV5 channel is exclusively present in sperm tail and is most abundant among the TRPV channels. This is the first report describing the endogenous presence of TRPV2 and TRPV3 channels in the sperm of any species. Using confocal imaging and super-resolution imaging, we demonstrate that though the TRPV channels are evolutionarily closely related, they have distinct localization pattern in the duck sperm, which could impact their role in fertilization.  相似文献

Germ cell (GC) transplantation (GCT) is a proven powerful reproductive technique to enhance the production efficiency of domesticated animals and aid to the recovery of endangered germ lines. In mammals, several methods have been adopted for the eradication of GCs such as treatment with cytotoxic drugs, irradiation, cold ischaemia and hyperthermic treatment. Some of these methods have also been tried in fishes, and conditions for sterilization of gonads have been established. Here, we report the production of GC‐depleted male striped catfish Pangasianodon hypophthalmus in 12 weeks by the combination of heat and chemical treatments. The cytotoxic drug busulphan (40 mg/kg) was intraperitoneally injected into the animals at 2‐week intervals (six doses in total) and maintained in water at 38°C between weeks 1 and 12. The effectiveness of the treatment was assessed using gonadal index and histology. At the end of 12 weeks, very severe gonadal degeneration was observed in fish treated with the heat–chemical combination, and 100% of sampled fish (n = 5) were found devoid of endogenous GCs. On contrary, high temperature alone caused minor gonadal degeneration. Results obtained in this study suggest that endogenous GCs of large‐bodied fish such as P. hypophthalmus can also be sterilized by heat and chemical treatments within a considerably short period.  相似文献

The Horsfield's treeshrew (Tupaia javanica) is one of the shrews found on the island of Java that mainly feeds on fruits and occasionally insects. This shrew has an important role in the ecosystem as it spreads plant seeds and controls insects. To this day, there is no available knowledge about the structure of Horsfield's treeshrew's tongue. Therefore, the aim of this study was to provide data on the tongue structure of the Horsfield's treeshrew through two different methods. We investigated the tongue morphology and lingual papillae distribution of Tupaia javanica using 1) scanning electron microscopy (SEM) for microscopic structure and 2) histology for microscopic organization. Our macroscopic results revealed that the tongue of Tupaia javanica can be divided into three parts: apex, corpus and radix. The dorsal surface of the medial corpus is the median groove extending to the radix. Observation using SEM and light microscopy (LM) showed that the apex of the tongue bears three types of papillae: scale-like filiform, small filiform and fungiform. These are rosette filiform papillae, cornflower filiform papillae, conical papillae and fungiform papillae in the corpus. Meanwhile, the radix contains small filiform papillae, scale-like filiform papillae, vallate papillae, conical papillae and fungiform papillae. On the lateral posterior radix, Weber's gland reacts positively to PAS reaction. Taking together, our research shows that there are variations in the papillae on the tongue surface of Horsfield's treeshrew, and the variations of these papillae are very dependent on the pattern, type of food and the function of the tongue.  相似文献
